| 1 | Efficient generation of transgenic chickens using the spermatogonial stem cells in vivo and ex vivo transfection显示文摘The highly efficient novel methods to produce transgenic chickens were established by directly in-jecting the recombinant plasmid containing green fluorescent protein (GFP) gene into the cock's testis termed as testis-medianted gene transfer (TMGT), and transplanting transfected spermatogonial stem cells (TTSSCs). For the TMGT approach,four dosages of pEGFP-N1 DNA/cationic polymer complex were injected intratesticularly. The results showed: (1) 48 h after the injection,the percentages of testis cells expressing GFP were 4.0%, 8.7%, 10.2% and 13.6% in the 50, 100, 150 and 200 μg/mL group, re-spectively. The difference from the four dosage groups was significant (P<0.05). On day 25 after the injection, a dosage-dependent and time-dependent increase in the number of transgenic sperm was observed. The percentages of gene expression reached the summit and became stable from day 70 to 160, being 12.7%, 12.8%, 15.9% and 19.1%, respectively. The difference from the four dosage groups was also significant (P<0.05). (2) 70 d after the injection, strong green fluorescent could be observed in the seminiferous tubules by whole-mount in-situ hybridization. (3) 70 d after the injection, the semen was collected and used to artificially inseminate wild-type females. The blastoderms of F1 and F2 transgenic chicken expressed GFP were 56.2% (254/452) and 53.2% (275/517), respectively. The detec-tion of polymerase chain reaction (PCR) of F1 and F2 transgenic chicken blood genomic DNA showed that 56.5% (3/23) of F1 and 52.9% (9/17) of F2 were positive. Southern blot showed GFP DNA was in-serted in their genomic DNAs. (4) Frozen whole mount tissue sections of F1 and F2 transgenic chicken liver, heart, kidney and muscle showed that the rates of green fluorescent positive were between 50.0% and 66.7%. (5) With the TTSSCs method, SSCs ex vivo transfected with GFP were transplanted into recipient roosters whose endogenic SSCs had been resoluted. The donor SSCs settled and GFP ex-pression became readily detectable in the frozen whole mount tissue sections of recepient testes. Moreover, sperms carrying GFP could be produced normally. The results of artificially inseminating wild-type females with these sperms showed 12.5% (8/64) of offspring embryo expressed GFP and 11.1% (2/18) hatched chicks were tested transgenic. | 6 | Kinematics Analysis of Motion Simulation Subsystem for Unmanned Vehicle显示文摘Unmanned vehicle has attracted wide attention and interests throughout the world since it first deputed in the 1960s. However, the experimental methods for unmanned vehicle's intelligent behavior, such as semi-physical simulation and motion subsystem, have not been widely explored. First, the requirements of the motion subsystem in unmanned vehicle semi-physical facility are analyzed, and a six DOF parallel manipulator is selected to reproduce the pose of the vehicle. The link lengths of the motion subsystem are worked out under the given rotational angles of the vehicle. According to the geometric properties of tetrahedron, three joint positions of the top platform are determined, and the rest are obtained from the first three position vectors. Six constraint equations are set up based on the vertices on the top platform and the link lengths. In order to solve the six angle variables, a numerical algorithm built on the Newton-Raphson iterative method is presented, which is based on Taylor series expansion of six constraint equations. The pose of the top platform is ultimately calculated. The eigenvalues of the top platform are solved to obtain the natural frequencies of the motion subsystem. The coordinates of six joint centers on the top platform and six constraint equations can be realized by simple algebraic manipulation, which allows significant abbreviation in the formulation and provides a systematic way of obtaining the kinematic solution of the parallel manipulator. A numerical example is given and its efficacy is demonstrated by the inverse kinematics. | 8 | An adaptive offloading framework for Android applications in mobile edge computing显示文摘Mobile edge computing(MEC) provides a fresh opportunity to significantly reduce the latency and battery energy consumption of mobile applications. It does so by enabling the offloading of parts of the applications on mobile edges, which are located in close proximity to the mobile devices. Owing to the geographical distribution of mobile edges and the mobility