1An Investigation of Oxidative DNA Damage in Pharmacy Technicians Exposed to Antineoplastic Drugs in Two Chinese Hospitals Using The Urinary 8-OHdG Assay显示文摘Objective To investigate oxidative DNA damage in pharmacy technicians preparing antineoplastic drugs at the PIVAS (Pharmacy Intravenous Admixture Service) in two Chinese hospitals.Methods Urinary 8-OHdG served as a biomarker.5-Fluorouracil (5-FU) concentrations in air,masks and gloves were determined.The spill exposure of each PIVAS technician to antineoplastic drugs was investigated.Eighty subjects were divided into exposed group I,II,and control group I,II.Results 5-FU concentration ratios for gloves and masks in exposed group I were significantly higher than those in exposed group II (P<0.05 or P<0.01).The average urinary 8-OHdG concentrations in exposed group I,control group I,exposed group II,and control group II were 14.69±0.93,10.68±1.07,10.57±0.55,and 11.96±0.73 ng/mg Cr,respectively.Urinary 8-OHdG concentration in exposed group I was significantly higher than that in control group I or that in exposed group II (P<0.01).There was a significant correlation between urinary 8-OHdG concentrations and spill frequencies per technician (P<0.01).Conclusion There was detectable oxidative DNA damage in PIVAS technicians exposed to antineoplastic drugs.This oxidative DNA damage may be associated with their spill exposure experience and contamination of their personal protective equipment.HUANG Yu Wen JIAN Le ZHANG Mei Bian ZHOU Quan YAN Xiao Feng HUA Xu Dong ZHOU Ying HE Ji Liang 2012Biomedical and Environmental Sciences2012,25,1:30

3Combined acoustic radiation force impulse, aminotransferase to platelet ratio index and Forns index assessment for hepatic fibrosis grading in hepatitis B显示文摘AIM: To investigate the combined diagnostic accuracy of acoustic radiation force impulse(ARFI), aspartate aminotransferase to platelet ratio index(APRI) and Forns index for a non-invasive assessment of liver fibrosis in patients with chronic hepatitis B(CHB). METHODS: In this prospective study, 206 patients had CHB with liver fibrosis stages F0-F4 classified by METAVIR and 40 were healthy volunteers were measured by ARFI, APRI and Forns index separately or combined as indicated. RESULTS: ARFI, APRI or Forns index demonstrated a significant correlation with the histological stage(all P < 0.001). According to the AUROC of ARFI and APRI for evaluating fibrotic stages more than F2, ARFI showed an enhanced diagnostic accuracy than APRI(P < 0.05). The combined measurement of ARFI and APRI exhibited better accuracy than ARFI alone when evaluating ≥ F2 fibrotic stage(Z = 2.77, P = 0.006). Combination of ARFI, APRI and Forns index did not obviously improve the diagnostic accuracy compared to the combination of ARFI and APRI(Z = 0.958, P = 0.338). 7Topographic differentiations of biological soil crusts and hydraulic properties in fixed sand dunes, Tengger Desert显示文摘Biological soil crusts(BSCs) play an important role in surface soil hydrology. Soils dominated with moss BSCs may have higher infiltration rates than those dominated with cyanobacteria or algal BSCs. However, it is unnown whether improved infiltration in moss BSCs is accompanied by an increase in soil hydraulic conductivity or water retention capacity. We investigated this question in the Tengger Desert, where a 43-year-old revegetation program has promoted the formation of two distinct types of BSCs along topographic positions, i.e. the moss-dominated BSCs on the interdune land and windward slopes of the fixed sand dunes, and the algal-dominated BSCs on the crest and leeward slopes. Soil water retention capacity and hydraulic conductivity were measured using an indoor evaporation method and a field infiltration method. And the results were fitted to the van Genuchten–Mualem model. Unsaturated hydraulic conductivities under greater water pressure(<–0.01 MPa) and water retention capacities in the entire pressure head range were higher for both crust types than for bare sand. However, saturated and unsaturated hydraulic conductivities in the near-saturation range(>–0.01 MPa) showed decreasing trends from bare sand to moss crusts and to algal crusts. 