Syntheses and Insecticidal Activity of Spirocyclic Tetronic Acid Derivatives Containing Oxime Ether Moiety显示文摘In order to develop novel spirocyclic tetronic acid lead compounds,a series of new spirocyclic tetronic acid derivatives containing oxime ether moiety was synthesized and bioassayed. The stmctures of 16 target compounds were characterized by lH NMR spectra,13C NMR spectra and high-resolution mass spectrometer(HRMS).Preliminary bioassays indicated that most of the title compounds displayed excellent insecticidal activity against Aphis fabae and Nilapmyata lugens at 100 mg/L.In particular, compound 6k showed the similar activity(LC50=6.87 mg/L)against A.fabae as the spirotetramat(LC50=4.56 mg/L)and had better efIects(LC50=l.64 mg/L)against N.lugens in comparison with spirotetramat(LC5o=7.90 mg/L).The study showed that compound 6k exhibited more promising and broad-spectrum insecticide activity and may serve as a new insecticidal agent for sucking pests. | ZHAO Yang CHENG Jingli XIE Zhengang FANG Shaowei ZHAO Jinhao | 2020 | Chemical Research in Chinese Universities2020,36,5: | 3
Capsid destabilization and epitope alterations of human papillomavirus 18 in the presence of thimerosal显示文摘Thimerosal has been widely used as a preservative in drug and vaccine products for decades.Due to the strong propensity to modify thiols in proteins, conformational changes could occur due to covalent bond formation between ethylmercury(a degradant of thimerosal)and thiols.Such a conformational change could lead to partial or even complete loss of desirable protein function. This study aims to investigate the effects of thimerosal on the capsid stability and antigenicity of recombinant human papillomavirus(HPV)18 virus-like particles(VLPs).Dramatic destabilization of the recombinant viral capsid upon thimerosal treatment was observed.Such a negative effect on the thermal stability of VLPs preserved with thimerosal was shown to be dependent on the thimerosal concentration. Two highly neutralizing antibodies,13H12 and 3C3,were found to be the most sensitive to thimerosal treatment. The kinetics of antigenicity loss, when monitored with 13H12 or 3C3 as probes, yielded two distinctly different sets of kinetic parameters, while the data from both monoclonal antibodies(mAbs)followed a biphasic exponential decay model. The potential effect of thimerosal on protein function, particularly for thiolcontaining proteinaceous active components, needs to be comprehensively characterized during formulation development when a preservative is necessary. | Xiaofen Huang Yike Li Meifeng Nie Mingxi Yue Yufang Li Zhijie Lin Huirong Pan Mujin Fang Ting Wu Shaowei Li Jun Zhang Ningshao Xia Qinjian Zhao | 2021 | Journal of Pharmaceutical Analysis2021,11,5: | 1
Ion–Electron Coupling Enables Ionic Thermoelectric Material with New Operation Mode and High Energy Density显示文摘Ionic thermoelectrics(i-TE) possesses great potential in powering distributed electronics because it can generate thermopower up to tens of millivolts per Kelvin. However,as ions cannot enter external circuit, the utilization of i-TE is currently based on capacitive charge/discharge, which results in discontinuous working mode and low energy density. Here,we introduce an ion–electron thermoelectric synergistic(IETS)effect by utilizing an ion–electron conductor. Electrons/holes can drift under the electric field generated by thermodiffusion of ions, thus converting the ionic current into electrical current that can pass through the external circuit. Due to the IETS effect, i-TE is able to operate continuously for over 3000 min. Moreover, our i-TE exhibits a thermopower of 32.7 mV K^(-1) and an energy density of 553.9 J m^(-2), which is more than 6.9 times of the highest reported value. Consequently, direct powering of electronics is achieved with i-TE. This work provides a novel strategy for the design of high-performance i-TE materials. | Yongjie He Shaowei Li Rui Chen Xu Liu George Omololu Odunmbaku Wei Fang Xiaoxue Lin Zeping Ou Qianzhi Gou Jiacheng Wang Nabonswende Aida Nadege Ouedraogo Jing Li Meng Li Chen Li Yujie Zheng Shanshan Chen Yongli Zhou Kuan Sun | 2023 | Nano-Micro Letters2023,15,7: | 1

