| 1 | Effects of histone acetylation and DNA methylation on p21^(WAF1)regulation显示文摘Cell cycle progression is regulated by interactions betweencyclins and cyclin-dependent kinases (CDKs). p21wAF1 is oneof the CIP/KIP family which inhibits CDKs activity. Increasedexpression of p21WAF1 may play an important role in thegrowth arrest induced in transformed calls. Although thestability of the p21wAF1 mRNA could be altered by differentsignals, cell differentiation and numerous influencingfactors. However, recent studies suggest that two knownmechanisms of epigenesis, i. e. gene inactivation bymethylation in promoter region and changes to an inactivechromatin by histone deacetylation, seem to be the bestcandidate mechanisms for inactivation of p21WAF1. To date,almost no coding region p21wAF1 mutations have been foundin tumor cells, despite extensive screening of hundreds ofvarious tumors. Hypermethylation of the p21WAF1 promoterregion may represent an alternative mechanism by which thep21WAF1/ClPl gene can be inactivated. The reduction of cellularDNMT protein levels also induces a corresponding rapidincrease in the cell cycle regulator p21wAF1 proteindemonstrating a regulatory link between DNMT and p21WAF1which is independent of methylation of DNA. Both histonehyperacetylation and hypoacetylation appear to be importantin the carcinoma process, and induction of the p21WAF1 geneby histone hyperacetylation may be a mechanism by whichdietary fiber prevents carcinogenesis. | 2 | Predicting Cadmium Safety Thresholds in Soils Based on Cadmium Uptake by Chinese Cabbage显示文摘Cadmium(Cd), a common toxic heavy metal in soil, has relatively high bioavailability, which seriously threatens agricultural products. In this study, 8 different soils with contrasting soil properties were collected from different regions in China to investigate the Cd transfer coefficient from soil to Chinese cabbage(Brassica chinensis L.) and the threshold levels of Cd in soils for production of Chinese cabbage according to the food safety standard for Cd. Exogenous Cd(0–4 mg kg^(-1)) was added to the soils and equilibrated for 2 weeks before Chinese cabbage was grown under greenhouse conditions. The influence of soil properties on the relationship between soil and cabbage Cd concentrations was investigated. The results showed that Cd concentration in the edible part of Chinese cabbage increased linearly with soil Cd concentration in 5 soils, but showed a curvilinear pattern with a plateau at the highest dose of exogenous Cd in the other 3 soils. The Cd transfer coefficient from soil to plant varied significantly among the different soils and decreased with increasing soil p H from 4.7 to 7.5. However, further increase in soil pH to > 8.0 resulted in a significant decrease in the Cd transfer coefficient. According to the measured Cd transfer coefficient and by reference to the National Food Safety Standards of China, the safety threshold of Cd concentration in soil was predicted to be between 0.12 and 1.7 mg kg^(-1) for the tested soils. The predicted threshold values were higher than the current soil quality standard for Cd in 5 soils, but lower than the standard in the other 3 soils. | 3 | Modification of the right subclavian vein catheterization and its anatomic basis and techniques显示文摘Background Several million subclavian-vein catheters are placed in patients each year to enable caregivers to administer chemotherapy, total parenteral nutrition, or long-term antibiotics or to manage preoperative fluids. Subclavian venipuncture requires the position of a deep vein to be identified with only surface landmarks. But the traditional right subclavian vein (RSV) catheterization (primitive procedures) is not the answer for all patients. The precise location of the vein is not known, and it is important to select the most appropriate method to achieve central venous access safely in any given patient. To modify the primitive procedures of the RSV catheterization for greater success and reduce the complications, anatomic studies and ultrasonography were conducted and clinical applications were validated. Methods Anatomical observation and measurement of the RSV and its adjacent structures were performed on 20 adult cadavers according to modified procedures. The RSV catheterization of 2900 cases was carried out by the modified procedure, 500 of these cases were observed by ultrasonography after the operation. Results The anatomical studies and clinical application showed that the insertion point differs from the bodily form of fatness or leptosome. The clinical data revealed that in the 2900 cases which were performed with the modified approach, the success rate was 98.90% (2868 cases), the failure rate was 1.10% (32 cases), and the complication rate is 0.79% (23 cases), and the catheterization time is (31.2±10.5) minutes. Five hundred and sixty cases of the RSV catheterization were carried out by the recommended insertion procedure; the results were compared with the modified approach and the traditional approach. The successful rate of the traditional approach was 73.0%, of which the complication rate was 6.1%; the two approaches were significantly different (successful rate: χ 2=626.642, P <0.01; complication