| 1 | Autophagy Plays a Potential Role in the Process of Sea Cucumber Body Wall “Melting” Induced by UV Irradiation显示文摘The changes of tissue appearances and structures in the process of UV-induced 'melting' for sea cucumber (Stichopus japonicus) body wall were studied.And the localization and determination of acid phosphatase (ACP),Cathepsin B and Cathepsin L activities were also investigated.The results show that the connective tissue was damaged with many hollows emerging and the regular collagen bundles were broken apart into irregular fragments.Margination of condensed chromatin at the nuclear membrane was observed.Both Golgi’s body and endoplasmic reticulum swelled,curled,and eventually double-or multi-lamellar vesicles were formed.A number of autophagic vesicles distributed in all through the whole cytoplasm.ACP be-comes more active after UV irradiation.The activities of cathepsin B and cathepsin L increased in UV-treated sea cucumbers and both achieved their maximum under certain conditions.It indicates that autophagy plays a potential role in the 'melting' process for sea cucumber body wall induced by UV irradiation. | ZHU Beiwei ZHENG Jie ZHANG Zongshen DONG Xiuping ZHAO Lulu Tada Mikiro | 2008 | Wuhan University Journal of Natural Sciences2008,13,2: | 23 |

Ta MYB72 is a MYB family transcription factor localized to the nucleus. Three Ta MYB72 homologs,Ta MYB72-A, Ta MYB72-B and Ta MYB72-D, cloned from hexaploid wheat were mapped to the short arm of the group 6 chromosomes. Under the long-day conditions,over-expression of the Ta MYB72 in rice shortened the flowering time by approximately 12 d. | 3 | Bimetallic nickel cobalt sulfides with hierarchical coralliform architecture for ultrafast and stable Na-ion storage显示文摘A series of bimetallic nickel cobalt sulfides with hierarchical micro/nano architectures were fabricated via a facile synthesis strategy of bimetallic micro/nano structure precursor construction-anion exchange via solvothermal method. Among the nickel cobalt sulfides with different Ni/Co contents, the coral-like Ni1.01Co1.99S4 (Ni/Co, 1/2) delivers ultrafast and stable Na-ion storage performance (350 mAh·g−1 after 1,000 cycles at 1 A·g−1 and 355 mAh·g−1 at 5 A·g−1). The remarkable electrochemical properties can be attributed to the enhanced conductivity by co-existence of bimetallic components, the unique coral-like micro/nanostructure, which could prevent structural collapse and self-aggregation of nanoparticles, and the easily accessibility of electrolyte, and fast Na+ diffusion upon cycling. Detailed kinetics studies by a galvanostatic intermittent titration technique (GITT) reveal the dynamic change of Na+ diffusion upon cycling, and quantitative kinetic analysis indicates the high contribution of pseudocapacitive behavior during charge-discharge processes. Moreover, the ex-situ characterization analysis results further verify the Na-ion storage mechanism based on conversion reaction. | 12 | Exploration of the potential mechanism of Danggui Shaoyao powder in the treatment of endometriosis based on bioinformatics显示文摘Background:Endometriosis is a hormone-and organ-dependent disease with unclear pathogenesis.Danggui Shaoyao powder(DSP)has been used for the treatment of endometriosis for many years and has been proven to be effective,but its underlying molecular mechanisms remain unclear.In this study,we explored the molecular targets and pathways of DSP in the treatment of endometriosis mainly from the perspective of network pharmacology,and provided some novel ideas for managing this disease.Methods:In the present study,we focused on the underlying mechanisms between DPS and endometriosis,via oral bioavailability screening,drug similarity assessment,target identification,network analysis,drug relocation,and molecular docking using network pharmacology.Results:By bioinformatics,54 active components of DSP were predicted from the 534 components provided by the TCMSP database.We then found 85 validated targets and 6059 predicted targets for herbal components,and 2241 targets for endometriosis.After mapping,there were 255 targets in common.After that,network analysis was performed to screen key networks and core targets.Ninetynine indirect targets were collected from 14 direct ones.GO and KEGG analyses identified 63 enriched functions and 10 pathways(P<.01).According to the disease target,dipyridamole could undergo drug repositioning for endometriosis(score?1).The mechanism of action of dipyridamole on endometriosis was compared with that of DSP.Finally,eight targets,namely,AR,ESR1,HMGCR,NOS2,NR3C1,PPARG,PTGES,and PTGS2,were validated by molecular docking,suggesting that the probable molecular mechanisms by which DSP treats endometriosis are through seven validated pathways:hsa05200,hsa04915,hsa01100,hsa00900,hsa04920,hsa01130,and hsa00590.Conclusion:In this study,we uncovered the mechanism by which DSP can treat endometriosis using a drugecomponentetarget interaction network.This approach can provide some novel ideas for the development of Chinese medicine and the clinical management of endometriosis. | Xiuping Zhang Lulu Zhang Qi Wang Xinhui Sun Yang Dong Yu Xing Xiaona Ma | 2019 | Journal of Traditional Chinese Medical Sciences2019,6,4: | 1 |
| 13 | JUNB mediates oxaliplatin resistance via the MAPK signaling pathway in gastric cancer by chromatin accessibility and transcriptomic analysis显示文摘tients,and chemotherapy resistance is one of the main reasons for treatment failure.Thus,it is important to reveal the mechanism of oxaliplatin resistance and to seek effective intervention strategies to improve chemotherapy sensitivity,thereby improving the survival and prognosis of gastric cancer patients.To understand the molecular mechanisms of oxaliplatin resistance,we generate an oxaliplatin-resistant gastric cancer cell line and conduct assay for transposase-accessible chromatin sequencing(ATAC-seq)and RNA sequencing(RNA-seq)for both parental and oxaliplatin-resistant AGS cells.A total of 3232 genomic regions are identified to have higher accessibility in ox-aliplatin-resistant cells,and DNA-binding motif analysis identifies JUNB as the core transcription factor in the regulatory network.JUNB is overexpressed in oxaliplatin-resistant gastric cancer cells,and its upregulation is associated with poor prognosis in gastric cancer patients,which is validated by our tissue microarray data.Moreover,chromatin immunoprecipitation sequencing(ChIP-seq)analysis reveals that JUNB binds to the tran-scriptional start site of key genes involved in the MAPK signaling pathway.Knockdown of JUNB inhibits the MAPK signaling pathway and restores sensitivity to oxaliplatin.Combined treatment with the ERK inhibitor piperlongu-mine or MEK inhibitor trametinib effectively overcomes oxaliplatin resistance.This study provides evidence that JUNB mediates oxaliplatin resistance in gastric cancer by activating the MAPK pathway.The combination of MAPK inhibitors with oxaliplatin overcomes resistance to oxaliplatin,providing a promising treatment opportunity for oxaliplatin-resistant gastric cancer patients. | Suyao Li Yichou Wei Xun Sun Mengling Liu Mengxuan Zhu Yitao Yuan Jiayu Zhang Yu Dong Keshu Hu Sining Ma Xiuping Zhang Bei Xu Hesheng Jiang Lu Gan Tianshu Liu | 2023 | Acta Biochimica et Biophysica Sinica2023,55,11: | 0 |
