16Moving interdisciplinary science forward:integrating participatory modelling with mathematical modelling of zoonotic disease in Africa显示文摘This review outlines the benefits of using multiple approaches to improve model design and facilitate multidisciplinary research into infectious diseases,as well as showing and proposing practical examples of effective integration.It looks particularly at the benefits of using participatory research in conjunction with traditional modelling methods to potentially improve disease research,control and management.Integrated approaches can lead to more realistic mathematical models which in turn can assist with making policy decisions that reduce disease and benefit local people.The emergence,risk,spread and control of diseases are affected by many complex bio-physical,environmental and socio-economic factors.These include climate and environmental change,land-use variation,changes in population and people’s behaviour.The evidence base for this scoping review comes from the work of a consortium,with the aim of integrating modelling approaches traditionally used in epidemiological,ecological and development research.A total of five examples of the impacts of participatory research on the choice of model structure are presented.Example 1 focused on using participatory research as a tool to structure a model.Example 2 looks at identifying the most relevant parameters of the system.Example 3 concentrates on identifying the most relevant regime of the system(e.g.,temporal stability or otherwise),Example 4 examines the feedbacks from mathematical models to guide participatory research and Example 5 goes beyond the so-far described two-way interplay between participatory and mathematical approaches to look at the integration of multiple methods and frameworks.This scoping review describes examples of best practice in the use of participatory methods,illustrating their potential to overcome disciplinary hurdles and promote multidisciplinary collaboration,with the aim of making models and their predictions more useful for decision-making and policy formulation.Catherine Grant Giovanni Lo Iacono Vupenyu Dzingirai Bernard Bett Thomas R.A.Winnebah Peter M.Atkinson 2016Infectious Diseases of Poverty2016,5,1:1
17MFSD7c functions as a transporter of choline at the blood–brain barrier显示文摘Mutations in the orphan transporter MFSD7c(also known as Flvcr2),are linked to Fowler syndrome.Here,we used Mfsd7c knockout(Mfsd7c–/–)mice and cell-based assays to reveal that MFSD7c is a choline transporter at the blood–brain barrier(BBB).We performed comprehensive metabolomics analysis and detected differential changes of metabolites in the brains and livers of Mfsd7c–/–embryos.Particularly,we found that choline-related metabolites were altered in the brains but not in the livers of Mfsd7c–/–embryos.Thus,we hypothesized that MFSD7c regulates the level of choline in the brain.Indeed,expression of human MFSD7c in cells significantly increased choline uptake.Interestingly,we showed that choline uptake by MFSD7c is greatly increased by choline-metabolizing enzymes,leading us to demonstrate that MFSD7c is a facilitative transporter of choline.Furthermore,single-cell patch clamp analysis showed that the import of choline by MFSD7c is electrogenic.Choline transport function of MFSD7c was shown to be conserved in vertebrates,but not in yeasts.We demonstrated that human MFSD7c is a functional ortholog of HNM1,the yeast choline importer.We also showed that several missense mutations identified in patients exhibiting Fowler syndrome had abolished or reduced choline transport activity.Mice lacking Mfsd7c in endothelial cells of the central nervous system suppressed the import of exogenous choline from blood but unexpectedly had increased choline levels in the brain.Stable-isotope tracing study revealed that MFSD7c was required for exporting choline derived from lysophosphatidylcholine in the brain.Collectively,our work identifies MFSD7c as a choline exporter at the BBB and provides a foundation for future work to reveal the disease mechanisms of Fowler syndrome.Xuan Thi Anh Nguyen Thanh Nha Uyen Le Toan Q.Nguyen Hoa Thi Thuy Ha Anna Artati Nancy C.P.Leong Dat T.Nguyen Pei Yen Lim Adelia Vicanatalita Susanto Qianhui Huang Ling Fam Lo Ngah Leong Isabelle Bonne Angela Lee Jorge L.Granadillo Catherine Gooch Dejie Yu Hua Huang Tuck Wah Soong Matthew Wook Chang Markus R.Wenk Jerzy Adamski Amaury Cazenave-Gassiot Long N.Nguyen 2024Cell Research2024,34,3:0
18Transcatheter Device Closure of a Perimembranous Ventricular Septal Defect in Congenitally Corrected Transposition of the Great Arteries显示文摘The majority of patients with congenitally corrected transposition of the great arteries,also known as transposition of the great arteries{S,L,L}have ventricular septal defects(VSD),most commonly perimembranous VSD(pmVSD).Transcatheter device closure of pmVSD in these patients has not been widely described.We present a case of device closure of pmVSD in L-TGA with an Amplatzer Duct Occluder II(ADOII)device using a deployment starting in the subpulmonary left ventricle.The case demonstrates some of the technical advantages of the ADOII device for VSD closure,specifically its low profile,symmetric shape,and soft material.These characteristics are advantageous in closing conventional pmVSD but are especially useful in patients with challenging anatomic substrates.Catherine E.Tomasulo Lindsay S.Rogers Lauren Andrade Michael LO’Byrne 2022Congenital Heart Disease2022,17,2:0