rate: χ 2=80.708, P <0.01). | 4 | Current-limiting characteristics of saturated iron-core fault current limiters in VSC-HVDC systems based on electromagnetic energy conversion mechanism显示文摘A common method to examine the current-limiting performance of saturated iron-core fault current limiter(SI-FCL) in high-voltage direct-current transmission based on voltage source converter(VSC-HVDC) systems is to solve differential equations based on the system fault transient characteristics and the equivalent inductance calculation equation. This method analyzes the fault current of the VSC-HVDC system in the time domain. However, it is computationally complex and cannot directly reflect the relationship between parameters and the currentlimiting effect of the SI-FCL.In this paper,the relationship between the magnetic flux density and magnetic field energy of the SI-FCL is analyzed. The energy exchange between the DC capacitor and the SI-FCL in the DC short circuit fault process is analyzed. From the perspective of electromagnetic energy conversion, the criterion for determining the current-limiting ability of the SI-FCL in the transient process is given based on the parameters of the SI-FCL and VSC-HVDC system. On this basis, the characteristics of the DC side fault current and the capacitor voltage when the SI-FCL has current-limiting ability are examined.Based on the parameters of the SI-FCL and VSC-HVDC system, a method for calculating the fault current peak value and capacitor voltage drop time is given. | 5 | High-Q whispering gallery modes in a polymer microresonator with broad strain tuning显示文摘We have reported the high-Q whispering gallery modes(WGMs) in a polydimethylsiloxane(PDMS) optical microresonators with broad tuning range. The PDMS microresonators are fabricated at the center of two collimating fiber tips, which can be controlled by the piezoelectric stage. Through stretching the fiber stem, the tuning range of WGMs are demonstrated more than 50 nm.Further investigations demonstrated that the WGM shift has a high force sensitivity(~ 19.7 pm/μN) of the gravitation when the microcavity is stretched by a weight. | 7 | A mechanically tuned Kerr comb in a dispersion-engineered silica microbubble resonator显示文摘This paper discusses the developement and investigation of a silica microbubble resonator(MBR) that is optimized to cancel mode dispersion with material dispersion, at a wavelength of approximately 1550 nm and maintain a quality factor of an optical mode as large as 5.4 × 10~7. Benefitting from the near-zero dispersion and high quality factor, a primary optical comb is generated in the MBR using cascaded four-wave mixing processes, which span over 300 nm with several tens of teeth. Furthermore, the frequency comb could be gradually tuned by mechanically stretching the MBR. | 11 | Diurnal and seasonal variations in carbon fluxes in bamboo forests during the growing season in Zhejiang province, China显示文摘Bamboo forest is an important forest type in subtropical China and is characterized by fast growth and high carbon sequestration capacity. However, the dynamics of carbon fluxes during the fast growing period of bamboo shoots and their correlation with environment factors are poorly understood. We measured carbon dioxide exchange and climate variables using open-path eddy covariance methods during the 2011 growing season in a Moso bamboo forest(MB, Phyllostchys edulis) and a Lei bamboo forest(LB, Phyllostachys violascens) in Zhejiang province,China. The bamboo forests were carbon sinks during the growing season. The minimum diurnal net ecosystem exchange(NEE) at MB and LB sites were-0.64 and-0.66 mg C m^(-2) s^(-1), respectively. The minimum monthly NEE, ecosystem respiration(RE), and gross ecosystem exchange(GEE) were-99.3 ± 4.03, 76.2 ±2.46, and^(-1)91.5 ± 4.98 g C m^(-2) month^(-1), respectively,at MB site, compared with-31.8 ± 3.44, 70.4 ± 1.41,and^(-1)57.9 ± 4.86 g C m^(-2) month^(-1), respectively, at LB site. Maximum RE was 92.1 ± 1.32 g C m^(-2) month^(-1) at MB site and 151.0 ± 2.38 g C m^(-2) month^(-1) at LB site.Key control factors varied by month during the growing season, but across the whole growing season, NEE and GEE at both sites showed similar trends in sensitivities to photosynthetic active radiation and vapor pressure deficit,and air temperature had the strongest correlation with RE at both sites. Carbon fluxes at LB site were more sensitive to soil water content compared to those at MB site. | 12 | Phosphorylated TDP-43 Staging of Primary Age-Related Tauopathy显示文摘Primary age-related tauopathy(PART) is characterized by tau neurofibrillary tangles(NFTs) in the absence of amyloid plaque pathology. In the present study,we analyzed the distribution patterns of phosphorylated43-kDa TAR DNA-binding protein(pTDP-43) in the brains of patients with PART. Immunohistochemistry and immunofluorescence double-labeling in multiple brain regions was performed on brain tissues from PART,Alzheimer's disease(AD), and aging control cases. We examined the regional distribution patterns of pTDP-43 intraneuronal inclusions in PART with Braak NFT stages[ 0 and B IV, and a Thal phase of 0(no beta-amyloid present). We found four stages which indicated