1The ATF/CREB site is the key element for transcription of the human RNA methyltransferase like 1 (RNMTL1) gene, a newly discovered 17p13.3 gene显示文摘The human RNA methyltransferase like i gene (RNMTL1) is one of thirteen newly discovered geneswithin a 116 Kb segment of the chromosome 17p13.3 that suffers from a high frequent loss of heterozygosityin human hepatocellular carcinoma in China[1-5]. To understand the molecular mechanisms underlyingtranscription control of the RNMTL1 gene in human cancers, we decline using of the conventional approachwhere the cis-elements bound by the known transcription factors are primary targets, and carried out thesystematic analyses to dissect the promoter structure and identify/characterize the key cis-elements thatare responsible for its strong expression in cell. The molecular approaches applied included 1, the primerextension for mapping of the transcription starts; 2, the transient transfection/reporter assays on a largenumber of deletion and site-specific mutants of the promoter segment for defining the minimal promoterand the crucial elements within; and 3, the electrophoresis mobility shift assay with specific antibodies forreconfirming the nature of the transcription factors and their cognate cis-elements. We have shown that theinteraction of an ATF/CREB element (-38 to -31) and its cognate transcription factors play a predominantrole in the promoter activity of the RNMTL1 gene. The secondary DNA structures of the ATF/CREBelement play a more vital role in the protein-DNA interaction. 2Perspectives on gene expression regulation techniques in Drosophila显示文摘Gene expression regulation, including loss-of-function and gain-of-function assays, is a powerful method to study developmental and disease mechanisms. Drosophila melanogaster is an ideal model system particularly well-equipped with many genetic tools. In this review, we describe and discuss the gene expression regulation techniques recently developed and their applications, including the CRISPR/Cas9-triggered heritable mutation system, CRISPR/dCas9-based transcriptional activation(CRISPRa) system,and CRISPR/dCas9-based transcriptional repression(CRISPRi) system, as well as the next-generation transgenic RNAi system. 3circBIRC6 contributes to the development of non-small cell lung cancer via regulating microRNA-217/amyloid beta precursor protein binding protein 2 axis显示文摘Background:Circular RNAs(circRNAs)are considered to be important regulators in cancer biology.In this study,we focused on the effect of circRNA baculoviral inhibitor of apoptosis protein(IAP)repeat containing 6(circBIRC6)on non-small cell lung cancer(NSCLC)progression.Methods:The NSCLC and adjacent non-tumor tissues were collected at Shanghai Ninth People's Hospital.Quantitative real-time polymerase chain reaction was conducted for assessing the levels of circBIRC6,amyloid beta precursor protein binding protein 2(APPBP2)messenger RNA(mRNA),baculoviral IAP repeat containing 6 mRNA(BIRC6),and microRNA-217(miR-217).Western blot assay was adopted for measuring the protein levels of APPBP2,E-cadherin,N-cadherin,and vimentin.Colony formation assay,transwell assay,and flow cytometry analysis were utilized for evaluating cell colony formation,metastasis,and apoptosis.Dualluciferase reporter assay and RNA immunoprecipitation assay were carried out to determine the interaction between miR-217 and circBIRC6 and APPBP2 in NSCLC tissues.The murine xenograft model assay was used to investigate the function of circBIRC6 in tumor formation in vivo.Differences were analyzed via Student's t test or one-way analysis of variance.Pearson's correlation coefficient analysis was used to analyze linear correlation.Results:CircBIRC6 was overexpressed in NSCLC tissues and cells.Knockdown of circBIRC6 repressed the colony formation and metastasis and facilitated apoptosis of NSCLC cells in vitro and restrained tumorigenesis in vivo.Mechanically,circBIRC6 functioned as miR-217 sponge to promote APPBP2 expression in NSCLC cells.MiR-217 inhibition rescued circBIRC6 knockdown-mediated effects on NSCLC cell colony formation,metastasis,and apoptosis.Overexpression of miR-217 inhibited the malignant phenotypes of NSCLC cells,while the effects were abrogated by elevating APPBP2.Conclusion:CircBIRC6 aggravated NSCLC cell progression by elevating APPBP2 via sponging miR-217,which might provide a fresh perspective on NSCLC therapy.Da Ni Jiping Teng Youshuang Cheng Zhijun Zhu Bufeng Zhuang Zhiyin Yang 2022Chinese Medical Journal2022,,6:2

9De?ning gene networks controlling the maintenance and function of the differentiation niche by an in vivo systematic RNAi screen显示文摘In the Drosophila ovary, escort cells (ECs) extrinsically control germline stem cell (GSC) maintenance and progeny differentiation. However, the underlying mechanisms remain poorly understood. In this study,we identified 173 EC genes for their roles in controlling GSC maintenance and progeny differentiation by using an in vivo systematic RNAi approach. Of the identified genes, 10 and 163 are required in ECs to promote GSC maintenance and progeny differentiation, respectively. The genes required for progeny differentiation fall into different functional categories, including transcription, mRNA splicing, protein degradation, signal transduction and cytoskeleton regulation. In addition, the GSC progeny differentiation defects caused by defective ECs are often associated with BMP signaling elevation, indicating that preventing BMP signaling is a general functional feature of the differentiation niche. Lastly, exon junction complex (EJC) components, which are essential for mRNA splicing, are required in ECs to promote GSC progeny differentiation by maintaining ECs and preventing BMP signaling. 20Effect of intracellular water release on hydro-mechanical behaviors of high kitchen waste content municipal solid waste显示文摘The release of intracellular water during degradation process contributes to the great leachate production and settlement of landfilled high kitchen waste content MSW(HKWC-MSW). An oven-drying and absorbent-paper combined method was proposed to measure the intracellular and interparticle water contents of HKWC-MSW. Two degradation experiments were carried out to study the release process of intracellular water and its effect on the hydro-mechanical behaviors of HKWC-MSW.It was found that the two degradation experiments showed similar degradation behaviors with BOD/COD decreasing with time in the early stage. In the first degradation experiment, most intracellular water was released during the first two months, and the degradation of degradable matter in kitchen waste(KW) was much slower than the release process of intracellular water. The particle size became smaller and the overall grain specific gravity increased during the fast release process of intracellular water.In the second degradation experiment, after two-year degradation, the total leachate production was about 45.2% of the initial wet weight of HKWC-MSW specimen. Water retention capacity θ_f increased from 0.23 to 0.58 during 1–69 d, which might be caused by the decrease of particle size and compression of waste skeleton. As almost all the intracellular water was released after 80-day degradation, during the latter stage of leachate drainage under gravity, θ_f decreased and was close to the total volumetric water content. The total compression strain was about 0.39. The secondary compression strain during 1–80 d(i.e., about 0.07)was considered to be mainly resulted by the release of intracellular water and the subsequent drainage of leachate, and it accounted for about 22.6% of the total secondary compression strain.XU XiaoBing NI Da CHEN YunMin XU Hui YANG QinFeng XU SiFa 2019Science China(Technological Sciences)2019,62,11:1
