| 4 | Bioinspired urease-powered micromotor as an active oral drug delivery carrier in stomach显示文摘Self-propelling micro-and nano-motors(MNMs)have been extensively investigated as an emerging oral drug delivery carrier for gastrointestinal(GI)tract diseases.However,the propulsion of current MNMs reported so far is mostly based on the redox reaction of metals(such as Zn and Mg)with severe propulsion gas generation,remaining non-degradable residue in the GI tract.Here,we develop a bioinspired enzyme-powered biopolymer micromotor mimicking the mucin penetrating behavior of Helicobacter pylori in the stomach.It converts urea to ammonia and the subsequent increase of pH induces local gel-sol transition of the mucin layer facilitating the penetration into the stomach tissue layer.The successful fabrication of micromotors is confirmed by high-resolution transmission electron microscopy,electron energy loss spectroscopy,dynamic light scattering analysis,zeta-potential analysis.In acidic condition,the immobilized urease can efficiently converted urea to ammonia,comparable with that of neutral condition because of the increase of surrounding pH during propulsion.After administration into the stomach,the micromotors show enhanced penetration and prolonged retention in the stomach for 24 h.Furthermore,histological analysis shows that the micromotors are cleared within 3 days without causing any toxicity in the GI tract.The enhanced penetration and retention of the micromotors as an active oral delivery carrier in the stomach would be successfully harnessed for the treatment of various GI tract diseases. | Hyunsik Choi Sang Hoon Jeong Tae Yeon Kim Jeeyoon Yi Sei Kwang Hahn | 2022 | Bioactive Materials2022,7,3: | 3 |

| 6 | Immune enhancement effect of an herb complex extract through the activation of natural killer cells and the regulation of cytokine levels in a cyclophosphamide-induced immunosuppression rat model显示文摘Objective: To investigate the effects of a herb complex extract(HCE) prepared from Cornus officinalis Sieb. Et Zucc., Eriobotrya japonica Lindley, and olive leaves on immune response of mouse spleen NK cells in vitro and in vivo analysis. Methods: The activity of natural killer(NK) cells was measured in splenocytes and YAC-1 cells. Mice were immunosuppressed using cyclophosphamide(5 mg/kg body weight). Three different doses of HCE(200, 400, and 800 mg/kg body weight) and red ginseng extract(800 mg/kg body weight) which was used as standard immunomodulatory herb were administered orally for 4 weeks. The body weight, dietary, water intake, organs(liver, thymus, and spleen) weight, completed blood count, and cytokines(tumor necrosis factor alpha, interferon gamma, and interleukin-2) production was measured. Results: At the maximum concentration of HCE, the activity of NK cells was increased by 48.5%. HCE increased liver, spleen, and thymus weights without altering numbers of white blood cells, lymphocytes, and neutrophils in a cyclophosphamide-induced immunosuppression rat model. However, HCE recovered the inhibited cytokine expression; HCE(800 mg/kg) increased cytokines levels. The results indicate the immune enhancement potential of this HCE. Conclusion: The HCE enhances immunity by increasing NK cell activity, regulating cytokine levels, and maintaining spleen weight. | 7 | Laser micromachining of CNT/Fe/Al_2O_3 nanocomposites显示文摘CNT/Fe/Al2O3 mixed powders were synthesized from Fe/Al2O3 nanopowders using thermal CVD for the homogeneous dispersion of carbon nanotubes CNTs. CNTs consisted of MWNT, and the diameter was approximately 20-30 nm. After sintering, CNTs were homogenously located throughout Al2O3 grain boundary and were buckled. A femto-second laser installed with special optical systems was used for micromachining of the nanocomposites. The relationship between material ablation rate and energy fluence was theoretically investigated and compared with experimental results from cross-sectional SEM analysis. | 8 | Whole-genome,transcriptome,and methylome analyses provide insights into the evolution of platycoside