potentially sequential dissemination of pTDP-43 in PART. Stage I was characterized by the presence of pTDP-43 lesions in the amygdala, stage II by such lesions in the hippocampus,stage III by spread of pTDP-43 to the neocortex, and stage IV by pTDP-43 lesions in the putamen, pallidum, and insular cortex. In general, the distribution pattern of pTDP-43 pathology in PART cases was similar to the early TDP-43 stages reported in AD, but tended to be more restricted to the limbic system. However, there were some differences in the distribution patterns of pTDP-43 between PART and AD, especially in the dentate gyrus of the hippocampus. | 13 | Inversion of the three-dimensional temperature structure of mesoscale eddies in the Northwest Pacific based on deep learning显示文摘Mesoscale eddies,which are mainly caused by baroclinic effects in the ocean,are common oceanic phenomena in the Northwest Pacific Ocean and play very important roles in ocean circulation,ocean dynamics and material energy transport.The temperature structure of mesoscale eddies will lead to variations in oceanic baroclinity,which can be reflected in the sea level anomaly(SLA).Deep learning can automatically extract different features of data at multiple levels without human intervention,and find the hidden relations of data.Therefore,combining satellite SLA data with deep learning is a good way to invert the temperature structure inside eddies.This paper proposes a deep learning algorithm,eddy convolution neural network(ECN),which can train the relationship between mesoscale eddy temperature anomalies and sea level anomalies(SLAs),relying on the powerful feature extraction and learning abilities of convolutional neural networks.After obtaining the temperature structure model through ECN,according to climatic temperature data,the temperature structure of mesoscale eddies in the Northwest Pacific is retrieved with a spatial resolution of 0.25°at depths of 0–1000 m.The overall accuracy of the ECN temperature structure is verified using Argo profiles at the locations of cyclonic and anticyclonic eddies during 2015–2016.Taking 10%error as the acceptable threshold of accuracy,89.64%and 87.25%of the cyclonic and anticyclonic eddy temperature structures obtained by ECN met the threshold,respectively. | Fangjie Yu Zeyuan Wang Shuai Liu Ge Chen | 2021 | Acta Oceanologica Sinica2021,40,10: | 2 |
| 14 | Bayesian uncertainty analysis of SA turbulence model for supersonic jet interaction simulations显示文摘The Reynolds Averaged Navier-Stokes(RANS) models are still the workhorse in current engineering applications due to its high efficiency and robustness. However, the closure coefficients of RANS turbulence models are determined by model builders according to some simple fundamental flows, and the suggested values may not be applicable to complex flows, especially supersonic jet interaction flow. In this work, the Bayesian method is employed to recalibrate the closure coefficients of Spalart-Allmaras(SA) turbulence model to improve its performance in supersonic jet interaction problem and quantify the uncertainty of wall pressure and separation length. The embedded model error approach is applied to the Bayesian uncertainty analysis. Firstly, the total Sobol index is calculated by non-intrusive polynomial chaos method to represent the sensitivity of wall pressure and separation length to model parameters. Then, the pressure data and the separation length are respectively served as calibration data to get the posterior uncertainty of model parameters and Quantities of Interests(Qo Is). The results show that the relative error of the wall pressure predicted by the SA turbulence model can be reduced from 14.99% to 2.95% through effective Bayesian parameter estimation. Besides, the calibration effects of four likelihood functions are systematically evaluated. The posterior uncertainties of wall pressure and separation length estimated by different likelihood functions are significantly discrepant, and the Maximum a Posteriori(MAP) values of parameters inferred by all functions show better performance than the nominal values. Finally, the closure coefficients are also estimated at different jet total pressures. | 16 | i4OilS pill, an Operational Marine Oil Spill Forecasting Model for Bohai Sea显示文摘Oil spill models can effectively simulate the trajectories and fate of oil slicks, which is an essential element in contingency planning and effective response strategies prepared for oil spill accidents. However, when applied to offshore areas such as the Bohai Sea, the trajectories and fate of oil slicks would be affected by time-varying factors in a regional scale, which are assumed to be constant in most of the present models. In fact, these factors in offshore regions show much more variation over time than in the deep sea, due to offshore bathymetric and climatic characteristics. In this paper, the challenge of parameterizing these offshore factors is tackled. The remote sensing data of the region are used to analyze the modification of wind-induced drift factors, and a well-suited solution is established in parameter correction mechanism for oil spill models. The novelty of the algorithm is the self-adaptive modification