| 1 | A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. | 2 | Invasion and control of water hyacinth (Eichhornia crassipes)in China显示文摘By the time of primary 21st century, water hyacinth had become a serious environmental problem in China. Water hyacinth contributes to the major part of ecological hazards from the invasion of foreign plant species, which is estimated about USD 7 billion a year in values. In the past 10 years, herbicides glyphosate, 2,4-D and paraquat have been used in controlling water hyacinth in China. Al- though the herbicides provided effective control on the weed in some areas, they could not provide the sustainable inhibition on the weed population, while would lead to pollution on water at various levels. At present, the herbicide application on water hyacinth is forbidden in many areas of China such as Shanghai. In this situation, the asexual reproduction inhibitor, KWH02, was invented for controlling water hyacinth and it provided about 70% of growth inhibition without any risk of dead plant pollution. It has been about 10 years for bio-control of water hyacinth in China. Works focused on mainly the efficacy and safety of the utilization of foreign insects. Researches on microorganism herbicides to control water hyacinth were started and obtained primary achievements in recent years. Although there are different opinions on how to face the water hyacinth problem in China, it is accepted widely that the control methods should be high efficient and safe with low cost. | 3 | The Genesis Mechanism of the Mantle Fluid Action and Evolution in the Ore-Forming Process: A Case Study of the Laowangzhai Gold Deposit in Yunnan, China显示文摘Based on petrological studies of the wall rocks, mineralizing rocks, ores and veins from the Laowangzhai gold deposit, it is discovered that along with the development of silication, carbonation and sulfidation, a kind of black opaque ultra-microlite material runs through the spaces between grains, fissures and cleavages. Under observations of the electron microprobe, scanning electronic microscopy and energy spectrum, this kind of ultra-microlite material is confirmed to consist of ultra microcrystalline quartz, silicate, sulfides and carbonates, as well as rutile, scheelite and specularite (magnetite), showing characters of liquation by the analyses of SEM and energy spectrum. The coexistence of immiscibility and precipitating co-crystallization strongly suggests that the mineralizing fluid changed from the melt to the hydrothermal fluid. Combined with the element geochemical researches, it is realized that the ultra-microlite aggregate is the direct relics of the mantle fluid behaving like a melt and supercritical fluid, which goes along with the mantle-derived magma and will escape from the magma body at a proper time. | 4 | Risk factors for progression to acute-on-chronic liver failure during severe acute exacerbation of chronic hepatitis B virus infection显示文摘BACKGROUND Acute exacerbation in patients with chronic hepatitis B virus(HBV) infection results in different severities of liver injury. The risk factors related to progression to hepatic decompensation(HD) and acute-on-chronic liver failure(ACLF) in patients with severe acute exacerbation(SAE) of chronic HBV infection remain unknown.AIM To identify risk factors related to progression to HD and ACLF in compensated patients with SAE of chronic HBV infection.METHODS The baseline characteristics of 164 patients with SAE of chronic HBV infection were retrospectively reviewed. Independent risk factors associated with progression to HD and ACLF were identified. The predictive values of our previously established prediction model in patients with acute exacerbation(AE model) and the model for end-stage liver disease(MELD) score in predicting the development of ACLF were evaluated.RESULTS Among 164 patients with SAE, 83(50.6%) had compensated liver cirrhosis(LC),43 had progression to HD without