1Modelling of low-temperature exhumation rate in Dabie Mountain based on (U-Th)/He and fission-track thermochronological data显示文摘While the high-temperature exhumation process in the Dabie Mountain has been well documented, the low-temperature exhumation of this area since Cretaceous, especially since Late Cretaceous, is relatively less studied. Low-temperature thermochronology provides one of the important approaches to solve this problem. Based on the data of fission track and (U-Th)/He analysis of apaptites and zircons from the granitoid and metamorphic rocks in the Dabie Mountain, this paper applies Mancktelow’s and Braun’s methods to estimating the exhumation rates and to drawing the regional differential exhumation pattern since Cretaceous, especially since Late Cretaceous by taking into consideration factors such as heat transport, heat advection, topography and heat production, which could influence geothermal field in the shallow crust. Since Cretaceous, the exhumation rate (0.08-0.10 km/Ma) in the region around Tiantangzhai and in the south of Tanlu fault zone is larger than the rate (0.04-0.07 km/Ma) in other areas of the Dabie Mountain. 2Preliminary experimental results by the prototype of Sanya Incoherent Scatter Radar显示文摘In the past decades,the Incoherent Scatter Radar(ISR)has been demonstrated to be one of the most powerful instruments for ionosphere monitoring.The Institute of Geology and Geophysics at the Chinese Academy of Sciences was founded to build a state-ofthe-art phased-array ISR at Sanya(18.3°N,109.6°E),a low-latitude station on Hainan Island,named the Sanya ISR(SYISR).As a first step,a prototype radar system consisting of eight subarrays(SYISR-8)was built to reduce the technical risk of producing the entire large array.In this work,we have summarized the preliminary experimental results based on the SYISR-8.The amplitude and phase among 256 channels were first calibrated through an embedded internal monitoring network.The mean oscillation of the amplitude and phase after calibration were about 1 dB and 5°,respectively,which met the basic requirements.The beam directivity was confirmed by crossing screen of the International Space Station.The SYISR-8 was further used to detect the tropospheric wind profile and meteors.The derived winds were evaluated by comparison with independent radiosonde and balloon-based GPS measurements.The SYISR-8 was able to observe several typical meteor echoes,such as the meteor head echo,range-spread trail echo,and specular trail echo.These results confirmed the validity and reliability of the SYISR-8 system,thereby reducing the technical risk of producing the entire large array of the SYISR to some extent.XinAn Yue WeiXing Wan Han Xiao LingQi Zeng ChangHai Ke BaiQi Ning Feng Ding BiQiang Zhao Lin Jin Chen Li MingYuan Li JunYi Wang HongLian Hao Ning Zhang 2020Earth and Planetary Physics2020,4,6:3
3Enhanced osteoarthritis therapy by nanoengineered mesenchymal stem cells using biomimetic CuS nanoparticles loaded with plasmid DNA encoding TGF-β1显示文摘Mesenchymal stem cells(MSCs)therapy shows the potential benefits to relieve clinical symptoms of osteoarthritis(OA),but it is uncertain if it can repair articular cartilage lesions-the main pathology of OA.Here,we prepared biomimetic cupper sulfide@phosphatidylcholine(CuS@PC)nanoparticles(NPs)loaded with plasmid DNA(pDNA)encoding transforming growth factor-beta 1(TGF-β1)to engineer MSCs for enhanced OA therapy via cartilage regeneration.We found that the NPs not only promoted cell proliferation and migration,but also presented a higher pDNA transfection efficiency relative to commercial transfection reagent lipofectamine 3000.The resultant CuS/TGF-β1@PC NP-engineered MSCs(termed CTP-MSCs)were better than pure MSCs in terms of chondrogenic gene expression,glycosaminoglycan deposition and type II collagen formation,favoring cartilage repair.Further,CTP-MSCs inhibited extracellular matrix degradation in interleukin-1β-induced chondrocytes.Consequently,intraarticular administration of CTP-MSCs significantly enhanced the repair of damaged cartilage,whereas pure MSCs exhibited very limited effects on cartilage regeneration in destabilization of the medial meniscus(DMM)surgical instability mice.Hence,this work provides a new strategy to overcome the limitation of current stem cell therapy in OA treatment through developing more effective nanoengineered MSCs.Yu Cai Cuixi Wu Qianhua Ou Muhui Zeng Song Xue Jieli Chen Yao Lu Changhai Ding 2023Bioactive Materials2023,,1:2
4Direct conversion of mouse astrocytes into neural progenitor cells and specific lineages of neurons显示文摘Background:Cell replacement therapy has been envisioned as a promising treatment for neurodegenerative diseases.Due to the ethical concerns of ESCs-derived neural progenitor cells(NPCs)and tumorigenic potential of iPSCs,reprogramming of somatic cells directly into multipotent NPCs has emerged as a preferred approach for cell transplantation.Methods:Mouse astrocytes were reprogrammed into NPCs by the overexpression of transcription factors(TFs)Foxg1,Sox2,and Brn2.The generation of subtypes of neurons was directed by the force expression of cell-type specific TFs Lhx8 or Foxa2/Lmx1a.Results:Astrocyte-derived induced NPCs(AiNPCs)share high similarities,including the expression of NPC-specific genes,DNA methylation patterns,the ability to proliferate and differentiate,with the wild type NPCs.The AiNPCs are committed to the forebrain identity and predominantly differentiated into glutamatergic and GABAergic neuronal subtypes.Interestingly,additional overexpression of TFs Lhx8 and Foxa2/Lmx1a in AiNPCs promoted cholinergic and dopaminergic neuronal differentiation,respectively.Conclusions:Our studies suggest that astrocytes can be converted into AiNPCs and lineage-committed AiNPCs can acquire differentiation potential of other lineages through forced expression of specific TFs.Understanding the impact of the TF sets on the reprogramming and differentiation into specific lineages of neurons will provide valuable strategies for astrocyte-based cell therapy in neurodegenerative diseases.Kangmu Ma Xiaobei Deng Xiaohuan Xia Zhaohuan Fan Xinrui Qi Yongxiang Wang Yuju Li Yizhao Ma Qiang Chen Hui Peng Jianqing Ding Chunhong Li Yunlong Huang Changhai Tian Jialin C.Zheng 2018Translational Neurodegeneration2018,7,1:2
5Inflammatory phenotype of osteoarthritis and its potential therapies显示文摘Introduction:Osteoarthritis(OA)is the most common joint disorder in the world but there are no approved therapies to slow disease progression.Historically,OA has been considered a wear and tear joint disease and efforts to identify and develop disease modifying therapies have primarily focused on inhibiting cartilage degradation directly.However,increasing studies have suggested that inflammation is a key player of joint pathology in some OA patients and also a link between obesity and OA.Therefore,targeting inflammation in these patients could be a promising therapeutic strategy.Areas Covered:In this review,we collate the latest evidence on the relationship between inflammation and OA which allows to propose the inflammatory phenotype of OA,and summarize major results of some emerging therapies targeting this phenotype in clinical trials which could implicate future directions for OA treatment.Expert Opinion:The authors believe early diagnosis,phenotyping OA patients and precise therapies would expedite the development of DMOADs targeting OA inflammatory phenotype.In the future,well-designed clinical trials including appropriate subgroups or phenotypes of patients according to underlying causes and mechanisms of OA are expected for investigational therapies.Yumei Cao Su'an Tang Changhai Ding 2021Rheumatology & Autoimmunity2021,1,2:1
