1SDF-1/CXCR4 axis modulates bone marrow mesenchymal stem cell apoptosis, migration and cytokine secretion显示文摘Bone marrow mesenchymal stem cells(MSCs)are considered as a promising cell source to treat the acute myocardial infarction.However,over 90%of the stem cells usually die in the first three days of transplantation.Survival potential,migration ability and paracrine capacity have been considered as the most important three factors for cell transplantation in the ischemic cardiac treatment.We hypothesized that stromal-derived factor-1(SDF-1)/CXCR4 axis plays a critical role in the regulation of these processes.In this study,apoptosis was induced by exposure of MSCs to H2O2 for 2 h.After re-oxygenation,the SDF-1 pretreated MSCs demonstrated a significant increase in survival and proliferation.SDF-1 pretreatment also enhanced the migration and increased the secretion of pro-survival and angiogenic cytokines including basic fibroblast growth factor and vascular endothelial growth factor.Western blot and RT-PCR demonstrated that SDF-1 pretreatment significantly activated the pro-survival Akt and Erk signaling pathways and up-regulated Bcl-2/Bax ratio.These protective effects were partially inhibited by AMD3100,an antagonist of CXCR4.We conclude that the SDF-1/CXCR4 axis is critical for MSC survival,migration and cytokine secretion.Xiaolei Liu Biyan Duan Zhaokang Cheng Xiaohua Jia Lina Mao Hao Fu Yongzhe Che Lailiang Ou Lin Liu Deling Kong 2011Protein & Cell2011,2,10:53
2Sugar metabolism and accumulation in the fruit of transgenic apple trees with decreased sorbitol synthesis显示文摘Both sorbitol and sucrose are synthesized in source leaves and transported to fruit for supporting fruit growth in tree fruit species of the Rosaceae family.In apple(Malus domestica),antisense suppression of aldose-6-phosphate reductase,the key enzyme for sorbitol synthesis,significantly decreased the sorbitol concentration but increased the sucrose concentration in leaves,leading to a lower sorbitol but a higher sucrose supply to fruit in these plants.In response to this altered carbon supply,the transgenic fruit had lower concentration of sorbitol and much higher concentration of glucose but similar levels of fructose,sucrose,and starch throughout fruit development relative to the untransformed control.Activities of sorbitol dehydrogenase,fructokinase,and sucrose phosphate synthase were lower,whereas activities of neutral invertase,sucrose synthase,and hexokinase were higher in the transgenic fruit during fruit development.Transcript levels of MdSOT1,MdSDHs,MdFK2,and MdSPS3/6 were downregulated,whereas transcript levels of MdSUC1/4,MdSUSY1-3,MdNIV1/3,MdHKs,and MdTMT1 were upregulated in the transgenic fruit.These findings suggest that the Sucrose cycle and the sugar transport system are very effective in maintaining the level of fructose and provide insights into the roles of sorbitol and sucrose in regulating sugar metabolism and accumulation in sorbitol-synthesizing species.Mingjun Li Pengmin Li Fengwang Ma Abhaya M.Dandekar Lailiang Cheng 2018Horticulture Research2018,5,1:12
3Increased activity of MdFRK2, a high-affinity fructokinase, leads to upregulation of sorbitol metabolism and downregulation of sucrose metabolism in apple leaves显示文摘To investigate the functions of fructokinase(FRK)in apple(Malus domestica)carbohydrate metabolism,we cloned the coding sequences of MdFRK1 and MdFRK2 from the‘Royal Gala’apple.The results showed that MdFRK2 expression was extremely high in shoot tips and young fruit.Analyses of heterologously expressed proteins revealed that MdFRK2 had a higher affinity for fructose than did MdFRK1,with Km values of 0.1 and 0.62 mM for MdFRK2 and MdFRK1,respectively.The two proteins,however,exhibited similar Vmax values when their activities were significantly inhibited by high concentrations of fructose.MdFRK2 ectopic expression was associated with a general decrease in fructose concentration in transgenic lines.In leaves,increased FRK activity similarly resulted in reduced concentrations of glucose and sucrose but no alterations in sorbitol concentration.When compared with those in the untransformed control,genes involved in sorbitol synthesis(A6PR)and the degradation pathway(SDH1/2)were significantly upregulated in transgenic lines,whereas those involved in sucrose synthesis(SPS1)and other degradation processes(SUSY4,NINV1/2,and HxK2)were downregulated.The activity of enzymes participating in carbohydrate metabolism was proportional to the level of gene expression.However,the growth performance and photosynthetic efficiency did not differ between the transgenic and wild-type plants.These results provide new genetic evidence to support the view that FRK plays roles in regulating sugar and sorbitol metabolism in Rosaceae plants.Jingjing Yang Lingcheng Zhu Weifang Cui Chen Zhang Dongxia Li Baiquan Ma Lailiang Cheng Yong-Ling Ruan Fengwang Ma Mingjun Li 2018Horticulture Research2018,5,1:9
4Effects of location within the tree canopy on carbohydrates,organic acids,amino acids and phenolic compounds in the fruit peel and flesh from three apple(Malus×domestica)cultivars显示文摘Fruits from three cultivars of apple(Malus × domestica Borkh.)—‘McIntosh’,‘Gala’and‘Mutsu’—were harvested from the exterior and interior of the tree canopy.Peel and flesh tissues were sampled separately to determine how the position of the fruit on the tree might affect the levels of the primary and secondary metabolites in the fruit.Fruit from the outer-canopy had a higher fresh weight and a higher soluble solids content compared with inner-canopy fruit.Both the flesh and peel of the outer-canopy fruit had higher concentrations of soluble sugars and sugar alcohols,but lower starch concentrations than the inner-canopy fruit.Canopy position did not significantly affect malic acid concentrations,except in the peel of‘McIntosh’and the flesh of‘Mutsu’.Although levels of ascorbic and succinic acids were higher in the peel of the outer-canopy fruit,the responses of other organic acids to canopy position depended on tissue type and cultivar.Except for histidine,lysine,threonine and glycine,most amino acids accumulated at higher concentrations in the inner-canopy fruit.By contrast,levels of phenolic compounds from both the peel and flesh were significantly higher in the outer-canopy fruit.The significant effects of location within the canopy on both primary metabolites and secondary metabolites demonstrate the importance of light exposure on apple fruit quality.Fengjuan Feng Mingjun Li Fengwang Ma Lailiang Cheng 2014Horticulture Research2014,1,1:7