ACLF, and 29 had progression to ACLF within 28 d after admission. Independent risk factors associated with progression to HD were LC and low alanine aminotransferase. Independent risk factors for progression to ACLF were LC, high MELD score, high aspartate aminotransferase(AST) levels, and low prothrombin activity(PTA). The area under the receiver operating characteristic of the AE model [0.844, 95%confidence interval(CI): 0.779-0.896] was significantly higher than that of MELD score(0.690, 95%CI: 0.613-0.760, P < 0.05) in predicting the development of ACLF.CONCLUSION In patients with SAE of chronic HBV infection, LC is an independent risk factor for progression to both HD and ACLF. High MELD score, high AST, and low PTA are associated with progression to ACLF. | 5 | Reversal of multi-drug resistance by pSUPER-shRNA-mdr1 in vivo and in vitro显示文摘AIM:To explore the possibility of reversing multi-drug resistance(MDR) to HepG2/mdr1 in vitro and in vivo with RNA interference(RNAi) . METHODS:HepG2/mdr1 was obtained by cloning the whole gene mdr1 into HepG2 cells.shRNA targeting sequence was designed to be homologous to the P-gp encoding MDR1 mRNA consensus sequence.pSUPERshRNA/mdr1 was constructed using the enzymedigested technique.HepG2/mdr1 cells were transfected with vectors of pSUPER-shRNA/mdr1 to measure their efficacy by real-time PCR for mdr1 mRNA,flow cytometry(FCM) for P-gp expression,and Rhodamine efflux,MTT method for HepG2/mdr1 function,respectively.In vivo,mice tumors were treated by injecting pSUPER-shRNA/mdr1 in situ and into intraabdominal cavity.Tumors were collected to create cell suspension and cryosections after chemothearpy with adiramycin and mytomycin.The cell suspension was incubated in RPMI-1640 supplemented with G418 to screen stable cells for appreciating the reversal of MDR. Cryosections were treated with immunohistochemistry technique to show the effectiveness of transfection and the expression of P-gp. RESULTS:pSUPER-shRNA/mdr1 was successfullyconstructed,which was confirmed by sequencing. The MDR phenotype of HepG2/mdr1 was decreased significantly in vitro transfection.HepG2/mdr1 showing its MDR was reversed notably in P-gp expression(11.0%vs 98.2%,P<0.01) .Real-time PCR showed that mRNA/mdr1 was lower in test groups than in control groups(18.73±1.33 vs 68.03±2.21,P<0.001) . Compared with HepG2,the sensitivity of HepG2/mdr1 and HepG2/mdr1-dsRNA cells to ADM was decreased by 1.64 times and 15.6 times,respectively. The accumulation of DNR in positive groups was decreased evidently.In vivo,the p-gp expression in positive groups was significantly lower than that in control groups(65.1%vs 94.1%,P<0.05) .The tumor suppressing rate in test groups was 57.8%. After chemotherapy,the growth rate in test groups was lower than that in control groups(700.14±35.61 vs 1659.70±152.54,P<0.05) .Similar results were also observed under fluorescence microscope,and confirmed by Image-Pro Plus 4.5 analysis. | 6 | DDIT4 promotes gastric cancer proliferation and tumorigenesis through the p53 and MAPK pathways显示文摘Background:Gastric cancer(GC)is one of the most common malignancies worldwide,particularly in China.DNA damage-inducible transcript 4(DDIT4)is a mammalian target of rapamycin inhibitor and is induced by various cellular stresses;however,its critical role in GC remains poorly understood.The present study aimed to investigate the poten-tial relationship and the underlying mechanism between DDIT4 and GC development.Methods:We used western blotting,real-time polymerase chain reaction,and immunohistochemical or immunoflu-orescence to determine DDIT4 expression in GC cells and tissues.High-content screening,cell counting kit-8 assays,colony formation,and in vivo tumorigenesis assays were performed to evaluate cell proliferation.Flow cytometry was used to investigate cell apoptosis and cell cycle distribution.Results:DDIT4 was upregulated in GC cells and tissue.Furthermore,downregulating DDIT4 