| 4 | Structural optimization and biological evaluation of 1,5-disubstituted pyrazole-3-carboxamines as potent inhibitors of human 5-lipoxygenase显示文摘Human 5-lipoxygenase(5-LOX)is a well-validated drug target and its inhibitors are potential drugs for treating leukotriene-related disorders.Our previous work on structural optimization of the hit compound 2 from our in-house collection identified two lead compounds,3a and 3b,exhibiting a potent inhibitory profile against 5-LOX with IC_(50) values less than 1 mmol/L in cell-based assays.Here,we further optimized these compounds to prepare a class of novel pyrazole derivatives by opening the fusedring system.Several new compounds exhibited more potent inhibitory activity than the lead compounds against 5-LOX.In particular,compound 4e not only suppressed lipopolysaccharide-induced inflammation in brain inflammatory cells and protected neurons from oxidative toxicity,but also significantly decreased infarct damage in a mouse model of cerebral ischemia.Molecular docking analysis further confirmed the consistency of our theoretical results and experimental data.In conclusion,the excellent in vitro andin vivo inhibitory activities of these compounds against 5-LOX suggested that these novel chemical structures have a promising therapeutic potential to treat leukotriene-related disorders. | Yu Zhou Jun Liu Mingyue Zheng Shuli Zheng Chunyi Jiang Xiaomei Zhou Dong Zhang Jihui Zhao Deju Ye Mingfang Zheng Hualiang Jiang Dongxiang Liu Jian Cheng Hong Liu | 2016 | Acta Pharmaceutica Sinica B2016,6,1: | 1 |

| 8 | Interaction between dodecyl oxypropyl β-hydroxyltrimethylammonium bromide and Xanthan:MesoDyn simulation and binding isotherm measurements显示文摘MesoDyn density functional simulation method is used to study the interactions between dodecyl oxypropyl β-hydroxyltrimethylammonium bromide (C12NBr) and Xanthan (XC). The micro dynamic process of aggregate formation and the aggregate morphology are reported. Interaction between XC and nonyphenyloxypropyl β-hydroxyltrimethylammonium bromide (C9phNBr) is compared with that between XC and C12NBr. Simulation results show that the aggregate morphology of XC/C12NBr and XC/C9phNBr is of rod-like shape with helix characteristic. The binding of C9phNBr to XC is more difficult than that of C12NBr to XC. In addition, three stages for the dynamic evolution of surfactant binding to XC are observed. | 11 | Water-in-Water Emulsions,Ultralow Interfacial Tension,and Biolubrication显示文摘A water-in-water(W/W)emulsion consists of droplets formed by the spontaneous liquid-liquid separation of two immiscible aqueous phases.The inherent properties of the W/W interfaces,low or ultralowinterfacial tension(γ_(W/W)=1-1000μN/m)and a large thickness of several nanometers,beget the poor inherent stability of emulsions.Herein,we report a nanofibril emulsifier having Schiff base reactivity to generate a W/W emulsion.The W/W emulsion has superior stability(stable>6 months)because collagen nanofibrils,acting as a stabilizer of W/W emulsions,can simultaneously satisfy the requirements of sizeandoverall coverage ratio of the phase interfaces.W/Wemulsions having γ_(W/W)∼10μN/m were used as synthetic synovial fluids,showing superior lubrication performance with a coefficient of friction in the range of 0.003-0.011,which has been demonstrated to be suitable for joint lubrication.An intraarticular injection assessment further confirmed this protective effect on articular cartilage in vivo.Our study reveals the mechanism of emulsion stabilization and opens up the possibility of osteoarthritis(OA)treatment using the biolubrication effects of W/W emulsions for lubricated artificial implant surfaces. | Yitong Wang Jin Yuan Yunpeng Zhao Ling Wang Luxuan Guo Lei Feng Jiwei Cui Shuli Dong Shanhong Wan Weimin Liu Heinz Hoffmann Kiet Tieu Jingcheng Hao | 2022 | CCS Chemistry2022,4,6: | 0 |