biosynthesis in Platycodon grandiflorus,a medicinal plant显示文摘Triterpenoid saponins(TSs)are common plant defense phytochemicals with potential pharmaceutical properties.Platycodon grandiflorus(Campanulaceae)has been traditionally used to treat bronchitis and asthma in East Asia.The oleanane-type TSs,platycosides,are a major component of the P.grandiflorus root extract.Recent studies show that platycosides exhibit anti-inflammatory,antiobesity,anticancer,antiviral,and antiallergy properties.However,the evolutionary history of platycoside biosynthesis genes remains unknown.In this study,we sequenced the genome of P.grandiflorus and investigated the genes involved in platycoside biosynthesis.The draft genome of P.grandiflorus is 680.1 Mb long and contains 40,017 protein-coding genes.Genomic analysis revealed that the CYP716 family genes play a major role in platycoside oxidation.The CYP716 gene family of P.grandiflorus was much larger than that of other Asterid species.Orthologous gene annotation also revealed the expansion ofβ-amyrin synthases(bASs)in P.grandiflorus,which was confirmed by tissue-specific gene expression.In these expanded gene families,we identified key genes showing preferential expression in roots and association with platycoside biosynthesis.In addition,wholegenome bisulfite sequencing showed that CYP716 and bAS genes are hypomethylated in P.grandiflorus,suggesting that epigenetic modification of these two gene families affects platycoside biosynthesis.Thus whole-genome,transcriptome,and methylome data of P.grandiflorus provide novel insights into the regulation of platycoside biosynthesis by CYP716 and bAS gene families. | Jungeun Kim Sang-Ho Kang Sin-Gi Park Tae-Jin Yang Yi Lee Ok Tae Kim Oksung Chung Jungho Lee Jae-Pil Choi Soo-Jin Kwon Keunpyo Lee Byoung-Ohg Ahn Dong Jin Lee Seung-il Yoo In-Gang Shin Yurry Um Dae Young Lee Geum-Soog Kim Chang Pyo Hong Jong Bhak Chang-Kug Kim | 2020 | Horticulture Research2020,7,1: | 2 |

| 11 | Hypothyroidism affects astrocyte and microglial morphology in type 2 diabetes显示文摘In the present study,we investigated the effects of hypothyroidism on the morphology of astrocytes and microglia in the hippocampus of Zucker diabetic fatty rats and Zucker lean control rats.To induce hypothyroidism,Zucker lean control and Zucker diabetic fatty rats at 7 weeks of age orally received the vehicle or methimazole,an anti-thyroid drug,treatment for 5 weeks and were sacrificed at 12 weeks of age in all groups for blood chemistry and immunohistochemical staining.In the methimazole-treated Zucker lean control and Zucker diabetic fatty rats,the serum circulating triiodothyronine(T3)and thyroxine(T4)levels were significantly decreased compared to levels observed in the vehicle-treated Zucker lean control or Zucker diabetic fatty rats.This reduction was more prominent in the methimazole-treated Zucker diabetic fatty group.Glial fibrillary acidic protein immunoreactive astrocytes and ionized calcium-binding adapter molecule 1(Iba-1)-immunoreactive microglia in the Zucker lean control and Zucker diabetic fatty group were diffusely detected in the hippocampal CA1 region and dentate gyrus.There were no significant differences in the glial fibrillary acidic protein and Iba-1 immunoreactivity in the CA1 region and dentate gyrus between Zucker lean control and Zucker diabetic fatty groups.However,in the methimazole-treated Zucker lean control and Zucker diabetic fatty groups,the processes of glial fibrillary acidic protein immunoreactive astrocytes and Iba-1 immunoreactive microglia,were significantly decreased in both the CA1region and dentate gyrus compared to that in the vehicle-treated Zucker lean control and Zucker diabetic fatty groups.These results suggest that diabetes has no effect on the morphology of astrocytes and microglia and that hypothyroidism during the onset of diabetes prominently reduces the processes of astrocytes and microglia. | Sung Min Nam Yo Na Kim Dae Young Yoo Sun Shin Yi Jung Hoon Choi In Koo Hwang Je Kyung Seong Yeo Sung Yoon | 2013 | Neural Regeneration Research2013,8,26: | 2 |
