1	通过单细胞测序发现结肠癌双系起源的新型癌基因——SLC12A5显示文摘单细胞测序是一种为划定遗传关系和个性化癌症管理确认重要驱动基因的功能强大的工具。本研究运用单细胞测序分析1例结肠癌。群体遗传学分析确定了肿瘤群体中两个独立的克隆。作为早期致癌事件的大部分肿瘤克隆包含APC和TP 53基因突变,然而少数克隆却包含CDC27和PABPC1基因的优势突变。 APC和TP53基因突变在少数克隆中的缺失说明这两个克隆来自两个细胞起源。对于另外21例整个组织外显子测序并检查其体细胞突变的等位基因频谱,揭示了结肠癌克隆起源具有异质性。该研究确定SLC12A5基因的突变,显示其在单细胞水平的高频突变,然而群体水平上却呈低频突变。 SLC12A5突变的功能特征揭示其在结肠癌中潜在的致癌作用。该研究提供了结肠癌单细胞水平上第一个广谱外显子组的证据,支持结肠癌双系克隆起源以及人群中的低频突变可能发挥的重要促癌作用。	Yu C Yu J Yao X 张俊许春伟张博	2014	临床与实验病理学杂志2014，30，12：	4
