2甲基硒酸对人高转移大细胞肺癌细胞株L9981增殖和凋亡作用及其分子机制的初步研究显示文摘背景与目的已有的研究表明硒具有防癌作用和抑制乳腺癌、前列腺癌细胞株增殖的作用,但有关甲基硒酸(MSA)是否具有抗肺癌的作用目前尚不清楚。本研究的目的是探讨MSA对人高转移大细胞肺癌细胞株L9981是否具有生长抑制及诱导凋亡的作用,并对其分子机制作初步探讨。方法应用体外细胞培养技术,以MSA处理L9981细胞株,应用台盼蓝计数法、克隆形成实验和流式细胞术检测MSA处理L9981细胞株前后细胞株体外增殖、克隆形成和凋亡水平的变化。应用流式细胞术检测MSA处理L9981细胞株前后细胞周期、细胞周期相关蛋白和凋亡相关蛋白表达水平的变化。结果①MSA在大于0.5μmol/L的浓度时可显著抑制L9981细胞株的增殖(P<0.05),细胞被阻滞于G0/G1期。②MSA浓度在2.5μmol/L时可诱导L9981细胞株进入凋亡(P<0.05)。③MSA在5μmol/L时可明显抑制L9981细胞株的克隆形成能力(P<0.05)。④MSA能显著上调P53、P21、Fas、FasL和Bax蛋白表达水平。结论①MSA可显著抑制人高转移大细胞肺癌细胞株L9981的体外增殖和克隆形成能力,并诱导细胞凋亡;②MSA的抗肺癌作用可能与其调控肺癌细胞株L9981细胞周期和细胞凋亡相关基因表达有关。刘洁薇 钟晓蓉 周清华 Allen C.Gao 王艳萍 朱文 马力 张芷旋 2006中国肺癌杂志2006,9,2:6
3Machine-learning informed prediction of high-entropy solid solution formation:Beyond the Hume-Rothery rules显示文摘The empirical rules for the prediction of solid solution formation proposed so far in the literature usually have very compromised predictability.Some rules with seemingly good predictability were,however,tested using small data sets.Based on an unprecedented large dataset containing 1252 multicomponent alloys,machine-learning methods showed that the formation of solid solutions can be very accurately predicted(93%).The machine-learning results help identify the most important features,such as molar volume,bulk modulus,and melting temperature.Zongrui Pei Junqi Yin Jeffrey A.Hawk David E.Alman Michael C.Gao 2020npj Computational Materials2020,,1:6

5Effects of effective stress and temperature on permeability of sandstone from CO2-plume geothermal reservoir显示文摘Rock is generally complex and heterogeneous,therefore the heterogeneity effects of effective stress and temperature on permeability should be taken into account.In this study,two-part Hooke's model(TPHM) is introduced to understand the influences of effective stress and temperature on permeability of soft and hard parts(two parts) of rock based on coupling thermo-hydro-mechanical tests.Under a fixed temperature level(25 ℃.35 ℃.50 ℃.65 ℃.80 ℃.90 ℃ and 95 ℃).the tests were carried out in a conventional triaxial system whereas the confining pressure was remained at 50 MPa.and the pore pressure was increased to the specified levels step by step.i.e.8 MPa,18 MPa.28 MPa.38 MPa.41 MPa,44 MPa.46 MPa and 48 MPa.The temperature-dependent relationships for two parts permeabilities are proposed on the basis of the initial test results.We point out that temperature of 65 ℃-90 ℃ is the threshold for the development of CO_2-plume geothermal(CPC) reservoir sandstone cracking under low effective stress(2-9 MPa) based on the relationship between temperature and soft part permeability.Furthermore,we discuss the effect of temperature on the two parts in the rock.The results indicate that as the temperature increases from 25 ℃ to 65 ℃.the flow channel in the hard part has a stronger response to temperature than that in the soft part at a fixed effective stress level,which is opposite to the situation of effective stress.Considering that natural rock is generally heterogeneous with non-uniform pore structure,we suggest a physical interpretation of the phenomenon that before the thermal cracking threshold the two parts have different responses to temperature.Y.Z.Sun L.Z.Xie B.He C.Gao J.Wang 2016Journal of Rock Mechanics and Geotechnical Engineering2016,8,6:4
6Adaptive pathways and emerging strategies overcoming treatment resistance in castration resistant prostate cancer显示文摘The therapies available for prostate cancer patients whom progress from hormonesensitive to castration resistant prostate cancer include both systemic drugs,including docetaxel and cabazitaxel,and drugs that inhibit androgen signaling such as enzalutamide and abiraterone.Unfortunately,it is estimated that up to 30%of patients have primary resistance to these treatments and over time even those who initially respond to therapy will eventually develop resistance and their disease will continue to progress regardless of the presence of the drug.Determining the mechanisms involved in the development of resistance to these therapies has been the area of intense study and several adaptive pathways have been uncovered.Androgen receptor(AR)mutations,expression of AR-V7(or other constitutively active androgen receptor variants),intracrine androgen production and overexpression of androgen synthesis enzymes such as Aldo-Keto Reductase Family 1,Member C3(AKR1C3)are among the many mechanisms associated with resistance to anti-androgens.In regards to the taxanes,one of the key contributors to drug resistance is increased drug efflux through ATP Binding Cassette