1The wonders of BMP9:From mesenchymal stem cell differentiation,angiogenesis,neurogenesis,tumorigenesis,and metabolism to regenerative medicine显示文摘Although bone morphogenetic proteins(BMPs)initially showed effective induction of ectopic bone growth in muscle,it has since been determined that these proteins,as members of the TGF-b superfamily,play a diverse and critical array of biological roles.These roles include regulating skeletal and bone formation,angiogenesis,and development and homeostasis of multiple organ systems.Disruptions of the members of the TGF-b/BMP superfamily result in severe skeletal and extra-skeletal irregularities,suggesting high therapeutic potential from understanding this family of BMP proteins.Although it was once one of the least characterized BMPs,BMP9 has revealed itself to have the highest osteogenic potential across numerous experiments both in vitro and in vivo,with recent studies suggesting that the exceptional potency of BMP9 may result from unique signaling pathways that differentiate it from other BMPs.The effectiveness of BMP9 in inducing bone formation was recently revealed in promising experiments that demonstrated efficacy in the repair of critical sized cranial defects as well as compatibility with bone-inducing bio-implants,revealing the great translational promise of BMP9.Furthermore,emerging evidence indicates that,besides its osteogenic activity,BMP9 exerts a broad range of biological functions,including stem cell differentiation,angiogenesis,neurogenesis,tumorigenesis,and metabolism.This review aims to summarize our current understanding of BMP9 across biology and the body.Sami Mostafa Mikhail Pakvasa Elam Coalson Allen Zhu Alex Alverdy Hector Castillo Jiaming Fan Alex Li Yixiao Feng Di Wu Elliott Bishop Scott Du Mia Spezia Alissa Li Ofir Hagag Alison Deng Winny Liu Mingyang Li Sherwin S·Ho Aravind Athiviraham Michael J·Lee Jennifer Moriatis Wolf Guillermo A·Ameer Hue H·Luu Rex C·Haydon Jason Strelzow Kelly Hynes Tong-Chuan He Russell R·Reid 2019Genes & Diseases2019,6,3:15
2Epidemiology of perforated peptic ulcer:Age-and gender-adjusted analysis of incidence and mortality显示文摘AIM:To investigate the epidemiological trends in inci-dence and mortality of perforated peptic ulcer(PPU)in a well-defined Norwegian population. METHODS:A retrospective,population-based,single-center,consecutive cohort study of all patients diag-nosed with benign perforated peptic ulcer.Included were both gastric and duodenal ulcer patients admitted to Stavanger University Hospital between January 2001 and December 2010.Ulcers with a malignant neoplasia diagnosis,verified by histology after biopsy or resection,were excluded.Patients were identified from the hospitals administrative electronic database using pertinent ICD-9 and ICD-10 codes(K25.1,K25.2,K25.5, K25.6,K26.1,K26.2,K26.5,K26.6).Additional searches using appropriate codes for relevant laparoscopic and open surgical procedures(e.g.,JDA 60,JDA 61,JDH 70 and JDH 71)were performed to enable a complete identification of all patients.Patient demographics,presentation patterns and clinical data were retrieved from hospital records and surgical notes.Crude and adjusted incidence and mortality rates were estimated by using national population demographics data. RESULTS:In the study period,a total of 172 patients with PPU were identified.The adjusted incidence rate for the overall 10-year period was 6.5 per 100 000 per year(95%CI:5.6-7.6)and the adjusted mortality rate for the overall 10-year period was 1.1 per 100 000 per year(95%CI:0.7-1.6).A non-significant decline in ad-justed incidence rate from 9.7 to 5.6 occurred during the decade.The standardized mortality ratio for the whole study period was 5.7(95%CI:3.9-8.2),while the total 30-d mortality was 16.3%.No difference in in-cidence or mortality was found between genders.However,for patients≥60 years,the incidence increased over 10-fold,and mortality more than 50-fold,compared to younger ages.The admission rates outside office hours were high with almost two out of three(63%) admissions seen at evening/night time shifts and/or during weekends.The observed seasonal variations in admissions were not statistically significant. 3Overexpression of an ABA biosynthesis gene using a stress-inducible promoter enhances drought resistance in petunia显示文摘The response of plants to drought stress includes reduced transpiration as stomates close in response to increased abscisic acid(ABA)concentrations.Constitutive overexpression of 9-cis-epoxycarotenoid dioxygenase(NCED),a key enzyme in ABA biosynthesis,increases drought resistance,but causes negative pleiotropic effects on plant growth and development.We overexpressed the tomato NCED(LeNCED1)in petunia plants under the control of a stress-inducible promoter,rd29A.Under water stress,the transgenic plants had increased transcripts of NCED mRNA,elevated leaf ABA concentrations,increased concentrations of proline,and a significant increase in drought resistance.The transgenic plants also displayed the expected decreases in stomatal conductance,transpiration,and photosynthesis.After 14 days without water,the control plants were dead,but the transgenic plants,though wilted,recovered fully when re-watered.Well-watered transgenic plants grew like non-transformed control plants and there was no effect of the transgene on seed dormancy.Alejandro C Estrada-Melo Chao Ma Michael S Reid Cai-Zhong Jiang 2015Horticulture Research2015,2,1:12