10Force-induced charge carrier storage:a new route for stress recording显示文摘Stress sensing is the basis of human-machine interface,biomedical engineering,and mechanical structure detection systems.Stress sensing based on mechanoluminescence(ML)shows significant advantages of distributed detection and remote response to mechanical stimuli and is thus expected to be a key technology of next-generation tactile sensors and stress recorders.However,the instantaneous photon emission in ML materials generally requires real-time recording with a photodetector,thus limiting their application fields to real-time stress sensing.In this paper,we report a force-induced charge carrier storage(FICS)effect in deep-trap ML materials,which enables storage of the applied mechanical energy in deep traps and then release of the stored energy as photon emission under thermal stimulation.The FICS effect was confirmed in five ML materials with piezoelectric structures,efficient emission centres and deep trap distributions,and its mechanism was investigated through detailed spectroscopic characterizations.Furthermore,we demonstrated three applications of the FICS effect in electronic signature recording,falling point monitoring and vehicle collision recording,which exhibited outstanding advantages of distributed recording,longterm storage,and no need for a continuous power supply.The FICS effect reported in this paper provides not only a breakthrough for ML materials in the field of stress recording but also a new idea for developing mechanical energy storage and conversion systems.Yixi Zhuang Dong Tu Changjian Chen Le Wang Hongwu Zhang Hao Xue Conghui Yuan Guorong Chen Caofeng Pan Lizong Dai Rong-Jun Xie 2020Light(Science & Applications)2020,9,1:10
11Ca_(1−x)Li_(x)Al_(1−x)Si_(1+x)N_(3):Eu^(2+) solid solutions as broadband,color-tunable and thermally robust red phosphors for superior color rendition white light-emitting diodes显示文摘Color rendition,luminous efficacy and reliability are three key technical parameters for white light-emitting diodes(wLEDs)that are dominantly determined by down-conversion phosphors.However,there is usually an inevitable trade-off between color rendition and luminescence efficacy because the spectrum of red phosphor(that is,spectral broadness and position)cannot satisfy them simultaneously.In this work,we report a very promising red phosphor that can minimize the aforementioned trade-off via structure and band-gap engineering,achieved by introducing isostructural LiSi_(2)N_(3) into CaAlSiN_(3):Eu^(2+).The solid solution phosphors show both substantial spectra broadening(88→117 nm)and blueshift(652→642 nm),along with a significant improvement in thermal quenching(only a 6%reduction at 150℃),which are strongly associated with electronic and crystal structure evolutions.The broadband and robust red phosphor thus enables fabrication of super-high color rendering wLEDs(Ra=95 and R9=96)concurrently with the maintenance of a high-luminous efficacy(101 lm W^(−1)),validating its superiority in highperformance solid state lightings over currently used red phosphors.Le Wang Rong-Jun Xie Yuanqiang Li Xiaojun Wang Chong-Geng Ma Dong Luo Takashi Takeda Yi-Ting Tsai Ru-Shi Liu Naoto Hirosaki 2016Light(Science & Applications)2016,5,1:9

13Mechanical Properties and Microstructure of In Situ TiB_2-7055 Composites显示文摘In order to fabricate a kind of high strength particulate reinforced aluminum-matrix composites, the high strength aluminum alloy 7055 was selected as a matrix. Composites reinforced with varying amounts of TiB2 particles were synthesized using the in situ method, and their mechanical properties and microstructure were analyzed. It is found that the in situ TiB2 particles sized from 50 to 400 nm uniformly disperse in the matrix. With the weight fraction of TiB2 particles increasing, the elastic modulus as well as the yield strength and the ultimate tensile strength increase, while the ductility decrease. 14Whole-genome sequencing provides insights into the genetic diversity and domestication of bitter gourd(Momordica spp.)显示文摘Bitter gourd(Momordica charantia)is a popular cultivated vegetable in Asian and African countries.To reveal the characteristics of the genomic structure,evolutionary trajectory,and genetic basis underlying the domestication of bitter gourd,we performed whole-genome sequencing of the cultivar Dali-11 and the wild small-fruited line TR and resequencing of 187 bitter gourd germplasms from 16 countries.The major gene clusters(Bi clusters)for the biosynthesis of cucurbitane triterpenoids,which confer a bitter taste,are highly conserved in cucumber,melon,and watermelon.Comparative analysis among cucurbit genomes revealed that the Bi cluster involved in cucurbitane triterpenoid biosynthesis is absent in bitter gourd.Phylogenetic analysis revealed that the TR group,including 21 bitter gourd germplasms,may belong to a new species or subspecies independent from M.charantia.Furthermore,we found that the remaining 166 M.charantia germplasms are geographically differentiated,and we identified 710,412,and 290 candidate domestication genes in the South Asia,Southeast Asia,and China populations,respectively.This study provides new insights into bitter gourd genetic diversity and domestication and will facilitate the future genomics-enabled improvement of bitter gourd.Junjie Cui Yan Yang Shaobo Luo Le Wang Rukui Huang Qingfang Wen Xiaoxia Han Nansheng Miao Jiaowen Cheng Ziji Liu Changyuan Zhang Chengcheng Feng Haisheng Zhu Jianwen Su Xinjian Wan Fang Hu Yu Niu Xiaoming Zheng Yulan Yang Dai Shan Zhensheng Dong Weiming He Narinder P.S.Dhillon Kailin Hu 2020Horticulture Research2020,7,1:7
