1Optimization of phenol degradation by Candida tropicalis Z-04 using Plackett-Burman design and response surface methodology显示文摘Statistical experimental designs were used to optimize the process of phenol degradation by Candida tropicalis Z-04,isolated from phenol-degrading aerobic granules.The most important factors influencing phenol degradation (p<0.05),as identified by a two-level Plackett-Burman design with 11 variables,were yeast extract,phenol,inoculum size,and temperature.Steepest ascent method was undertaken to determine the optimal regions of these four significant factors.Central composite design (CCD) and response surface analysis were adopted to further investigate the mutual interactions between these variables and to identify their optimal values that would generate maximum phenol degradation.The analysis results indicated that interactions between yeast extract and temperature,phenol and temperature,inoculum size and temperature affected the response variable (phenol degradation) significantly.The predicted results showed that the maximum removal efficiency of phenol (99.10%) could be obtained under the optimum conditions of yeast extract 0.41 g/L,phenol 1.03 g/L,inoculum size 1.43% (V/V) and temperature 30.04°C.These predicted values were further verified by validation experiments.The excellent correlation between predicted and experimental values confirmed the validity and practicability of this statistical optimum strategy.This study indicated the excellent ability of C.tropicalis Z-04 in degrading high-strength phenol.Optimal conditions obtained in this experiment laid a solid foundation for further use of this microorganism in the treatment of highstrength phenol effluents.Jiangya Zhou Xiaojuan Yu Cong Ding Zhiping Wang Qianqian Zhou Hao Pao Weimin Ca 2011Journal of Environmental Sciences2011,23,1:48
2Analysis on the decadal scale variation of the dust storm in North China显示文摘In this paper, the temporal variation characteristics of the dust storm in North China are investigated. Based on power spectrum analysis and wavelet analysis, 1956-1970 and 1985-1999 are chosen as the high-frequency and low-frequency dust storm decades respectively. Analysis results clearly show that the spring and wintertime anomalous atmospheric circulation between these two dec-ades are significantly different. Compared with the former decade, there are a strengthened polar vortex, enhanced westerlies near 50°N, and a weak East Asian major though in the winter of the latter decade. The north and center parts of the Siberian high and the Aleutian low become weak. The southerly and easterly wind anomalies appear over the north and east parts of China, which implies the weakening of East Asian winter monsoon. Meanwhile, northern China experiences warmer winters and wetter springs, which are in favor of the weakening of dust storm intensity in North China. There are significant out-of-phase relationships between dust frequency and wintertime westerly intensity, as well as between dust frequency and Arctic Oscillation. 4No apparent lock-in depth of the Laschamp geomagnetic excursion: Evidence from the Malan loess显示文摘The first-order chronological framework of the long-term Chinese loess/paleosol sequences was based mainly on magnetostratigraphy. However, there remain arguments on the lock-in ages of the major geomagnetic reversals. This study systematically compared the stratigraphic locations of the Laschamp excursion and Henrich events (H4 at~39 ka and H5 at~48 ka) recorded by the Luochuan loess profile in the central Chinese Loess Plateau. 5Characterization of cancer stem-like cells in the side population cells of human gastric cancer cell line MKN-45显示文摘Objective:Side population(SP) cells may play a crucial role in tumorigenesis and the recurrence of cancer.Many kinds of cell lines and tissues have demonstrated the presence of SP cells,including several gastric cancer cell lines.This study is aimed to identify the cancer stem-like cells in the SP of gastric cancer cell line MKN-45.Methods:We used fluorescence activated cell sorting(FACS) to sort SP cells in the human gastric carcinoma cell line MKN-45(cells labeled with Hoechst 33342) and then characterized the cancer stem-like properties of SP cells.Results:This study found that the SP cells had higher clone formation efficiency than major population(MP) cells.Five stemness-related gene expression profiles,including OCT-4,SOX-2,NANOG,CD44,and adenosine triphosphate(ATP)-binding cassette transporters gene ABCG2,were tested in SP and MP cells using quantitative real-time reverse transcription polymerase chain reaction(RT-PCR).Western blot was used to show the difference of protein expression between SP and MP cells.Both results show that there was significantly higher protein expression in SP cells than in MP cells.When inoculated into non-obese diabetic/severe combined immunodeficiency(NOD/SCID) mice,SP cells show higher tumorigenesis tendency than MP cells.Conclusions:These results indicate that SP cells possess cancer stem cell properties and prove that SP cells from MKN-45 are gastric cancer stem-like cells.Hai-hong ZHANG Ai-zhen CA Xue-ming WEI Li DING Feng-zhi LI Ai-ming ZHENG Da-jiang DAI Rong-rong HUANG Hou-jun CAO Hai-yang ZHOU Jian-mei WANG Xue-jing WANG Wei SHI Heng ZHU Xiao-ying YUAN Lin CHEN 2013Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2013,14,3:9

8Applications of stable isotopes to study plant-animal relationships in terrestrial ecosystems显示文摘As natural tracers, stable isotopes have been extensively used in plant physiological, ecological and envi- ronmental research. Recently, animal physiological ecologists have also applied stable isotope techniques to study plant- animal relationships. The isotopic compositions of animal body generally reflect and integrate their diets over a time period ranging from hours to years to the lifetime of an indi- vidual. When animal living habitat changes or animals move to a new environment, the animal isotopic compositions will shift accordingly. Thus, stable isotope signatures of an ani- mal can truly reflect its food sources, habitat, distribution and movement patterns during a given time period. More- over, by analyzing animal-tissue isotopic compositions at different temporal scales, we can improve our understanding of animal adaptation to environmental changes. Stable iso- tope technique also provides an ideal tool to study animal foodweb relationship and community structure because of isotopic fractionation during the processes of nutrient as- similation by animals. Stable isotope technique can continu- ously measure animal trophic position in a foodweb, which can eventually reveal the predator-prey relationship and its role in determining matter balance and energy flow in the entire ecosystem. Stable isotope technique has been one of the most important and efficient tools in studying plant-animal relationship. 10Internet-based collaborative pseudo-dynamic testing of multi-span bridge structure显示文摘In this paper, the first Internet-based collaborative pseudo-dynamic testing of a multi-span bridge system in China's Mainland is presented in detail, which has been carried out by the collaboration of Hunan University (HNU), Harbin Institute of Technology (HIT) and Tsinghua University (TU) in China's Mainland, and the University of Southern California (USC) in the USA. The prototype system of Russion River Bridge at Southern California in America is illustrated. Different testing specimens, testing equipments and substructure boundary conditions at HNU and HIT laboratories are described. A network protocol platform for collaborative pseudo-dynamic testing, called NetSLab, and some function modules developed on the use of its standardized interface are introduced. Aiming at the MTS system widely used in structural testing, two approaches are presented to implement the communication interface between NetSLab and testing equipment, respectively, studied by HNU and HIT. The integrated networked testing system and collaborative testing procedures are presented.