Efficacy of comprehensive cognitive health management for Shanghai community older adults with mild cognitive impairment显示文摘Background The management of modifiable risk factors and comorbidities may impact the future trajectory of cognitive impairment, but easy-to-implement management methods are lacking.Aims This study investigated the effects of simple but comprehensive cognitive health management practices on the cognitive function of older adults in the community with normal cognition(NC)and mild cognitive impairment(MCI).Methods The comprehensive cognitive health management programme included a psychiatric assessment of the cognitive risk factors for those in the intervention groups and individualised recommendations for reducing the risks through self-management supported by regular medical professional follow-up.The intervention groups for this study included 84 elderly participants with NC and 43 elderly participants with MCI who received comprehensive cognitive health management. The control groups included 84 elderly participants with NC and 43 elderly participants with MCI who matched the intervention group’s general characteristics and scale scores using the propensity matching score analysis. The Montreal Cognitive Assessment(MoCA)scale and Geriatric Depression Scale(GDS)scores were compared after a 1-year follow-up.Results For older adults with MCI in the intervention group,MoCA scores were higher at the 1-year followup than at baseline(24.07(3.674)vs 22.21(3.052),p=0.002).For the MoCA subscales, the intervention group’s abstract and delayed memory scores had significantly increased during the 1-year follow-up.Furthermore,in a generalised linear mixed model analysis, the interaction effect of group×follow-up was statistically significant for the MCI group(F=6.61,p=0.011;coefficients=5.83).Conclusions After the comprehensive cognitive health management intervention, the older adults with MCI in the community showed improvement at the 1-year follow-up.This preliminary study was the first to demonstrate an easy-to-implement strategy for modifying the cognitive risk factors of elderly individuals with MCI in the community, providing new insight into early-stage intervention for dementia. | Jiayuan Qiu Lu Zhao Shifu Xiao Shaowei Zhang Ling Li Jing Nie Li Bai Shixing Qian Yang Yang Michael Phillips Meiqing Sheng Yuan Fang Xia Li | 2022 | General Psychiatry2022,35,4: | 0
Two antibodies show broad, synergistic neutralization against SARS-CoV-2 variants by inducing conformational change within the RBD显示文摘Continual evolution of the severe acute respiratory syndrome coronavirus(SARS-CoV-2)virus has allowed for its gradual evasion of neutralizing antibodies(nAbs)produced in response to natural infection or vaccination. The rapid nature of these changes has incited a need for the development of superior broad nAbs(bnAbs)and/or the rational design of an antibody cocktail that can protect against the mutated virus strain. Here,we report two angiotensin-converting enzyme 2 competing nAbs—8H12 and 3E2—with synergistic neutralization but evaded by some Omicron subvariants.Cryo-electron microscopy reveals the two nAbs synergistic neutralizing virus through a rigorous pairing permitted by rearrangement of the 472-489 loop in the receptor-binding domain to avoid steric clashing.Bispecific antibodies based on these two nAbs tremendously extend the neutralizing breadth and restore neutralization against recent variants including currently dominant XBB.1.5.Together,these findings expand our understanding of the potential strategies for the neutralization of SARS-CoV-2 variants toward the design of broad-acting antibody therapeutics and vaccines. | Hui Sun Tingting Deng Yali Zhang Yanling Lin Yanan Jiang Yichao Jiang Yang Huang Shuo Song Lingyan Cui Tingting Li Hualong Xiong Miaolin Lan Liqin Liu Yu Li Qianjiao Fang Kunyu Yu Wenling Jiang Lizhi Zhou Yuqiong Que Tianying Zhang Quan Yuan Tong Cheng Zheng Zhang Hai Yu Jun Zhang Wenxin Luo Shaowei Li Qingbing Zheng Ying Gu Ningshao Xia | 2024 | Protein & Cell2024,15,2: | 0
