1The GEON service-oriented architecture for Earth Science applications显示文摘The Geosciences Network(GEON)project has been developing cyberinfrastructure for data sharing in the Earth Science community based on a serviceoriented architecture.The layered architecture consists of Core,Middleware,and Applications services.Core services provide system-level functions(e.g.user authentication),Middleware services provide generic capabilities(e.g.catalog search),and Application services provide functions that users directly interact with,including applications that are specific to Earth Sciences.The GEON‘service stack’includes a standardized set of these services and the corresponding software modules.The GEON Portal provides Web-based access to these services via a set of portlets.This service-oriented approach has enabled GEON to expand to new partner sites and leverage GEON services for other projects.To facilitate interoperation in a distributed geoinformatics environment,GEON is focusing on standards for distributed search across federated catalogs.Chaitan Baru Sandeep Chandra Kai Lin Ashraf Memon Choonhan Youn 2009International Journal of Digital Earth2009,2,S01:3

17Fragment antigen binding domains (F_(ab)s) as tools to study assembly-line polyketide synthases显示文摘The crystallization of proteins remains a bottleneck in our fundamental understanding of their functions.Therefore,discovering tools that aid crystallization is crucial.In this review,the versatility of fragment-antigen binding domains(F_(ab)s)as protein crystallization chaperones is discussed.F_(ab)s have aided the crystallization of membrane-bound and soluble proteins as well as RNA.The ability to bind three F_(ab)s onto a single protein target has demonstrated their potential for crystallization of challenging proteins.We describe a high-throughput workflow for identifying F_(ab)s to aid the crystallization of a protein of interest(POI)by leveraging phage display technologies and differential scanning fluorimetry(DSF).This workflow has proven to be especially effective in our structural studies of assembly-line polyketide synthases(PKSs),which harbor flexible domains and assume transient conformations.PKSs are of interest to us due to their ability to synthesize an unusually broad range of medicinally relevant compounds.Despite years of research studying these megasynthases,their overall topology has remained elusive.One F ab in particular,1B2,has successfully enabled X-ray crystallographic and single particle cryo-electron microscopic(cryoEM)analyses of multiple modules from distinct assembly-line PKSs.Its use has not only facilitated multidomain protein crystallization but has also enhanced particle quality via cryoEM,thereby enabling the visualization of intact PKS modules at near-atomic(3–5Å)resolution.The identification of PKS-binding F_(ab)s can be expected to continue playing a key role in furthering our knowledge of polyketide biosynthesis on assembly-line PKSs.Katarina M.Guzman Chaitan Khosla 2022Synthetic and Systems Biotechnology2022,7,1:0
18Mediating among GeoSciML resources显示文摘Integration of data across multiple independently developed data sources can be challenging due to a variety of heterogeneities that exist across such systems.Data mediation technologies provide approaches for overcoming these heterogeneities.Standards such as Geoscience Markup Language can address some of the heterogeneity issues by providing schema standards which sources can adhere to.This article addresses the issue of semantic heterogeneity across information resources by using domain ontologies and registering schema elements and data values to such ontologies.Registering data to ontologies provides a powerful search and data integration capability across disparate geoscience information resources.Chaitan Baru Kai Lin 2009International Journal of Digital Earth2009,2,S01:0
