1Exposure to blast shock waves via the ear canal induces deficits in vestibular afferent function in rats显示文摘The ears are air-filled structures that are directly impacted during blast exposure.In addition to hearing loss and tinnitus,blast victims often complain of vertigo,dizziness and unsteady posture,suggesting that blast exposure induces damage to the vestibular end organs in the inner ear.However,the underlying mechanisms remain to be elucidated.In this report,single vestibular afferent activity and the vestibuloocular reflex(VOR)were investigated before and after exposure to blast shock waves(~20 PSI)delivered into the left external ear canals of anesthetized rats.Single vestibular afferent activity was recorded from the superior branch of the left vestibular nerves of the blast-treated and control rats one day after blast exposure.Blast exposure reduced the spontaneous discharge rates of the otolith and canal afferents.Blast exposure also reduced the sensitivity of irregular canal afferents to sinusoidal head rotation at 0.5e2Hz.Blast exposure,however,resulted in few changes in the VOR responses to sinusoidal head rotation and translation.To the best of our knowledge,this is the first study that reports blast exposure-induced damage to vestibular afferents in an animal model.These results provide insights that may be helpful in developing biomarkers for early diagnosis of blast-induced vestibular deficits in military and civilian populations.Yue Yu Jun Huang Xuehui Tang Jerome Allison David Sandlin Dalian Ding Yi Pang Chunming Zhang Tianwen Chen Nathan Yin Lan Chen William Mustain Wu Zhou Hong Zhu 2020Journal of Otology2020,15,3:3

16Kif4A mediates resistance to neoadjuvant chemoradiotherapy in patients with advanced colorectal cancer via regulating DNA damage response显示文摘More and more patients with advanced colorectal cancer(CRC)have benefited from surgical resection or ablation following neoadjuvant chemoradiotherapy(nCRT),but nCRT may be ineffective and have potential risks to some patients.Therefore,it is necessary to discover effective biomarkers for predicting the nCRT efficacy in CRC patients.Chromokinesin Kif4A plays a critical role in mitosis,DNA damage repair and tumorigenesis,but its relationship with nCRT efficacy in advanced CRC remains unclear.Here,we find that Kif4A expression in pretreated tumor tissue is positively correlated with poorer tumor regression after receiving nCRT(P=0.005).Knockdown of endogenous Kif4A causes an increased sensitivity of CRC cells to chemotherapeutic drugs 5-fluorouracil(5-FU)and Cisplatin(DDP),while overexpression of Kif4A enhances resistance of CRC cells to the chemotherapeutic drugs.Furthermore,depending on its motor domain and tail domain,Kif4A regulates DNA damage response(DDR)induced by 5-FU or DDP treatment in CRC cells.In conclusion,we demonstrate that Kif4A may be a potential independent biomarker for predicting the nCRT efficacy in advanced CRC patients,and Kif4A regulates chemosensitivity of CRC cells through controlling DDR.Rui Zhang Shuanghui Liu Bojiang Gong Wenran Xie Youjuan Zhao Liang Xu Yi Zheng Shengnan Jin Chunming Ding Chang Xu Zhixiong Dong 2022Acta Biochimica et Biophysica Sinica2022,54,7:1

20Development of novel-nanobody-based lateral-flow immunochromatographic strip test for rapid detection of recombinant human interferon a2b显示文摘Recombinant human interferon a2b(rhIFNa2b)is widely used as an antiviral therapy agent for the treatment of hepatitis B and hepatitis C.The current identification test for rhIFNa2b is complex.In this study,an anti-rhIFNa2b nanobody was discovered and used for the development of a rapid lateral flow strip for the identification of rhIFNa2b.RhIFNa2b was used to immunize an alpaca,which established a phage nanobody library.After five steps of enrichment,the nanobody I22,which specifically bound rhIFNa2b,was isolated and inserted into the prokaryotic expression vector pET28a.After subsequent purification,the physicochemical properties of the nanobody were determined.A semiquantitative detection and rapid identification assay of rhIFNa2b was developed using this novel nanobody.To develop a rapid test,the nanobody I22 was coupled with a colloidal gold to produce lateral-flow test strips.The developed rhIFNa2b detection assay had a limit of detection of 1 mg/mL.The isolation of I22 and successful construction of a lateral-flow immunochromatographic test strip demonstrated the feasibility of performing ligand-binding assays on a lateral-flow test strip using recombinant protein products.The principle of this novel assay is generally applicable for the rapid testing of other commercial products,with a great potential for routine use in detecting counterfeit recombinant protein products.Xi Qin Maoqin Duan Dening Pei Jian Lin Lan Wang Peng Zhou Wenrong Yao Ying Guo Xiang Li Lei Tao Youxue Ding Lan Liu Yong Zhou Chuncui Jia Chunming Rao Junzhi Wang 2022Journal of Pharmaceutical Analysis2022,12,2:0
