1Oxymatrine reduces neuroinflammation in rat brain A signaling pathway显示文摘Cerebral neuroinflammation models were established by injecting 10 μg lipopolysaccharide into the hippocampus of male Sprague-Dawley rats. The rats were treated with an intraperitoneal injection of 120, 90, or 60 mg/kg oxymatrine daily for three days prior to the lipopolysaccharide injection. Twenty-four hours after model induction, the hippocampus was analyzed by real-time quantitative PCR, and the cerebral cortex was analyzed by enzyme-linked immunosorbent assay and western blot assay. The results of the enzyme-linked immunosorbent assay and the real-time quantitative PCR showed that the secretion and mRNA expression of the pro-inflammatory cytokines interleukin-1β and tumor necrosis factor-α were significantly decreased in the hippocampus and cerebral cortex of model rats treated with oxymatrine. Western blot assay and real-time quantitative PCR analysis indicated that toll-like receptor 4 mRNA and protein expression were significantly decreased in the groups receiving different doses of oxymatrine. Additionally, 120 and 90 mg/kg oxymatrine were shown to reduce protein levels of nuclear factor-κB p65 in the nucleus and of phosphorylated IκBα in the cytoplasm of brain cells, as detected by western blot assay. 2Tuning the intermolecular interaction of A_(2)-A_(1)-D-A_(1)-A_(2) type non-fullerene acceptors by substituent engineering for organic solar cells with ultrahigh V_(OC) of ~1.2 V显示文摘For non-fullerene acceptors(NFAs)with linear A_(2)-A_(1)-D-A_(1)-A_(2) backbone,there are three kinds of possible intermolecular interaction,A_(1)-A_(1),A_(1)-A_(2) and A_(2)-A_(2) stacking.Hence,it is a huge challenge to control this interaction and investigate the effect of intermolecular stacking model on the photovoltaic performance.Here,we adopt a feasible strategy,by utilizing different substituent groups on terminal A2 unit of dicyanomethylene rhodanine(RCN),to modulate this stacking model.According to theoretical calculation results,the molecule BTA3 with ethyl substituent packs via heterogeneous interaction between A_(2) and A_(1) unit in neighboring molecules.Surprisingly,the benzyl group can effectively transform the aggregation of BTA5 into homogeneous packing of A_(2)-A_(2) model,which might be driven by the strong interaction between benzyl and A1(benzotriazole)unit.However,different with benzyl,phenyl end group impedes the intermolecular interaction of BTA4 due to the large steric hindrance.When using a BTA-based D-π-A polymer J52-F as donor according to“Same-A-Strategy”,BTA3-5 could achieve ultrahigh open-circuit voltage(VOC)of 1.17–1.21 V.Finally,BTA5 with benzyl groups realized an improved power conversion efficiency(PCE)of 11.27%,obviously higher than that of BTA3(PCE=9.04%)and BTA4(PCE=5.61%).It is also worth noting that the same trend can be found when using other four classic p-type polymers of P3HT,PTB7,PTB7-Th and PBDB-T.This work not only investigates the intermolecular interaction of A_(2)-A_(1)-D-A_(1)-A_(2) type NFAs for the first time,but also provides a straightforward and universal method to change the interaction model and improve the photovoltaic performance.Xiaochen Wang Ailing Tang Jing Yang Mengzhen Du Jianfeng Li Gongqiang Li Qiang Guo Erjun Zhou 2020Science China Chemistry2020,63,11:5
3A non-ACE2 competing human single-domain antibody confers broad neutralization against SARS-CoV-2 and circulating variants显示文摘The current COVID-19 pandemic has heavily burdened the global public health system and may keep simmering for years.The frequent emergence of immune escape variants have spurred the search for prophylactic vaccines and therapeutic antibodies that confer broad protection against SARS-CoV-2 variants.Here we show that the bivalency of an affinity maturated fully human singledomain antibody(n3113.1-Fc)exhibits exquisite neutralizing potency against SARS-CoV-2 pseudovirus,and confers effective prophylactic and therapeutic protection against authentic SARS-CoV-2 in the host cell receptor angiotensin-converting enzyme 2(ACE2)humanized mice.The crystal structure of n3113 in complex with the receptor-binding domain(RBD)of SARS-CoV-2,combined with the cryo-EM structures of n3113 and spike ecto-domain,reveals that n3113 