in GC cells inhibited proliferation both in vitro and in vivo and increased 5-fluorouracil-induced apoptosis and cell cycle arrest.In contrast,ectopic expression of DDIT4 in normal gastric epithelial cells promoted proliferation and attenuated chemosensitivity.Further analysis indicated that the mitogen-activated protein kinase and p53 signaling pathways were involved in the suppression of proliferation,and increased chemosensitivity upon DDIT4 downregulation.Conclusion:DDIT4 promotes GC proliferation and tumorigenesis,providing new insights into the role of DDIT4 in the tumorigenesis of human GC. | Feng Du Lina Sun Yi Chu Tingyu Li Chao Lei Xin Wang Mingzuo Jiang Yali Min Yuanyuan Lu Xiaodi Zhao Yongzhan Nie Daiming Fan | 2018 | Cancer Communications2018,38,1: | 10 |
| 7 | Studies on Reducing Nitrogen Excretion: Ⅰ. Net Energy Requirement of Finishing Pigs Maximizing Performance and Carcass Quality Fed Low Crude Protein Diets Supplemented with Crystalline Amino Acids显示文摘Two experiments were conducted to investigate the effects of net energy (NE) level on the performance and carcass traits of finishing pigs fed low crude protein (CP) diets supplemented with crystalline amino acids (CAA). A total of 216 (Exp.1) and 360 (Exp.2) barrows (Yorkshire×Landrace×Duroc) were allotted to one of six treatments (n=6). The experimental diets were based on corn and soybean meal, and consisted of a highCP diet that contained approximately 16% CP and 2.50 Mcal/kg of NE as well as five lowCP diets in which the CP level of the diet was reduced by approximately four percentage units but was supplemented with crystalline lysine, methionine, threonine, and tryptophan. The low CP diets provided 2.64, 2.58, 2.50, 2.42, or 2.36 Mcal/kg in Exp.1 as well as 2.45, 2.40, 2.35, 2.30, or 2.25 Mcal/kg in Exp.2. In Exp.1, a linear (P=0.03) increase in weight gain was observed with decreasing NE level while the ratio of gain to feed was unaffected (P>0.05) by NE level. There was a linear increase (P=0.01) in the percentage of fatfree lean and a linear decrease (P=0.03) in the percentage of total fat with decreasing NE levels. In Exp.2, a significant quadratic (P=0.03) effect of NE level was observed for weight gain. The ratio of gain to feed demonstrated a significant (P<0.01) quadratic effect with pigs fed 2.35 and 2.40 Mcal/kg of NE. Pigs fed the diet containing 2.40 Mcal/kg of NE had the lowest percentage of total fat (36.95%) and the highest percentage of fatfree lean (49.36%). The overall results of these experiments indicate that feeding either a surplus or a deficiency of NE is detrimental to both pig performance and carcass composition when low CP diets supplemented with CAA are fed. | 9 | Effects of Dietary Net Energy Density and Standardized Ileal Digestible Lysine: Net Energy Ratio on the Performance and Carcass Characteristic of Growing-Finishing Pigs Fed Low Crude Protein Supplemented with Crystalline Amino Acids Diets显示文摘Two 3×3 factorial design trials with 1080 of Yorkshire×Landrace×Duroc barrows were conducted to investigate the effectsof dietary net energy (NE) density and standardized ileal digestible (SID) lysine:NE ratio on the performance of growing-finishing pigs and carcass characteristic of finishing pigs fed low crude protein (CP) supplemented with crystalline aminoacids (CAA) diets. Low CP corn-soybean meal basal diets (14% for Exp. 1 and 11% for Exp. 2) supplemented withcrystalline lysine, methionine, threonine, and tryptophan were formulated. Nine treatments with increasing NE densityand SID lysine:NE ratiowereused in each trial(Exp. 1: 9.66, 9.87, and 10.08 MJ kg-1 NE, 0.89, 0.96,and 1.03 g MJ-1 SID lysine:NE; Exp. 2: 9.83, 10.04, and 10.25 MJ kg-1 NE, 0.69, 0.76, and 0.83 g MJ-1 SID lysine:NE). In Exp. 1, we observed that theaverage daily gain (ADG) for barrows that received the diet containing 9.87 MJ kg-1 NE was significantly higher than thatof barrows fed a 9.66 MJ kg-1 NE diet (P<0.05), but no