2AFLP analysis revealed differences in genetic diversity of four natural populations of Manila clam (Ruditapes philippinarum) in China显示文摘The amplified fragment length polymorphism (AFLP) technology was used to analyze the genetic diversities in four natural populations of Manila clam (Ruditapes philippinarum), distributed in four sea areas of China, i.e., the Bohai Sea, the Huanghai Sea, the East China Sea and the South China Sea. Two hundred and sixty-four AFLP loci were analysed in 195 individuals and revealed high levels of genetic diversity. The percentage of polymorphic loci ranged from 92.13% to 96.06% and the Shannon’s information index was from 0.256 8 to 0.275 6. By analyzing molecular variance (AMOVA), it was found that there were high levels of genetic differentiation between populations of Qingdao and the other three sea areas. Cluster analysis by Nei’s pairwise distances grouped specimens by geographical origin, except the population of Qingdao. A conclusion can be drawn that there are high genetic diversities in the four natural populations of Manila clam in China and some distinct differences existed among and between the four populations. 4Clinical and laboratory survey of 65 Chinese patients with Leigh syndrome显示文摘Background Leigh syndrome is an inherited neurodegenerative disease that emerges in infancy and childhood and presents with a clinically heterogeneous variety of neuromuscular and non-neuromuscular disorders. It can result from the inheritance of mutations in either nuclear or mitochondrial DNA. In the current study, we performed a retrospective study in 65 patients in order to investigate the clinical and genetic characteristics of Leigh syndrome in Chinese patients. Methods Sixty-five unrelated cases (35 men and 30 women) who were hospitalized in the past 12 years were reviewed. Diagnosis was based on both the clinical presentation and the characteristic neuropathologic findings of bilateral symmetric necrotizing lesions in the basal ganglia and brain stem as detected using cranial computed tomography (CT) scan or magnetic resonance imaging (MRI). The differential diagnosis of organic acidurias and fatty acid ?-oxidation defects were performed. Specific point mutations and deletions in mitochondrial DNA (T8993G, T8993C, T9176C, A8344G, A3243G) were screened by PCR-restriction analysis and Southern blot. The SURF1 gene was sequenced. Skeletal muscle biopsies were performed in 17 (26.2%) of the patients. The diagnosis was confirmed by autopsy in 6 (9.2%) patients. Results The patients had various forms of metabolic encephalomyopathy. Fifty-nine (90.8%) of the patients had the typical neuroradiological features of Leigh syndrome, including symmetrical necrotizing lesions scattered within the basal ganglia, thalamus and brain stem. Twenty (30.8%) patients were confirmed by genetic, biochemical analysis and autopsy. Specific point mutations in mitochondrial DNA were found in 5 cases (7.7%). Of these, the A8344G mutation was detected in 2 patients. The T8993G, T8993C, and A3243G point mutations were identified in 3 other patients, respectively. SURF1 mutations associated with cytochrome c oxidase deficiency were identified in 8 (12.3%) families by DNA sequencing. A G604C mutation was identified in 6 (9.2%) patients. The genotypes of 52 patients remained unknown. Conclusions Leigh syndrome presents as a diverse array of clinical features and can result from specific mutations in nuclear or mitochondrial DNA. In this study, SURF1 mutations associated with cytochrome c oxidase deficiency were identified in 8 (12.3%) out of 65 patients with Leigh syndrome. It indicates that SURF1 mutations might be a common cause of Leigh syndrome in China. 7Mutual optimization of water utilization structure and industrial structure in arid inland river basins of Northwest China显示文摘Water is a key restricting factor of the economic development and eco-environmental protection in arid inland river basins of Northwest China. Although water supplies are short, the water utilization structure and the corresponding industrial structure are unbalanced. We constructed a System Dynamic Model for mutual optimization based on the mechanism of their interaction. This model is applied to the Heihe River Basin where the share of limited water resources among ecosystem, production and human living is optimized. Results show that, by mutual optimization, the water utilization structure and the industrial structures fit in with each other. And the relationships between the upper, middle and lower reaches of the Heihe River Basin can be harmonized. Mutual benefits of ecology, society and economy can be reached, and a sustainable ecology-production-living system can be obtained. 8An Estimation of Internal Soliton Forces on a Pile in the Ocean显示文摘Internal soliton forces on oil-platform piles in the ocean are estimated with the Morison Formula. Different from sur- face wave forces, which change only in magnitude along a pile, internal soliton forces can be distributed over the entire pile in the water and they change not only in magnitude but also in direction with depth. Our calculations show that the maximum total force caused by a soliton with its associated current of 2.1 m s-1 is nearly equal to the maximum total force exerted by a surface wave with a wavelength of 300 m and a wave-height of 18 m. The total internal soliton force is large enough to affect the operations of marine oil platforms and other facilities. 10Hepatitis C virus in human B lymphocytes transformed by Epstein-Barr virus in vitro by in situ reverse transcriptase-polymerase chain reaction显示文摘AIM To study persistence and replication ofheltitis C virus (HCV) in patients' peripheralblood mononuclear cells (PBMC) cultured invitro.METHODS Epstein-Barr virus (EBV) was usedto transform the hepatitis C virus from a HCVpositive patient to permanent lymphoblastoidcell lines (LCL). Positive and negative HCV RNAstrands of the cultured cells and growth mediawere detected by reverse transcriptase-polymerase chain reaction ( RT-PCR ) eachmonth. Core and NS5 proteins of HCV werefurther tested using immunohistochemical SPmethod and in situ RT-PCR.RESULTS HCV RNA positive strands wereconsistently detected the cultured cells for oneyear. The negative-strand RNA in LCL cells andthe positive-strand RNA in supernatants wereobserved intermittently. Immunohistochemicalresults medicated expression of HCV NS3 and Cproteins in LCL cytoplasm mostly. The positivesignal of PCR product was dark blue and mainlylocalized to the LCL cytoplasm. 