of mobile devices, the runtime environment of MEC is highly complex and dynamic. As a result, it is challenging for application developers to support computation offloading in MEC compared with the traditional approach in mobile cloud computing, where applications use only the cloud for offloading. On the one hand, developers have to make the offloading adaptive to the changing environment, where the offloading should dynamically occur among available computation nodes.On the other hand, developers have to effectively determine the offloading scheme each time the environment changes. To address these challenges, this paper proposes an adaptive framework that supports mobile applications with offloading capabilities in MEC. First, based on our previous study(DPartner), a new design pattern is proposed to enable an application to be dynamically offloaded among mobile devices, mobile edges,and the cloud. Second, an estimation model is designed to automatically determine the offloading scheme.In this model, different parts of the application may be executed on different computation nodes. Finally, an adaptive offloading framework is implemented to support the design pattern and the estimation model. We evaluate our framework on two real-world applications. | 12 | Promotion effect of adsorbed water/OH on the catalytic performance of Ag/activated carbon catalysts for CO preferential oxidation in excess H_2显示文摘Activated carbon (AC) supported silver catalysts were prepared by incipient wetness impregnation method and their catalytic performance for CO preferential oxidation (PROX) in excess H2 was evaluated. Ag/AC catalysts, after reduction in H2 at low temperatures (≤200℃) following heat treatment in He at 200℃ (He200H200), exhibited the best catalytic properties. Temperature-programmed desorption (TPD), X-ray diffraction (XRD) and temperature-programmed reduction (TPR) results indicated that silver oxides were produced during heat treatment in He at 200℃ which were reduced to metal silver nanoparticles in H2 at low temperatures (≤200℃), simultaneously generating the adsorbed water/OH. CO conversion was enhanced 40% after water treatment following heat treatment in He at 600℃. These results imply that the metal silver nanoparticles are the active species and the adsorbed water/OH has noticeable promotion effects on CO oxidation. However, the promotion effect is still limited compared to gold catalysts under the similar conditions, which may be the reason of low selectivity to CO oxidation in PROX over silver catalysts. The reported Ag/AC-S-He catalyst after He200H200 treatment displayed similar PROX of CO reaction properties to Ag/SiO2 . | 17 | Synthesis of hierarchical porous carbon with high surface area by chemical activation of(NH_(4))_(2)C_(2)O_(4) modified hydrochar for chlorobenzene adsorption显示文摘In this work,hydrothermal technique combined with KOH activation were employed to develop a series of porous carbons (NPCK-x) using tobacco stem as a low-cost carbon source and (NH_(4))_(2)C_(2)O_(4)as a novel nitrogen-doping agent.Physicochemical properties of NPCK-x were characterized by Brunauer-Emmett-Teller,field emission scanning electron microscopy,X-ray diffraction,Raman microscope,elemental analysis,and X-ray photoelectron spectroscopy.Results showed that the NPCK-x samples possessed large surface areas (maximum:2875 m^(2)/g),hierarchical porous structures,and high degree of disorder.N-containing functional groups decomposed during activation process,which could be the dominant reason for appearance of abundant mesopores and well-developed pore structure.Dynamic chlorobenzene adsorption experiments demonstrated that carbon materials with(NH_(4))_(2)C_(2)O_(4)modification exhibited higher adsorption capacity (maximum:1053 mg/g) than those without modification (maximum:723 mg/g).The reusability studies of chlorobenzene indicated that the desorption efficiency of (NH_(4))_(2)C_(2)O_(4)modified porous carbon reached90.40%after thermal desorption at 100℃ under N2atmosphere.Thomas model fitting results exhibited that the existence of mesopores accelerated the diffusion rate of chlorobenzene in porous carbon.Moreover,Grand Canonical Monte Carlo simulation was conducted to verify that micropores with pore sizes of 1.2–2 nm of the optimized porous carbon were the best adsorption sites for chlorobenzene and mesopores with pore sizes of 2–5 nm were also highly active sites for chlorobenzene adsorption. | Jie Wen Zewei Liu Hongxia Xi Bichun Huang | 2023 | Journal of Environmental Sciences2023,,4: | 0 |