mechanism of the drift factors derived from the remote sensing data for the targeted sea region, in respect to empirical constants in the present models. Considering this situation, a new regional oil spill model(i4Oil Spill) for the Bohai Sea is developed, which can simulate oil transformation and fate processes by Eulerian-Lagrangian methodology. The forecasting accuracy of the proposed model is proven by the validation results in the comparison between model simulation and subsequent satellite observations on the Penglai 19-3 oil spill accident. | 17 | Postnatal renin-angiotensin system inhibition prevents renal damage from prenatal inflammation in rats显示文摘OBJECTIVE:To assess the protective role of benazepril,an angiotensin-converting enzyme inhibitor,in renal damage caused by prenatal inflammation.METHODS:Saline or lipopolysaccharide were administered intraperitoneally to pregnant Sprague-Dawley rats on gestation days 8,10,and 12.After birth,offspring received either tap water or benazepril in water between 7 and 68 weeks.Blood pressure,blood urea nitrogen,creatinine,and 24-h urine volume were measured as indices of renal function.Hematoxylin,eosin,periodic acid-Schiff,and Sirius Red staining were used to evaluate renal damage.RESULTS:Postnatal benazepril treatment ameliorated hypertension and restored normal 24-h urine volume and blood urea nitrogen and serum creatinine levels.Benazepril treatment also reduced glycoprotein accumulation and fibrosis in the glomerulus and in tubular epithelial cells and inhibited nuclear factor-kappa B activation.CONCLUSION:Together with our previous findings that postnatal inhibition of nuclear factor-kappa B activation blocks intra-renal renin-angiotensin system activation,our current data demonstrate that intra-renal activation of the renin-angiotensin system interacts with nuclear factor-kappa B activation to cause renal damage in adulthood following prenatal inflammation. | Guo Wei Ji Yan Han Qi Yang Yao Wang Fangjie Yang Yongjian Deng Youcai Sun Xiongshan Li Xiaohui | 2020 | Journal of Traditional Chinese Medicine2020,40,4: | 2 |
| 18 | Progress and Knowledge Transfer from Science to Technology in the Research Frontier of CRISPR Based on the LDA Model显示文摘Purpose:This study explores the underlying research topics regarding CRISPR based on the LDA model and figures out trends in knowledge transfer from science to technology in this area over the latest 10 years.Design/methodology/approach:We collected publications on CRISPR between 2011 and2020 from the Web of Science,and traced all the patents citing them from lens.org.15,904 articles and 18,985 patents in total are downloaded and analyzed.The LDA model was applied to identify underlying research topics in related research.In addition,some indicators were introduced to measure the knowledge transfer from research topics of scientific publications to IPC-4 classes of patents.Findings:The emerging research topics on CRISPR were identified and their evolution over time displayed.Furthermore,a big picture of knowledge transition from research topics to technological classes of patents was presented.We found that for all topics on CRISPR,the average first transition year,the ratio of articles cited by patents,the NPR transition rate are respectively 1.08,15.57%,and 1.19,extremely shorter and more intensive than those of general fields.Moreover,the transition patterns are different among research topics.Research limitations:Our research is limited to publications retrieved from the Web of Science and their citing patents indexed in lens.org.A limitation inherent with LDA analysis is in the manual interpretation and labeling of'topics'.Practical implications:Our study provides good references for policy-makers on allocating scientific resources and regulating financial budgets to face challenges related to the transformative technology of CRISPR.Originality/value:The LDA model here is applied to topic identification in the area of transformative researches for the first time,as exemplified on CRISPR.Additionally,the dataset of all citing patents in this area helps to provide a full picture to detect the knowledge transition between S&T. | Yushuang Lyu Muqi Yin Fangjie Xi Xiaojun Hu | 2022 | Journal of Data and Information Science2022,7,1: | 2 |
| 19 | Identifying storm-induced wave origins using SAR wave mode data显示文摘Because of weak dissipation effects, swells generated by fierce storms can propagate across an entire ocean basin;therefore, observing swell generation and decay and retrieving storm characteristics from a swell by satellite remote sensing are possible. In this study, based on the dispersion relation and geometrical optics principle, we used SAR wave mode data from 2003 to 2010 provided by GlobWave to track swells with peak wavelengths of more than 300 m to locate a storm-generated far-traveling swell and present the swell field related to this 'static' origin. Through a comparison with ECMWF wind datasets, we conducted validations and explored some conditions that cause misjudgments in swell origins. Finally, we obtained the spatiotemporal distribution characteristics of satellite-observed swell origins(i.e., the fierce wind condition) and their evolution.