15Systematic identification of endogenous RNA polymeraseⅢpromoters for efficient RNA guidebased genome editing technologies in maize显示文摘Single-guide RNA(sg RNA) is one of the two core components of the CRISPR(clustered regularly interspaced short palindromic repeat)/Cas(CRISPR-associated) genome-editing technology. We established an in vitro Traffic Light Reporter(TLR) system, which is designated as the same colors as traffic lights such as green, red and yellow were produced in cells. The TLR can be readily used in maize mesophyll protoplast for a quick test of promoter activity. The TLR assay indicates the variation in transcription activities of the seven Pol III promoters, from 3.4%(U6-1) to over 21.0%(U6-6). The U6-2 promoter, which was constructed to drive sg RNA expression targeting the Zm Wx1 gene, yielded mutation efficiencies ranging from 48.5% to 97.1%. Based on the reported and unpublished data, the in vitro TLR assay results were confirmed to be a readily system and may be extended to other plant species amenable to efficient genome editing via CRISPR/Cas. 16Moxibustion upregulates hippocampal progranulin expression显示文摘In China,moxibustion is reported to be useful and has few side effects for chronic fatigue syndrome,but its mechanisms are largely unknown.More recently,the focus has been on the wealth of information supporting stress as a factor in chronic fatigue syndrome,and largely concerns dysregulation in the stress-related hypothalamic-pituitary-adrenal axis.In the present study,we aimed to determine the effect of moxibustion on behavioral symptoms in chronic fatigue syndrome rats and examine possible mechanisms.Rats were subjected to a combination of chronic restraint stress and forced swimming to induce chronic fatigue syndrome.The acupoints Guanyuan(CV4) and Zusanli(ST36,bilateral) were simultaneously administered moxibustion.Untreated chronic fatigue syndrome rats and normal rats were used as controls.Results from the forced swimming test,open field test,tail suspension test,real-time PCR,enzyme-linked immunosorbent assay,and western blot assay showed that moxibustion treatment decreased m RNA expression of corticotropin-releasing hormone in the hypothalamus,and adrenocorticotropic hormone and corticosterone levels in plasma,and markedly increased progranulin m RNA and protein expression in the hippocampus.These findings suggest that moxibustion may relieve the behavioral symptoms of chronic fatigue syndrome,at least in part,by modulating the hypothalamic-pituitary-adrenal axis and upregulating hippocampal progranulin.Tao Yi Li Qi Ji Li Jing-jing Le Lei Shao Xin Du Jing-cheng Dong 2016Neural Regeneration Research2016,11,4:6
17A comparative study of the characteristics of two Coxsackie A virus type 16 strains (genotype B)显示文摘Coxsackie A virus is one of the major pathogens associated with hand, foot and mouth disease (HFMD). The etiological characteristics of Coxsackie A virus type 16 (CA16) are thought to correlate with the pathological process of its infection. Two CA16 strains that were isolated from a severe HFMD patient presented with different plaque forms. This observation, along with biological analysis, indicated that the differences in the strains' biological characteristics, such as proliferation kinetics and immunogenicity, correlate with differences in their pathogenicity toward neonatal mice. Furthermore, these differences are thought to be associated with the sequence of the 5′ non-coding region of the viral genome and the VP1 structural region sequence. 