difference was observed when compared with barrows that receiveda diet containing 10.08 MJ kg-1 NE (P>0.05). TheADG was affected both linearly (P<0.05) and quadratically(P<0.05) as theSID lysine:NE ratio increased. The NE density or SID lysine:NE ratio and their interaction had no effect on the averagedaily feed intake (ADFI) and serum urea nitrogen (SUN) (P>0.05). The feed to gain ratio (F:G) decreased (linearly, P<0.05;quadratically, P<0.05) as the NE density or SID lysine:NE ratio increased, but no interaction of NE density and SID lysine:NE ratio on the F:G. In Exp. 2, the dietary NE density both linearly (P<0.05) and quadratically (P<0.05) affected the ADGand F:G. Barrows that received a diet containing 10.04 MJ kg-1 NE had a greater ADG (P<0.05) than barrows that receiveda 9.83 MJ kg-1 diet, but there was no difference (P>0.05) as compared with barrows that received a diet containing 10.25 MJkg-1 NE. F:G was significantly higher (P<0.05) for barrows that received a 9.83 MJ kg-1 diet when compared with barrowsthat received the other two diets. The ADG was significantly decreased (P<0.05) for pigs that received a diet containing0.69 g MJ-1 SID lysine:NE ratio when compared with pigs fed a diet containing 0.76 or 0.83 g MJ-1 SID lysine:NE ratio. TheNE density or SID lysine:NE ratio and their interaction had no effect on the average daily feed intake, SUN, and carcasscharacteristic for finishing barrows (P>0.05). | 10 | Repeatability and Reproducibility of Quantitative Assessment of the Retinal Microvasculature Using Optical Coherence Tomography Angiography Based on Optical Microangiography显示文摘Objective The aim of this study was to determine the repeatability and reproducibility of optical coherence tomography angiography(OCTA) based on optical microangiography(OMAG) measurements of macular vessels in normal eyes. Methods In this prospective cohort study, 40 eyes of 40 healthy volunteers underwent repeated OCTA(Cirrus HD-OCT 5000 angiography system, Carl Zeiss Meditec, Inc.) scans on two separate visit days. On each visit day, the eyes were scanned three times. The following parameters were used to quantitatively describe the OCTA images of the superficial vascular network: vessel area density(VAD), vessel skeleton density(VSD), vessel diameter index(VDI), vessel perimeter index(VPI), vessel complexity index(VCI), flux, and foveal avascular zone(FAZ). Coefficient of variation(CV) and intraclass correlation coefficient(ICC) were calculated for evaluating intravisit and intervisit repeatability, as well as interobserver reproducibility. Results The measurements showed high repeatability [CVs ≤ 4.2%(intravisit) and ≤ 4.6%(intervisit)] and interobserver reproducibility(ICCs ≥ 0.923) for all parameters. | 16 | ~1H nuclear magnetic resonance-based metabolomic study on efficacy of Qingrehuatan decoction against abundant phlegm-heat syndrome in young adults with essential hypertension显示文摘OBJECTIVE: To observe the influence of Qingrehuatan decoction(QRHT) on serum metabolic profile in young essential hypertension(YEH) patients with abundant phlegm-heat syndrome and provide a basis for treatment with the decoction.METHODS: Twelve male YEH patients were randomly selected and serum samples were collected for examination before and after 4 weeks of thetreatment with QRHT. Twelve healthy males were randomly selected and their serum samples were collected as a control. All serum samples were detected using metabolomic technology with1 H nuclear magnetic resonance. Differences in metabolites were studied by principal component analysis and partial least squares-discriminate analysis,which produced scores and loadings plots.RESULTS: After 4 weeks of treatment, serum substances could be distinguished between the YEH patients with abundant phlegm-heat syndrome and the control patients. The specific serum endogenous metabolites tended to improve after the treatment. QRHT can appropriately increase the levels of