13Higher alcohol synthesis over Cu-Fe composite oxides with high selectivity to C_(2+)OH显示文摘Cu-Fe composite oxides were prepared by co-precipitation method and tested for higher alcohol synthesis from syngas. The selectivity to C2+OH and C6+OH in alcohol distribution was very high while the methane product fraction in hydrocarbon distribution was rather low, demonstrating a promising potential in higher alcohols synthesis from syngas. The distribution of alcohols and hydrocarbons approximately obeyed Anderson-Schulz-Flory distribution with similar chain growth probability, indicating alcohols and hydrocarbons derived from the same intermediates. The effects of Cu/Fe molar ratio, reaction temperature and gas hourly space velocity (GHSV) on catalytic performance were studied in detail. The sample with a Cu/Fe molar ratio of 10/1 exhibited the best catalytic performance. Higher reaction temperature accelerated water-gas-shift reaction and led to lower total alcohols selectivity. 16Effect of Danzhijiangtang capsule on monocyte chemoattractant protein-1 mRNA expression in newly diagnosed diabetes subclinical vascular lesions显示文摘AIM:To investigate the effect of Danzhijiangtang capsule(DJC) on monocyte chemoattractant protein-1(MCP-1) mRNA expression in newly diagnosed type 2 diabetes mellitus(T2DM) subclinical vascular lesions.METHODS:Sixty-two patients with newly diagnosed T2DM subclinical vascular lesions were randomly divided into a control group and treatment group of 31 cases each.Oral antidiabetic therapy with routine western medicine was conducted in both groups,and the treatment group was additionally treated with DJCs.The treatment course for both groups was 12 wk.Before and after treatment,the total efficiency and traditional Chinese medicine(TCM) syndrome score were calculated.The fasting plasma glucose(FPG),2-h plasma glucose(2hPG),fasting insulin(FINS),insulin resistance index(IRI),hemoglobin(Hb)A1c,blood lipids,and hemorheology indices were determined.In addition,the levels of vascular endothelial growth factors including thrombomodulin(TM),von Willebrand factor(vWF),P-selectin and MCP-1 mRNA were determined.RESULTS:After 12 wk of treatment,the TCM syndrome score was significantly decreased compared to before treatment in both groups.After treatment,FPG,2hPG,HbA1c,FINS,IRI,total cholesterol,triglycerides,low-density lipoprotein,high-density lipoprotein,whole blood low shear specific viscosity,plasma specific viscosity,TM,vWF,P-selectin and MCP-1 mRNA were significantly improved compared to before treatment in both groups.After treatment,the total efficiency and TCM syndrome score in the treatment group were better than in the control group.FINS,IRI,whole blood high shear specific viscosity,plasma specific viscosity,TM,vWF,P-selectin and MCP-1 mRNA level in the treatment group were significantly reduced after treatment compared with control group.CONCLUSION:DJCs are efficacious in supplementing qi,nourishing yin and invigorating blood circulation,and upregulate MCP-1 mRNA expression in patients with T2DM subclinical vascular lesions.Zhao-Hui Fang Yan Liu Tao-Tao Bao Ying-Qun Ni Jian Liu Guo-Bin Shi Ji-Ping Wu Jun-Ping Yang Hong Zhang 2013World Journal of Gastroenterology2013,19,19:9

20Spatial relationship of high-speed transportation construction and land-use efficiency and its mechanism:Case study of Shandong Peninsula urban agglomeration显示文摘Land-use efficiency is low for the urban agglomeration of China.High-speed transportation construction has been an important factor driving land use change.It is criti?cally importa nt to explore the spatial relati on ship between the high-speed transportation su?periority degree and land-use efficie ncy.We built a model to evaluate the ben efits of con?venient high-speed transportation using the relative density of highways and the distanee from high-speed rail stations and airports as a metric.We used 42 coun ties of the Shan dong Peninsula urban agglomeration as an example.Land?use efficiency was calculated by a DEA model with capital,labor,economic ben efits and envir on mental ben efits as input and output factors.We examined the spatial relati on ships betwee n high-speed tran sport superiority degree and land-use efficiency and obtained the following results.First,there are significant spatial differe nces in the relati on ships betwee n the high-speed transportati on superiority degree and land-use efficiency.Taking the two major cities of Jinan and Qingdao as the hubs,the core surrounding counties show significant spatial relationship between land-use efficiency and the high-speed transportation superiority degree.Spatial correlation declines as the distance from the hubs in creases.Land-use efficiency is less tha n high-speed tran sportation convenience in areas along the transportation trunks that are distant from the hub cities.Correlation is low in areas that are away from both hub cities and transportation trunk routes.Second,high-speed transportation has a positive relationship with land?use efficiency due to the mechanism of element agglomeration exogenous growth.Third,high-speed transportation facilitates the flow of goods,services and tech no logies between core cities and peripheral cities as space spillover(the hub effect).This alters the spatial pattern of regional land-use efficiency.Fin ally,the short-board effect caused by decreased high-speed tran sport corn struction can be balaneed by highway construction and the proper node layouts of high-speed rail stations and airports,resulting in a well-balaneed spatial pattern of land?use efficiency.CUI Xuegang FANG Chuanglin WANG Zhenbo BAO Chao 2019Journal of Geographical Sciences2019,29,4:8