Subfamily B Member 1(ABCB1).Targeting these resistance mechanisms using different strategies has led to various levels of success in overcoming resistance to current therapies.For instance,targeting AR-V7 with niclosamide or AKR1C3 with indomethacin can improve enzalutamide and abiraterone treatment.ABCB1 transport activity can be inhibited by the dietary constituent apigenin and antiandrogens such as bicalutamide which in turn improves response to docetaxel.A more thorough understanding of how drug resistance develops will lead to improved treatment strategies.This review will cover the current knowledge of resistance mechanisms to castration resistant prostate cancer therapies and methods that have been identified which may improve treatment response.Cameron M.Armstrong Allen C.Gao 2016Asian Journal of Urology2016,3,4:3
7Current strategies for targeting the activity of androgen receptor variants显示文摘Current therapies for advanced prostate cancer,such as enzalutamide and abiraterone,focus on inhibiting androgen receptor(AR)activity and reducing downstream signaling pathways to inhibit tumor growth.Unfortunately,cancer cells are very adaptable and,over time,these cells develop mechanisms by which they can circumvent therapeutics.One of the many mechanisms that have been discovered is the generation of AR variants.These variants are generated through alternative splicing of the full length AR and often lack the ligand binding domain.This leads to forms of the AR that are constitutively active that continue to promote prostate cancer cell growth even in the absence of ligand.The high prevalence of AR variants and their role in disease progression have prompted a number of studies investigating ways to inhibited AR variant expression and activity.Among these are the antihelminthic drug,niclosamide,which selectively promotes degradation of AR variants over full length AR and re-sensitizes anti-androgen resistant prostate cancer cells to treatment with enzalutamide and abiraterone.Other AR variant targeting mechanisms include interfering with AR variant co-activators and the development of drugs that bind to the DNA or N-terminal AR domains,which are retained in most AR variants.The clinical efficacy of treating prostate cancer by targeting AR variants is under investigation in several clinical trials.In this review,we provide an overview of the most relevant AR variants and discuss current AR variant targeting strategies.Cameron M.Armstrong Allen C.Gao 2019Asian Journal of Urology2019,6,1:3
8First-principles prediction of high-entropy-alloy stability显示文摘High entropy alloys(HEAs)are multicomponent compounds whose high configurational entropy allows them to solidify into a single phase,with a simple crystal lattice structure.Some HEAs exhibit desirable properties,such as high specific strength,ductility,and corrosion resistance,while challenging the scientist to make confident predictions in the face of multiple competing phases.We demonstrate phase stability in the multicomponent alloy system of Cr-Mo-Nb-V,for which some of its binary subsystems are subject to phase separation and complex intermetallic-phase formation.Our first-principles calculation of free energy predicts that the configurational entropy stabilizes a single body-centered cubic(BCC)phase from T=1700 K up to melting,while precipitation of a complex intermetallic is favored at lower temperatures.We form the compound experimentally and confirm that it develops as a single BCC phase from the melt,but that it transforms reversibly at lower temperatures.Rui Feng Peter K.Liaw Michael C.Gao Michael Widom 2017npj Computational Materials2017,,1:1

13B3 0749+460A: A New Repeat “Changing-look” Active Galactic Nucleus Associated with X-Ray Spectral Slope Variations显示文摘Here we report an identification of B3 0749+460 A as a new double-peaked local 'changing-look' active galactic nucleus(CL-AGN) in terms of our multi-epoch spectroscopic analysis. By comparing our new spectra taken in 2021 with the ones taken by the Sloan Digital Sky Survey in 2004, BOSS in 2013 and Ma NGA in 2016, we reveal type transitions of Seyfert(Sy) 1.9 → Sy1.8 → Sy1.9. In the transitions, the classical broad Hα emission fades away after 2013 and disappears in our 2021 spectrum, although the absence of broad Hβ can be traced back to at least 2016. A follow-up observation in X-rays by the Swift/XRT reveals that(1) the X-ray emission level gradually decreased since 2005;and(2)the X-ray spectrum is soft in the optical 'turn-off' state and hard in the 'turn-on' state.