4A basic helix-loop-helix transcription factor, PhFBH4, regulates flower senescence by modulating ethylene biosynthesis pathway in petunia显示文摘The basic helix-loop-helix(bHLH)transcription factors(TFs)play important roles in regulating multiple biological processes in plants.However,there are few reports about the function of bHLHs in flower senescence.In this study,a bHLH TF,PhFBH4,was found to be dramatically upregulated during flower senescence.Transcription of PhFBH4 is induced by plant hormones and abiotic stress treatments.Silencing of PhFBH4 using virus-induced gene silencing or an antisense approach extended flower longevity,while transgenic petunia flowers with an overexpression construct showed a reduction in flower lifespan.Abundance of transcripts of senescence-related genes(SAG12,SAG29)was significantly changed in petunia PhFBH4 transgenic flowers.Furthermore,silencing or overexpression of PhFBH4 reduced or increased,respectively,transcript abundances of important ethylene biosynthesis-related genes,ACS1 and ACO1,thereby influencing ethylene production.An electrophoretic mobility shift assay showed that the PhFBH4 protein physically interacted with the G-box cis-element in the promoter of ACS1,suggesting that ACS1 was a direct target of the PhFBH4 protein.In addition,ectopic expression of this gene altered plant development including plant height,internode length,and size of leaves and flowers,accompanied by alteration of transcript abundance of the gibberellin biosynthesis-related gene GA2OX3.Our results indicate that PhFBH4 plays an important role in regulating plant growth and development through modulating the ethylene biosynthesis pathway.Jing Yin Xiaoxiao Chang Takao Kasuga Mai Bui Michael S Reid Cai-Zhong Jiang 2015Horticulture Research2015,2,1:7
5Controlling plant architecture by manipulation of gibberellicacid signalling in petunia显示文摘Since stem elongation is a gibberellic acid(GA)response,GA inhibitors are commonly used to control plant height in the production of potted ornamentals and bedding plants.In this study,we investigated interfering with GA signaling by using molecular techniques as an alternative approach.We isolated three putative GID1 genes(PhGID1A,PhGID1B and PhGID1C)encoding GA receptors from petunia.Virus-induced gene silencing(VIGS)of these genes results in stunted growth,dark-green leaves and late-flowering.We also isolated the gai mutant gene(gai-1)from Arabidopsis.We have generated transgenic petunia plants in which the gai mutant protein is over-expressed under the control of a dexamethasone-inducible promoter.This system permits induction of the dominant Arabidopsis gai mutant gene at a desired stage of plant development in petunia plants by the application of dexamethasone(Dex).The induction of gai in Dex-treated T1 petunia seedlings caused dramatic growth retardation with short internodes.Yin-Chih Liang Michael S Reid Cai-Zhong Jiang 2014Horticulture Research2014,1,1:5

14Observation or resection of pancreatic intraductal papillary mucinous neoplasm:An ongoing tug of war显示文摘An increasing number of patients are being referred to pancreatic centres around the world due to often incidentally discovered cystic neoplasms of the pancreas.The evaluation and management of pancreatic cystic neoplasms is a controversial topic and with existing guidelines based on a lack of strong evidence there is discordance between centres and guidelines with regard to when to offer surgery and when to favour surveillance.The frequency,duration and modality of surveillance is also controversial as this is resource-consuming and must be balanced against the perceived benefits and risks involved.While there is consensus that the risk of malignancy should be balanced against the lifeexpectancy and comorbidities,the indications for surgery and surveillance strategies vary among the guidelines.Thus,the tug of war between surveillance or resection continues.Here we discuss the recommendations from guidelines with further accumulating data and emerging reports on intraductal papillary mucinous neoplasm in the literature.Jan Rune Aunan Nigel B.Jamieson Kjetil Sφreide 2019World Journal of Gastrointestinal Oncology2019,11,12:2