glucose, lactic acid, citric acid, high-density lipoprotein, phosphatidylcholine, glycerophosphate choline, hydroxybutyrate, alanine, and glutamate.QRHT could also decrease the levels of low-density lipoprotein/very low-density lipoprotein, lipids,N-acetyl glycoprotein, and O-acetyl glycoprotein.CONCLUSION: QRHT can effectively ameliorate metabolic disorders in YEH Patients with abundant phlegm- heat syndrome.1H NMR-based metabolomic technology can provide an objective basis for the treatment of YEH patients with abundant phlegm-heat syndrome using QRHT. | Feng Xuanchao Yang Zheng Chu Yuguang Du Bai Su Mei Li Yi Wang Yinghong Jiang Chunying Hu Yuanhui | 2015 | Journal of Traditional Chinese Medicine2015,35,1: | 6 |

| 19 | Genetic and Antibiotic Resistance Characteristics ofCampylobacterjejuni Isolated from Diarrheal Patients,Poultry and Cattle in Shenzhen显示文摘Objective To investigate genetic and antibiotic resistance characteristics of Campylobacter jejuni(C. jejuni) isolated from Shenzhen. Methods Multilocs sequence typing and agar dilution methods were used to define the genotype and antibiotic resistance of C. jejuni, respectively. Results In total, 126 C. jejuni strains were isolated. The prevalence of C. jejuni was 5.3% in diarrheal patients. The prevalence in poultry meat(36.5%) was higher than that in cattle meat(1.1%). However, the prevalence in poultry cloacal swabs(27.0%) was lower than that in cattle stool(57.3%). Sixty-two sequence types were obtained, among which 27 of the STs and 10 alleles were previously unreported. The most frequently observed clonal complexes were ST-21(11.9%), ST-22(10.3%), and ST-403(7.1%). ST-21, ST-45, ST-354, ST-403, and ST-443 complexes overlapped between isolates from patients and cattle, whereas ST-45 and ST-574 complexes overlapped between isolates from patients and poultry. All C. jejuni were resistant to at least one antibiotic. The highest resistance rate was toward ciprofloxacin(89.7%), followed by tetracycline(74.6%), and nalidixic acid(69.0%). Conclusion This is the first report of the genotypes and antibiotic resistance of C. jejuni in Shenzhen. | 20 | Exogenous hydrogen sulfide inhibits apoptosis by regulating endoplasmic reticulum stress-autophagy axis and improves myocardial reconstruction after acute myocardial infarction显示文摘During acute myocardial infarction,endoplasm ic reticulum(ER)stress-induced autophagy and apoptosis have been shown as im portant pathogeneses of myocardial reconstruction.Im portantly,hydrogen sulfide(H2S),as a third endogenous gas signaling molecule,exerts strong cytoprotective effect on anti-ER stress,autophagy regulation and antiapoptosis.Here,we showed that H2 S treatm ent inhibits apoptosis by regulating ER stress-autophagy axis and im proves myocardial reconstruction after acute myocardial infarction.We found that H2S intervention im proved left ventricle function,reduced glycogen deposition in myocardial tissue mesenchyme,and inhibited apoptosis.Moreover,the expressions of fibrosis indicators(Col3a1 and Col1a2),ER stress-related proteins(CHOP and BIP/ERP78),autophagy-related proteins(Beclin and ATG5),apoptosis protein(Bax),as well as fibrosis protein Col4a3bp were all decreased after treatm ent w ith H2 S.H2 S adm inistration also m aintained MMP/TIMP balance.Mechanistically,H2S activated the PI3K/AKT signaling pathway.In addition,H2S treatm ent also reduced the expressions of ER stress-related proteins,autophagy-related proteins,and apoptins in in wtro experiments.Interestingly,activation of ER stress-autophagy axis could reverse the inhibitory effect of H2S on myocardial apoptosis.Altogether,these results suggested that exogenous H2S suppresses myocardial apoptosis by blocking ER stress-autophagy axis,which in turn reverses cardiac rem odeling after myocardial infarction. | Yaling Li Maojun Liu Jiali Yi Xiong Song Xia Zheng Da Liu Sen Wang Chun Chu Jun Yang | 2020 | Acta Biochimica et Biophysica Sinica2020,52,12: | 5 |
