| 1 | Resveratrol inhibits matrix metalloproteinases to attenuate neuronal damage in cerebral ischemia:a molecular docking study exploring possible neuroprotection显示文摘The main pathophysiology of cerebral ischemia is the structural alteration in the neurovascular unit, coinciding with neurovascular matrix degradation. Resveratrol has been reported to be one of the most potent chemopreventive agents that can inhibit cellular processes associated with ischemic stroke. Matrix metalloproteinases(MMPs) has been considered as a potential drug target for the treatment of cerebral ischemia. To explore this, we tried to investigate the interaction of resveratrol with MMPs through molecular docking studies. At 30 minutes before and 2 hours after cerebral ischemia/reperfusion induced by occlusion of the middle cerebral artery, 40 mg/kg resveratrol was intraperitoneally administered. After resveratrol administration, neurological function and brain edema were significantly alleviated, cerebral infarct volume was significantly reduced, and nitrite and malondialdehyde levels in the cortical and striatal regions were significantly decreased. The molecular docking study of resveratrol and MMPs revealed that resveratrol occupied the active site of MMP-2 and MMP-9. The binding energy of the complexes was –37.848672 k J/mol and –36.6345 k J/mol for MMP-2 and MMP-9, respectively. In case of MMP-2, Leu 164, Ala 165 and Thr 227 were engaged in H-Bonding with resveratrol and in case of MMP-9, H-bonding was found with Glu 402, Ala 417 and Arg 424 residues. | 2 | Piroxicam-mediated modulatory action of 5-hydroxytryptamine serves as a “brake” on neuronal excitability in ischemic stroke显示文摘Our previous studies indicated an increase in extracellular γ-aminobutyric acid(GABA) in rodent's ischemic brain after Piroxicam administration, leading to alleviation of glutamate mediated excitotoxicity through activation of type A GABA receptor(GABAA). This study was to investigate if GABAA activation by Piroxicam affects extracellular 5-hydroxytryptamine or not. High performance liquid chromatography revealed that there was a significant decrease in extracellular 5-hydroxytryptamine release in ischemic cerebral cortex and striatum in Piroxicam pre-treated rat brains. | 3 | A possible therapeutic potential of quercetin through inhibition of μ-calpain in hypoxia induced neuronal injury:a molecular dynamics simulation study显示文摘The neuroprotective property of quercetin is well reported against hypoxia and ischemia in past studies.This property of quercetin lies in its antioxidant property with blood-brain barrier permeability and anti-inflammatory capabilities.μ-Calpain,a calcium ion activated intracellular cysteine protease causes serious cellular insult,leading to cell death in various pathological conditions including hypoxia and ischemic stroke.Hence,it may be considered as a potential drug target for the treatment of hypoxia induced neuronal injury.As the inhibitory property of μ-calpain is yet to be explored in details,hence,in the present study,we investigated the interaction of quercetin with μ-calpain through a molecular dynamics simulation study as a tool through clarifying the molecular mechanism of such inhibition and determining the probable sites and modes of quercetin interaction with the μ-calpain catalytic domain.In addition,we also investigated the structure-activity relationship of quercetin with μ-calpain.Affinity binding of quercetin with μ-calpain had a value of –28.73 k J/mol and a Ki value of 35.87 μM that may be a probable reason to lead to altered functioning of μ-calpain.Hence,quercetin was found to be an inhibitor of μ-calpain which might have a possible therapeutic role in hypoxic injury. | Anand Kumar Pandey Swet Chand Shukla Pallab Bhattacharya Ranjana Patnaik | 2016 | Neural Regeneration Research2016,11,8: | 3 |

| 13 | Numerical analysis of hemodynamic parameters in stenosed arteries under pulsatile flow conditions显示文摘This research studies the changes in flow patterns and hemodynamic parameters of diverse shapes and sizes of stenosis.Six different shapes and sizes of stenosis are constructed to investigate the variations in hemodynamics as the morphology changes.Changes in shape(trapezoidal and bell-shaped)and sizes of stenosis change the stresses on the walls and their flow patterns.TAWSS and OSI results specify that trapezoidal stenosis exerts greater stress than bell-shaped stenosis.Also,as the length of the trapezoidal stenosis increases,the TAWSS increases,whereas the trend is the opposite for bell-shaped stenosis.Later,this paper also studies different degrees of stenosis extracted from real images.Changes in velocity flow patterns,wall shear stress(WSS),Time-averaged wall shear stress(TAWSS)and Oscillatory shear index(OSI)have been studied for these images.Results illustrate that the peak velocity rises drastically as the stenosis percentage increases.Negative velocity is seen close to the artery's walls,indicating flow separation.This flow separation region is seen throughout the cycle except in the accelerating flow region.An increase in stenosis also increases WSS and TAWSS drastically.Negative WSS is seen downstream of stenosis,indicating flow recirculation.Such negative WSS in the blood vessels also promotes endothelial dysfunction.OSI values greater than 0.2 are seen near the stenosis region,indicating atherosclerosis growth.Regions of high OSI and low TAWSS are also identified,indicating probable regions of plaque development. | Priyambada Praharaj Chandrakant Sonawane Anand Pandey Vikas Kumar Arundhati Warke Hitesh Panchal R.Ibrahim Chander Prakash | 2023 | Medicine in Novel Technology and Devices2023,,4: | 0 |

| 15 | Evaluation of thermostable endoglucanase in Paenibacillus lautus strain BHU3 for yield enhancement显示文摘In the present study,Paenibacillus lautus strain BHU3 isolated from landfill soil was evaluated for the presence of potential endoglucanases which are the first candidate of cellulase enzyme system to act on cellulose.In-silico analysis revealed high potential thermostable endoglucanases which can efficiently interact with cellulose.The most potent and thermostable endoglucanase(locus tag id.CPZ30_18280)belonged to glycosyl family-5 and had interaction energy of−12.981 kcal/mol for the best docked cluster containing three out of ten docking conformations,and Tm value of 73.3℃.MD simulation of 100 ns proved highly stable binding interactions of CPZ30_18280 endoglucanase with cellulose with root mean square deviation(RMSD)values ranging from 0.15 to 0.30 nm.Consistent interactions with characteristic active site residues(tyrosine,tryptophan and aspartate)of glycosyl family-5 endoglucanases were found.Further,to enhance the production of endoglucanases,the fermentation conditions were optimized employing approaches like one factor at a time(OFAT)and response surface methodology(RSM).Maximum activity of endoglucanase was determined at 60℃.The optimized condi-tions for enhanced production of endoglucanase(10.15 U/mL)were pH 6.63,yeast extract conc.3.44 g/L,wheat bran 3.59%,and inoculum size 2.65%.Hence,P.lautus strain BHU3 has enormous potential to synthesize highly efficient thermostable endoglucanases under optimized regime using agro-wastes.Thus,it could find immense industrial applications including large scale cellulose conversion to bioethanol. | Suman Yadav Anand Kumar Pandey Suresh Kumar Dubey | 2022 | Systems Microbiology and Biomanufacturing2022,2,4: | 0 |