18A Bunyavirus-Inducible Ubiquitin Ligase Targets RNA Polymerase IV for Degradation during Viral Pathogenesis in Rice显示文摘The ubiquitin-proteasome system(UPS)is an important post-translational regulatory mechanism that controls many cellular functions in eukaryotes.Here,we show that stable expression of P3 protein encoded by Rice grassy stunt virus(RGSV),a negative-strand RNA virus in the Bunyavirales,causes developmental abnormities similar to the disease symptoms caused by RGSV,such as dwarfing and excess tillering,in transgenic rice plants.We found that both transgenic expression of P3 and RGSV infection induce ubiquitination and UPS-dependent degradation of rice NUCLEAR RNA POLYMERASE D1a(OsNRPD1a),one of two orthologs of the largest subunit of plant-specific RNA polymerase IV(Pol IV),which is required for RNA-directed DNA methylation(RdDM).Furthermore,we identified a P3-inducible U-box type E3 ubiquitin ligase,designated as P3-inducible protein 1(P3IP1),which interacts with OsNRPD1a and mediates its ubiquitination and UPS-dependent degradation in vitro and in vivo.Notably,both knockdown of OsNRPD1 and overexpression of P3IP1 in rice plants induced developmental phenotypes similar to RGSV disease symptomss.Taken together,our findings reveal a novel virulence mechanism whereby plant pathogens target host RNA Pol IV for UPS-dependent degradation to induce disease symptoms.Our study also identified an E3 ubiquitin ligase,which targets the RdDM compotent NRPD1 for UPS-mediated degradation in rice.Chao Zhang Ying Wei Le Xu Kang-Cheng Wu Liang Yang Chao-Nan Shi Guo-Yi Yang Dong Chen Fei-Fei Yu Qi Xie Shou-Wei Ding Jian-Guo Wu 2020Molecular Plant2020,13,6:5
19Threading dislocation density comparison between GaN grown on the patterned and conventional sapphire substrate by high resolution X-ray diffraction显示文摘GaN epifilms are grown on the patterned sapphire substrates (PSS) (0001) and the conventional sapphire substrates (CSS) (0001) by metal-organic chemical vapor deposition (MOCVD) using a novel two-step growth. High resolution X-ray diffraction (HR-XRD) is used to investigate the threading dislocation (TD) density of the GaN epifilms. The TD density is calculated from the ω-scans full width at half maximum (FWHM) results of HR-XRD. The edge dislocation destiny of GaN grown on the PSS is 2.7×108 cm-2, which is less than on the CSS. This is confirmed by the results of atomic force microscopy (AFM) measurement. The lower TD destiny indicates that the crystalline quality of the GaN epifilms grown on the PSS is improved compared to GaN epifilms grown on the CSS. The residual strains of GaN grown on the PSS and CSS are compared by Raman Scattering spectra. 20Integrative taxonomy of the Russet Bush Warbler Locustella mandelli complex reveals a new species from central China显示文摘Background: The Russet Bush Warbler Locustella(previously Bradypterus) mandelli complex occurs in mountains in the eastern Himalayas, southern China, Vietnam, the Philippines, and Indonesia. The taxonomy has been debated,with one(L. seebohmi) to four(L. seebohmi, L. mandelli, L. montis and L. timorensis) species having been recognised.Methods: We used an integrative approach, incorporating analyses of morphology, vocalizations and a molecular marker, to re-evaluate species limits in the L. mandelli complex.Results: We found that central Chinese L. mandelli differed from those from India through northern Southeast Asia to southeast China in plumage, morphometrics and song. All were easily classified by song, and(wing + culmen)/tail ratio overlapped only marginally. Both groups were reciprocally monophyletic in a mitochondrial cytochrome b(cytb) gene tree, with a mean divergence of 1.0 ± 0.2%. They were sympatric and mostly altitudinally segregated in the breeding season in southern Sichuan province. We found that the Mt Victoria(western Myanmar) population differed vocally from other L. mandelli, but no specimens are available. Taiwan Bush Warbler L. alishanensis was sister to the L. mandelli complex, with the most divergent song. Plumage, vocal and cytb evidence supported the distinctness of the south Vietnamese L. mandelli idonea. The Timor Bush Warbler L. timorensis, Javan Bush Warbler L.montis and Benguet Bush Warbler L. seebohmi differed distinctly in plumage, but among-population song variation in L. montis exceeded the differences between some populations of these taxa, and mean pairwise cytb divergences were only 0.5–0.9%. We also found that some L. montis populations differed morphologically.Conclusions: We conclude that the central Chinese population of Russet Bush Warbler represents a new species,which we describe herein, breeding at mid elevations in Sichuan, Shaanxi, Hubei, Hunan and Guizhou provinces.The taxonomic status of the other allopatric populations is less clear. However, as they differ to a degree comparable with that of the sympatric L. mandelli and the new species, we elevate L. idonea to species status, and retain L. seebohmi and L. montis as separate species, the latter with timorensis as a subspecies.