binds to the side surface of up-state RBD with no competition with ACE2.The binding of n3113 to this novel epitope stabilizes spike in up-state conformations but inhibits SARS-CoV-2 S mediated membrane fusion,expanding our recognition of neutralization by antibodies against SARS-CoV-2.Binding assay and pseudovirus neutralization assay show no evasion of recently prevalent SARS-CoV-2 lineages,including Alpha(B.1.1.7),Beta(B.1.351),Gamma(P.1),and Delta(B.1.617.2)for n3113.1-Fc with Y58L mutation,demonstrating the potential of n3113.1-Fc(Y58L)as a promising candidate for clinical development to treat COVID-19.Zhenlin Yang Yulu Wang Yujia Jin Yuanfei Zhu Yanling Wu Cheng Li Yu Kong Wenping Song Xiaolong Tian Wuqiang Zhan Ailing Huang Shanshan Zhou Shuai Xia Xiaoxu Tian Chao Peng Cuicui Chen Yibing Shi Gaowei Hu Shujuan Du Yuyan Wang Youhua Xie Shibo Jiang Lu Lu Lei Sun Yuanlin Song Tianlei Ying 2021Signal Transduction and Targeted Therapy2021,6,12:1
4Drug-peptide supramolecular hydrogel boosting transcorneal permeability and pharmacological activity via ligand-receptor interaction显示文摘Boosting transcorneal permeability and pharmacological activity of drug poses a great challenge in the field of ocular drug delivery.In the present study,we propose a drug-peptide supramolecular hydrogel based on anti-inflammatory drug,dexamethasone(Dex),and Arg-Gly-Asp(RGD)motif for boosting transcorneal permeability and pharmacological activity via the ligand-receptor interaction.The drug-peptide(Dex-SA-RGD/RGE)supramolecular hydrogel comprised of uniform nanotube architecture formed spontaneously in phosphate buffered saline(PBS,pH=7.4)without external stimuli.Upon storage at 4℃,25℃,and 37℃ for 70 days,Dex-SA-RGD in hydrogel did not undergo significant hydrolysis,suggesting great long-term stability.In comparison to Dex-SA-RGE,Dex-SA-RGD exhibited a more potent in vitro anti-inflammatory efficacy in lipopolysaccharide(LPS)-activated RAW 264.7 macrophages via the inhibition of nuclear factorкB(NF-κB)signal pathway.More importantly,using drug-peptide supramolecular hydrogel labeled with 7-nitro-2,1,3-benzoxadiazole(NBD),the Dex-SA-K(NBD)RGD showed increased performance in terms of integrin targeting and cellular uptake compared to Dex-SA-K(NBD)RGE,as revealed by cellular uptake assay.On topical instillation in rabbit’s eye,the proposed Dex-SA-K(NBD)RGD could effectively enhance the transcorneal distribution and permeability with respect to the Dex-SA-K(NBD)RGE.Overall,our findings demonstrate the performance of the ligand-receptor interaction for boosting transcorneal permeability and pharmacological activity of drug.Lin Chen Jie Deng Ailing Yu Yuhan Hu Bo Jin Pengyuan Du Jianhong Zhou Lei Lei Yuan Wang Serhii Vakal Xingyi Li 2022Bioactive Materials2022,7,4:0
5Thermodynamic equilibrium diagram of the chlorine-titanium system显示文摘The chemical and electrochemical equilibria of the chlorine-titanium system in the presence of gaseous phase were investigated. Many species, which consisted of chlorine and titanium, were considered. Various thermodynamic equi- libria were calculated in the different pressures at different temperatures. The calculated results were shown as log p-1/T and E-T diagrams. These diagrams may be used as important tools for corrosion study and titanium production. 6Side chain engineering of quinoxaline-based small molecular nonfullerene acceptors for high-performance poly(3-hexylthiophene)-based organic solar cells显示文摘Poly(3-hexylthiophene)(P3HT)is one of the most used semiconducting polymers for organic photovoltaics because it has potential for commercialization due to its easy synthesis and stability.Although the rapid development of the small molecular non-fullerene acceptors(NFAs)have largely improved the power conversion efficiency(PCE)of organic solar cells(OSCs)based on other complicated p-type polymers,the PCE of P3HT-based OSCs is still low.In addition,the design principle and structure-properties correlation for the NFAs matching well with P3HTare still unclear and need to be investigated in depth.Here we designed a series of NFAs comprised of acceptor(A)and donor(D)units with an A2-A1-D-A1-A2 configuration.These NFAs are abbreviated as Qx3,Qx3 b and Qx3c,where indaceno[1,2-b:5,6-b′]dithiophene(IDT),quinoxaline(Qx)and 