| 1 | Reduction of tumorigenicity of SMMC-7721 hepatoma cells by vascular endothelial growth factor antisense gene therapy显示文摘AIM To test the hypothesis to block VEGFexpression of SMMC-7721 hepatoma cells mayinhibit tumor growth using the rat hepatomamodel.METHODS Amplifiy the 200 VEGF cDNAfragment and insert it into human U6 genecassette in the reverse orientation transcribingsmall antisense RNA which could specificallyinteract with VEGF165, and VEGF121 mRNA.Construct the retroviral vector containing thisantisense VEGF U6 cassette and package thereplication-deficient recombinant retrovirus.SMMC-7721 cells were transduced with thesevirus and positive clones were selected withG418. PCR and Southern blot analysis wereperformed to determine if U6 cassette integratedinto the genomic DNA of positive clone.Transfected tumor cells were evaluated for RNAexpression by ribonuclease protection assays.The VEGF protein in the supernatant of parentaltumor cells and genetically modified tumor cellswas determined with ELISA. In vitro and in vivogrowth properties of antisense VEGF cell clonein nude mice were analyzed.RESULTS Restriction enzyme digestion andPCR sequencing verified that the antisense VEGFRNA retroviral vector was successfullyconstructed. After G418 selection, resistantSMMC-7721 cell clone was picked up. PCR andSouthern blot analysis suggested that U6cassette was integrated into the cell genomicDNA. Stable SMMC-7721 cell clone transducedwith U6 antisense RNA cassette could express200bp small antisense VEGF RNA and secretereduced levels of VEGF in culture condition.Production of VEGF by antisense transgeneexpressing cells was 65 ± 10 ng / L per 106 cells,420 ± 45 ng/L per 106 cells in sense group and 485± 30 ng/L per 106 cells in the negative control group, (P<0.05). The antisense-VEGF cell clone appeared phenotypically indistinguishable from SMMC-7721 cells and SMMC-7721 cells transfected sense VEGF. The growth rate of the antisense-VEGF cell clone was the same as the control cells. When S. | 15 | A new dynamic pushback control method for reducing fuel-burn costs: Using predicted taxi-out time显示文摘Long departure-taxi-out time leads to significant airport surface congestion, fuel-burn costs, and excessive emissions of greenhouse gases. To reduce these undesirable effects, a Predicted taxi-out time-based Dynamic Pushback Control(PDPC) method is proposed. The implementation of this method requires two steps: first, the taxi-out times for aircraft are predicted by the leastsquares support-vector regression approach of which the parameters are optimized by an introduced improved Firefly algorithm. Then, a dynamic pushback control model equipped with a linear gate-hold penalty function is built, along with a proposed iterative taxiway queue-threshold optimization algorithm for solving the model. A case study with data obtained from Beijing International airport(PEK) is presented. The taxi-out time prediction model achieves predictive accuracy within 3 min and 5 min by 84.71% and 95.66%, respectively. The results of the proposed pushback method show that total operation cost and fuel-burn cost achieve a 14.0% and 21.1%reduction, respectively, as compared to the traditional K-control policy.(3) From the perspective of implementation, using PDPC policy can significantly reduce the queue length in taxiway and taxi-out time. The total operation cost and fuel-burn cost can be curtailed by 37.2% and 52.1%,respectively, as compared to the non-enforcement of any pushback control mechanism. | 16 | Three new alkaloids isolated from the stem tuber of Pinellia pedatisecta显示文摘The present study was designed to determine the chemical constituents of the stem tuber of Pinellia pedatisecta. The chemical constituents were isolated and purified by various chromatographic techniques, and their structures were elucidated on the basis of physicochemical properties and spectral data. Three new alkaloids(compounds 1, 2, and 3) were obtained and identified as 9-((5-methoxypyridin-2-yl)methyl)-9 H-purin-6-amine(1), 4-(2-(2, 5-dioxopyrrolidin-1-yl)ethyl)phenyl acetate(2), and N-(9-((5-methoxypyridin-2-yl)methyl)-9 H-purin-6-yl)acetamide(3). These compounds were evaluated for their cytotoxicity against human cervical cancer He La cells. | 17 | Prospects and Problems for Identification of Poisonous Plants in China using DNA Barcodes显示文摘Objective Poisonous plants are a deadly threat to public health in China. The traditional clinical diagnosis of the toxic plants is inefficient, fallible, and dependent upon experts. In this study, we tested the performance of DNA barcodes for identification of the most threatening poisonous plants in China. Methods Seventy-four accessions of 27 toxic plant species in 22 genera and 17 families were sampled and three DNA barcodes(matK, rbcL, and ITS) were amplified, sequenced and tested. Three methods, Blast, pairwise global alignment(PWG) distance, and Tree-Building were tested for discrimination power. Results The primer universality of all the three markers was high. Except in the case of ITS for Hemerocallis minor, the three barcodes were successfully generated from all the selected species. Among the three methods applied, Blast showed the lowest discrimination rate, whereas PWG Distance and Tree-Building methods were equally effective. The ITS barcode showed highest discrimination rates using the PWG Distance and Tree-Building methods. When the barcodes were combined, discrimination rates were increased for the Blast method. Conclusion DNA barcoding technique provides us a fast tool for clinical identification of poisonous plants in China. Based on the labeling schema by Lee, spatial curves both for facets and nodes are proposed and the main algorithms for coordinate translation, node Lsequence generation and visiting nodes are presented. In particular,constant time algorithms for generating node Lsequence are advanced by using bit manipulation operations,which can be easily implemented with hardware. In L curve the distance between three nodes of a facet is mostly limited in a range of small value, thus making fast access possible. Though codes of sibling facets are continuous, the difference between codes of some cousins may occasionally be very large and makes the distance of a few facets also very large, thus greatly increasing the mean node distance and the total traversing distance. | 20 | Effects of 1-methylcyclopropene and modified atmosphere packaging on fruit quality and superficial scald in Yali pears during storage显示文摘The Yali pear(Pyrus bretschneideri Rehd.) is susceptible to superficial scald during prolonged cold storage and at shelf life. This study investigated the effects of 1-methylcyclopropene(1-MCP) and modified atmosphere packaging(MAP) on changes of fruit quality and superficial scald during cold storage and at shelf life in Yali pear. Compared with MAP, the combination of MAP and 1-MCP(MAP+1-MCP) treatment reduced the carbon dioxide and ethylene content inside the packaging bag. The 1-MCP, MAP, and MAP+1-MCP treatments reduced the superficial scald index, malondialdehyde content, O_2^-· production rate and relative conductivity and inhibited the accumulation of α-farnesene and conjugated trienes in the peel. 1-MCP and MAP+1-MCP treatments maintained a higher phenolic content and enhanced the catalase and superoxide dismutase activities in the fruit, while reduced activities of lipoxygenase and polyphenol oxidase in the peel preceding the onset of superficial scald.