5High performance of a unique mesoporous polystyrene-based adsorbent for blood purification显示文摘A multi-functional polystyrene based adsorbent(NKU-9)with a unique mesoporous and a high surface area was prepared by suspension polymerization for removal of therapeutic toxins in blood purification.The adsorbent produced had an almost equal amount of mesopore distribution in the range from 2 to 50 nm.The adsorption of serum toxins with different molecular weights were examined by in vitro adsorption assays and compared with some clinical currently used adsorbents such as HA-330,Cytosorb and BL-300 which are produced by China,America and Japan,respectively.Test results indicated that the adsorption rate for pentobarbital by NKU-9 was 81.24%which is nearly as high as HA-330(81.44%).The latter adsorbent is currently used for acute detoxification treatment in China.To reach adsorption equilibrium,NKU-9 was faster than HA-330,which implies short treatment time.For the removal of middle molecular toxins such as b2-microglobulin(98.88%),NKU-9 performed better adsorptive selectivity than Cytosorb(92.80%).In addition,NKU-9 showed high performance for the removal of albumin-bound toxins(e.g.,bilirubin),and its adsorption rate for total bilirubin(80.79%)in plasma was 8.4%higher than that of anion exchange resin BL-300 which is currently used to eliminate bilirubin in clinic.Therefore,our results indicate that the newly developed adsorbent with a wide distribution and almost equal amount of mesopores is a multifunctional adsorbent for high efficient removal of serum toxins with different molecular weights which might be an excellent blood purification adsorbent especially to treat diseases that conventional medical methods are low or not efficient.Jian Chen Wenyan Han Jie Chen Wenhui Zong Weichao Wang Yue Wang Guanghui Cheng Chunran Li Lailiang Ou Yaoting Yu 2017Regenerative Biomaterials2017,4,1:3
6Effect of Metaxenia on Sugars and Organic Acids and Taste Quality of Bagged Fuji Apple Fruits显示文摘In order to provide reference for the selection of appropriate pollination varieties facilitating the improvement of the flavor quality of bagged Fuji apple,the metaxenia effects of 5 pollination varieties on sugars,organic acids and flavor quality of bagged Fuji fruit were studied.The results showed that the 5 pollination varieties had significant metaxenia effects on sugars,organic acids and flavor quality of bagged Fuji fruit.For the sugar components,the fruits pollinated with Golden Delicious and Jinxiuhaitang showed higher glucose contents,and the fruit pollinated with Gala had the highest sucrose content;the fruits pollinated by Gala,Golden Delicious and Ruby exhibited higher malic acids,and the oxalic acid content and the succinic acid content were higher in the fruits pollinated with Jinxiuhaitang and Ruby;and the fruit pollinated with Gala had the highest total sugar content,and the total acid content was higher in the fruits pollinated by Jinxiuhaitang and Ruby.The sugar-acid ratio was higher in the fruits pollinated with Gala and Golden Delicious,and the lowest in the fruit pollinated with Ruby.The sensory evaluation of fruits showed that the fruit pollinated with Ruby tasted sour-sweet,while the fruits pollinated with other 4 pollination varieties were sour and sweet moderately.Haibo WANG Chuanzeng WANG Lailiang CHENG Yuansheng CHANG Ping HE Linguang LI 2017Agricultural Biotechnology2017,6,4:2

10Wave-dominated,mesotidal headland-bay beach morphodynamic classsfications of the Shuidong Bay in South China显示文摘Beach morphodynamic classifications have achieved extensive acceptance in foreign coastal geomorphological studies.Three beaches located in different zones of a headland-bay coast are classified according to a dimensionless fall parameter,a relative tide range parameter and a dimensionless embayment scaling parameter.Synchronous data,including wave,tide,sediment and beach morphology,are respectively collected from the tangential beach,the transitional beach and the shadow beach of the Shuidong Bay during each spring tide for 16 successive months.The research results indicate that(1) the beach in the tangential zone falls between two major categories which are low tide terrace beaches with rips and barred beaches;the beach in the transitional zone exhibits two main types which are low tide bar/rip beaches and barred dissipative beaches;and the beach in the shadow zone mainly mirrors dissipative states with presence or absence of bars;and(2) the sequential changes and differences of beach states in different coastal zones reflect spatial and temporal variabilities of the headlandbay coast,totally meeting the actual measured beach morphology changes,showing that studies on wavedominated,meso-macrotidal beaches need to consider the influences of the tides.Meanwhile,the research mainly provides a framework about beach state studies,due to different beach states with different erosion patterns,which requires the need to strengthen the researches in this respect,in order to further enrich theoretical basis for a beach topography evolution,beach morphodynamic processes and beach erosion prevention in China.YU Jitao DING Yuanting CHENG Huangxin CAI Lailiang CHEN Zishen 2016Acta Oceanologica Sinica2016,35,7:1