2-(1,1-dicyanomethylene)rhodanine serve as the middle D,bridged A1 and the end group A2,respectively.By subtracting the phenyl side groups appended on both IDT and Qx skeletons,the absorption spectra,energy levels and crystallinity could be regularly modulated.When paired with P3 HT,three NFAs show totally different photovoltaic performance with PCEs of 3.37%(Qx3),6.37%(Qx3b)and 0.03%(Qx3 c),respectively.From Qx3 to Qx3b,the removing of phenyl side chain in the middle IDT unit results in the increase of crystallinity and electron mobility.However,after subtracting all the grafted phenyl side groups on both IDT and Qx units,the final molecule Qx3 c exhibits the lowest PCE of only 0.03%,which is mainly attributed to the serious phase-separation of the blend film.These results demonstrate that optimizing the substituted position of phenyl side groups for A2-A1-D-A1-A2 type NFAs is vital to regulate the optoelectronic property of molecule and morphological property of active layer for high performance P3HT-based OSCs.Bo Xiao Qianqian Zhang Gongqiang Li Mengzhen Du Yanfang Geng Xiangnan Sun Ailing Tang Yingliang Liu Qiang Guo Erjun Zhou 2020Science China Chemistry2020,63,2:0

8Anti-corrosive, weatherproof and self-healing polyurethane developed from hydrogenated hydroxyl-terminated polybutadiene toward surface-protective applications显示文摘Self-healing polyurethane(PU)faces aging deterioration due to active dynamic bonds,which remain a challenging predicament for practical use.In this work,a novel strategy is developed to address this predicament by leveraging the hydrophobicity and gas barrier of hydrogenated hydroxyl-terminated polybutadiene(HHPB).The dynamic oxime-carbamate bonds derived from 2,4-pentanedione dioxime(PDO)enable the elastomer to exhibit surface self-repairability upon applied mild heat and achieve~99.5%mechanical self-healing efficiency.The mechanical properties remained nearly intact after 30-d exposure to thermal oxidation,xenon lamp,acids,bases,and salts.Gas permeability,positron annihilation lifetime spectroscopy(PALS),and contact angle measurements reveal the pivotal role of gas barrier,free volume,and hydrophobicity in blocking undesirable molecules and ions which effectively protects the elastomer from deterioration.HHPB-PU also exhibits excellent adhesion to steel substrate.The shear strength achieves(3.02±0.42)MPa after heating at 80°C for 4 h,and(3.06±0.2)MPa after heating at 130°C for 0.5 h.Regarding its outstanding anti-corrosive and weatherproof performances,this self-healable elastomer is a promising candidate in surface-protective applications.Yuanyuan LIU Xin DU Hui WANG Yu YUAN Liuhe WEI Xingjiang LIU Ailing SUN Yuhan LI 2022Frontiers of Materials Science2022,16,2:0

10High fidelity virtual try-on network via semantic adaptation and distributed componentization显示文摘Image-based virtual try-on systems have significant commercial value in online garment shopping.However,prior methods fail to appropriately handle details,so are defective in maintaining the original appearance of organizational items including arms,the neck,and in-shop garments.We propose a novel high fidelity virtual try-on network to generate realistic results.Specifically,a distributed pipeline is used for simultaneous generation of organizational items.First,the in-shop garment is warped using thin plate splines(TPS)to give a coarse shape reference,and then a corresponding target semantic map is generated,which can adaptively respond to the distribution of different items triggered by different garments.Second,organizational items are componentized separately using our novel semantic map-based image adjustment network(SMIAN)to avoid interference between body parts.Finally,all components are integrated to generatethe overall result by SMIAN.A priori dual-modalinformation is incorporated in the tail layers of SMIAN to improve the convergence rate of the network.Experiments demonstrate that the proposed method can retain better details of condition information than current methods.Our method achieves convincing quantitative and qualitative results on existing benchmark datasets.Chenghu Du Feng Yu Minghua Jiang Ailing Hua Yaxin Zhao Xiong Wei Tao Peng Xinrong Hu 2022Computational Visual Media2022,8,4:0
