1DNA end binding activity and Ku70/80 heterodimer expression in human colorectal tumor显示文摘AIM: To determine the DNA binding activity and protein levels of the Ku70/80 heterodimer, the functional mediator of the NHEJ activity, in human colorectal carcinogenesis.METHODS: The Ku70/80 DNA-binding activity was determined by electrophoretic mobility shift assays in 20 colon adenoma and 15 colorectal cancer samples as well as matched normal colonic tissues. Nuclear and cytoplasmic protein expression was determined by immunohistochemistry and Western blot analysis.RESULTS: A statistically significant difference was found in both adenomas and carcinomas as compared to matched normal colonic mucosa (P<0.00). However, changes in binding activity were not homogenous with approximately 50% of the tumors showing a clear increase in the binding activity, 30% displaying a modest increase and 15% showing a decrease of the activity.Tumors, with increased DNA-binding activity, also showed a statistically significant increase in Ku70 and Ku86nuclear expression, as determined by Western blot and immunohistochemical analyses (P<0.001). Cytoplasmic protein expression was found in pathological samples, but not in normal tissues either from tumor patients or from healthy subjects.CONCLUSION: Overall, our DNA-binding activity and protein level are consistent with a substantial activation of the NHEJ pathway in colorectal tumors. Since the NHEJ is an error prone mechanism, its abnormal activation can result in chromosomal instability and ultimately lead to tumorigenesis.Paola Mazzarelli Paola Parrella Davide Seripa Emanuela Signori Giuseppe Perrone Carla Rabitti Domenico Borzomati Armando Gabbrielli Maria Giovanna Matera Carolina Gravina Marco Caricato Maria Luana Poeta Monica Rinaldi Sergio Valeri Roberto Coppola Vito Michele Fazio 2005World Journal of Gastroenterology2005,11,42:4
2Electrochemotherapy using thin-needle electrode improves recovery in feline nasal planum squamous cell carcinoma-a translational model显示文摘Aim:Cutaneous squamous cell carcinoma(cSCC)is a common disease in patients exposed to UV-light and human papillomavirus.Electrochemotherapy,a well-established treatment modality with minimum side effects in human and veterinary medicine, circumvents chemoresistance to bleomycin by the use of electric fields. However, patients are sensitive to the trauma produced by the insertion of the needles that lengthen recovery times, particularly cats with nasal planum cSCC.To address this matter,we developed thin-needles electrodes.Methods:Thin-needles electrodes developed using computer simulations and plant tissue models were compared to standard electrodes.A prospective non-randomized study recruiting 52 feline patients with nasal planum cSCC was performed.Local response, anorexia, and overall survival were evaluated. Results:Computer simulations and plant model experiments showed satisfactory results with both electrodes. The patients treated with the thin-needle electrode obtained similar local response rates compared to the standard group,OR 97.3%vs.80%,respectively(P<0.067).Most patients in the thin-needle group resumed eating in less than 48 h,as the anorexia was significantly lower(P<0.0001).Using the standard electrode, most patients took 3 to 5 days to resume normal feeding. The electric current circulating in the standard electrode was 44% higher, contributing to a longer duration of anorexia due to tissue damage. The overall survival in both groups was similar. Conclusion:Electrochemotherapy using thin-needle electrodes provides equivalent local response rates and overall survival compared with standard electrodes but significantly reduced return to appetite after the treatment. These results may be useful in the development of new electrodes for human patients.Matias Tellado Sebastian Michinski Joseph Impellizeri Guillermo Marshall Emanuela Signori Felipe Maglietti 2022Cancer Drug Resistance2022,5,3:1
