1PPARγ expression by rambutan peel extract in obesity rat model-induced high-calorie diet显示文摘Objective: To monitor the physiological characteristics and genes expression of obesity rat model after rambutan peel extract(RPE) treatment.Methods: Twenty-four 12-week-old male rats were divided into 4 groups: normal,obesity, obesity treated with ellagic acid(O-EA) and obesity treated with RPE30(ORPE30). Physiological characteristics were monitored by measuring body weight, calorie intake, size of adipocyte and level of triglyceride. Peroxisome proliferator activated receptor gamma(PPARg), CCAAT/enhancer-binding proteins a and fatty acid binding protein 4(FABP4) expression were observed using immunohistochemistry, Western blotting and quantitative RT-PCR methods.Results: Body weight gain of O-EA and O-RPE30 rats were lower than obesity group and size of adipocyte cells were smaller than obesity group(P < 0.05), but when we compared to normal group, those groups had higher body weight gain and larger adipocyte cells. The level of triglycerides, protein expression of PPARg and m RNA level of FABP4 genes were significantly downregulated on O-EA and O-RPE30 compared to obesity group(P < 0.05). Our results indicated that RPE had potential substance as inhibitor of body weight gain, declining of size of adipocyte, level of triglycerides, PPARg expression and m RNA level of FABP4 gene on obesity rat model.Conclusions: RPE have anti-obesity activity by inhibiting body weight gain, declining size of adipocyte, decreasing triglyceride, PPARg expression and m RNA level of FABP4 gene on obesity rat model.Sri Rahayu Lestari Muhammad Sasmito Djati Achmad Rudijanto Fatchiyah Fatchiyah 2015Asian Pacific Journal of Tropical Biomedicine2015,5,10:1

5Frequency–magnitude of landslides affected by the 27–29 November 2017 Tropical Cyclone Cempaka in Pacitan, East Java显示文摘Tropical cyclone(TC) Cempaka which occurred on 27–29 November 2017 has caused floods, landslides, and strong winds in certain areas of Java Island. Pacitan Regency was the most severely affected by TC Cempaka. The landslide frequency–area distribution curve of event inventory i.e. TC Cempaka can help to understand landslide susceptibility, hazard, vulnerability, and risk. Landslides were identified by using a local government database and by comparing pre-and post-event high-resolution satellite imageries. Field investigation was carried out in March 2018 to November 2018 to verify the landslide location and update the information. Power law, inverse gamma, and double Pareto model were employed to describe the frequency–magnitude of landslide(mLS) triggered by TC Cempaka. The exponent β values of power law, inverse gamma, and double Pareto were 2.6±0.28(fitted for 8.5% of dataset), 2.2±0.08(fitted for 83% of dataset), and 2.3±0.09(best fitted for dataset), respectively. The P-values were 0.51, 0.67, and 0.91 for power law, inverse gamma, and double Pareto, respectively. This study revealed that rollover occurred at 200 and 300 m2 for double Pareto and inverse gamma, respectively. The cutoff points totaled 1096.49 ± 236.44 and 7235.4 ± 1896.7 m2 for double Pareto and power law, respectively. Rollover phenomenon was real and existed in the dataset because it was far from the minimum resolvable size of the landslide that the authors can delineate from the satellite images. mLS for Pacitan was distributed at around 2 to 4. The magnitude of large landslides was 3.2, that of medium landslides was less than 3, and that of small landslides was almost 4. Numerical estimation calculated a fixed mLS=3.01. Comparison analysis of β values obtained from several landslide inventories triggered by heavy rainfall suggests that the variability of β is related to the intensity and duration of rainfall. Triggering events, such as intensity and duration of rainfall, affect the proportion of large landslides that occur in an area. 6Designing Cyclopentapeptide Inhibitor of Neuraminidase H5N1 Virus Through Molecular and Pharmacology Simulations显示文摘Highly Pathogenic Avian Influenza(HPAI) H5N1 has attracted much attention as a potential pandemic virus in humans, which makes death inevitable in humans. Neuraminidase(NA) has an important role in viral replication. Thus, it is an attractive target when designing anti-influenza virus drug. However, evolving viruses cause some anti-viral drugs to be ineffective, as they show resistance to them. Selection of peptides as drug candidates is important for the peptide-receptor activity and good selectivity. Cyclic bonds in the peptide ligand design aim to improve the stability of the system and remove the obstacles in drug metabolism. The design is based on the polarity of the ligand and amino acid residues in the active site of NA. The results are 4200 cyclic pentapeptides as potential lead compounds. Docking simulations were conducted using MOE 2008.10 and were screened based on the value of the binding energy(?Gbinding). ADME-Tox prediction assay was conducted on the selected ligands.Intra- and inter-molecular interactions, as well as changes in the form of bonds, were tested by molecular dynamics simulations at temperatures of 310 K and 312 K. The results of the docking simulations and toxicity prediction assay show that there are two ligands that have a residual interaction with the target protein: CLDRC and CIWRC. These two ligands have ?Gbindingvalues of –40.5854 and –39.9721 kcal/mol(1 kcal/mol = 4.18 k J/mol). These ligands are prone to be mutagenic and carcinogenic, and they have a good oral bioavailability. The results show that the molecular dynamics of both ligand CLDRC and CIWRC are more feasible at the temperature of 312 K.
