| 1 | Xenobiotics and loss of tolerance in primary biliary cholangitis显示文摘Data from genome wide association studies and geoepidemiological studies established that a com-bination of genetic predisposition and environmental stimulation is required for the loss of tolerance in primary biliary cholangitis(PBC).The serologic hallmark of PBC are the presence of high titer anti-mitochondrial autoantibodies(AMA)that recognize the lipoyl domain of the mitochondrial pyruvate dehydrogenase E2(PDC--E2)subunit.Extensive efforts have been directed to investigate the molecular basis of AMA.Recently,experimental data has pointed to the thesis that the breaking of tolerance to PDC--E2 is a pivotal event in the initial etiology of PBC,including environmental xenobiotics including those commonly found in cos-metics and food additives,suggesting that chemical modification of the PDC--E2 epitope may render its vulnerable to become a neo-antigen and trigger an immune response in genetically susceptible hosts.Here,we will discuss the natural history,genetics and immunobiology of PBC and structural constraints of PDC--E2 in AMA recognition which makes it vulnerable to chemical modification. | Jinjun Wang Guoxiang Yang Alana Mari Dubrovsky Jinjung Choi Patrick SC Leung | 2016 | World Journal of Gastroenterology2016，22，1： | 4 |
| 2 | Biodegradable nanocarriers for small interfering ribonucleic acid (siRNA) co-delivery strategy increase the chemosensitivity of pancreatic cancer cells to gemcitabine显示文摘Ribonucleic acid (RNA) interference (RNAi) therapies are promising cancertreatment modalities that can specifically target abnormal proto-oncogenes, thusimproving the therapeutic effect. For the treatment of pancreatic cancer, targetingone mutant proto-oncogene by RNAi usually does not yield the desired therapeuticefficiency. Both K-ras gene mutations and Notch1 overexpression are commonsymptoms in pancreatic cancer patients, and play a crucial role in pancreaticcancer cell drug resistance. In this study, biodegradable charged polyester-basedvectors (BCPVs) were synthesized for the co-delivery of K-ras and Notch1 smallinterfering ribonucleic acid (siRNA) into MiaPaCa-2 cells (pancreatic cancercell line) to overcome drug resistance to gemcitabine (GEM), a first-line chem-otherapeutic drug used in the clinic. BCPVs could effectively absorb negative siRNAto form a capsule-like structure, prevent siRNA from nuclease digestion in theserum, and promote effective siRNA cell internalization and endosomal escape.Through K-ras and Notch1 gene silencing in MiaPaCa-2 cells, BCPV-siRNAKsiRNAN^tcm nanocomplexes effectively reversed the epithelia-mesenchymaltransition (EMT) in MiaPaCa-2 cells, thereby greatly enhancing the sensitivity ofMiaPaCa-2 cells to GEM. MiaPaCa-2 cell proliferation, migration, and invasionwere effectively inhibited, and cell apoptosis was also significantly enhanced bythe synergistic antitumor effect of BCPV-siRNAK siRNAN^t~l nanocomplexesand GEM. These results suggest that this combination RNAi therapy can be usedto improve cancer cell sensitivity to chemotherapeutic drugs.
