1Targeting the alternative bile acid synthetic pathway for metabolic diseases显示文摘The gut microbiota is profoundly involved in glucose and lipid metabolism,in part by regulating bile acid(BA)metabolism and affecting multiple BA-receptor signaling pathways.BAs are synthesized in the liver by multi-step reactions catalyzed via two distinct routes,the classical pathway(producing the 12α-hydroxylated primary BA,cholic acid),and the alternative pathway(producing the non-12α-hydroxylated primary BA,chenodeoxycholic acid).BA synthesis and excretion is a major pathway of cholesterol and lipid catabolism,and thus,is implicated in a variety of metabolic diseases including obesity,insulin resistance,and nonalcoholic fatty liver disease.Additionally,both oxysterols and BAs function as signaling molecules that activate multiple nuclear and membrane receptor-mediated signaling pathways in various tissues,regulating glucose,lipid homeostasis,inflammation,and energy expenditure.Modulating BA synthesis and composition to regulate BA signaling is an interesting and novel direction for developing therapies for metabolic disease.In this review,we summarize the most recent findings on the role of BA synthetic pathways,with a focus on the role of the alternative pathway,which has been under-investigated,in treating hyperglycemia and fatty liver disease.We also discuss future perspectives to develop promising pharmacological strategies targeting the alternative BA synthetic pathway for the treatment of metabolic diseases.Wei Jia Meilin Wei Cynthia Rajani Xiaojiao Zheng 2021Protein & Cell2021,12,5:17
2Gut microbiota alterations are distinct for primary colorectal cancer and hepatocellular carcinoma显示文摘Colorectal cancer(CRC)and hepatocellular carcinoma(HCC)are the second and third most common causes of death by cancer,respectively.The etiologies of the two cancers are either infectious insult or due to chronic use of alcohol,smoking,diet,obesity and diabetes.Pathological changes in the composition of the gut microbiota that lead to intestinal inflammation are a common factor for both HCC and CRC.However,the gut microbiota of the cancer patient evolves with disease pathogenesis in unique ways that are affected by etiologies and environmental factors.In this review,we examine the changes that occur in the composition of the gut microbiota across the stages of the HCC and CRC.Based on the idea that the gut microbiota are an additional“lifeline”and contribute to the tumor microenvironment,we can observe from previously published literature how the microbiota can cause a shift in the balance from normal→inflammation→diminished inflammation from early to later disease stages.This pattern leads to the hypothesis that tumor survival depends on a less proinflammatory tumor microenvironment.The differences observed in the gut microbiota composition between different disease etiologies as well as between HCC and CRC suggest that the tumor microenvironment is unique for each case.Wei Jia Cynthia Rajani Hongxi Xu Xiaojiao Zheng 2021Protein & Cell2021,12,5:14
3MicroRNA aberrations:An emerging field for gallbladdercancer management显示文摘Gallbladder cancer(GBC) is infrequent but most lethal biliary tract malignancy characterized by an advanced stage diagnosis and poor survival rates attributed to absence of specific symptoms and effective treatment options. These necessitate development of early prognostic/predictive markers and novel therapeutic interventions. Micro RNAs(mi RNAs) are small, noncoding RNA molecules that play a key role in tumor biology by functioning like tumor suppressor- or oncogenes and their aberrant expression are associated with the pathogenesis of several neoplasms with overwhelming clinical implications. Since mi RNA signature is tissue specific, here, we focused on current data concerning the mi RNAs abberations in GBC pathogenesis. In GBC, mi RNAs with tumor suppressor activity(mi R-135-5p, mi R-335, mi R-34 a, mi R-26 a, mi R-146b-5p, Mir-218-5p, mi R-1, mi R-145, mir-130a) were found downregulated, while those with oncogenic property(mi R-20 a, mi R-182, mir-155) were upregulated. The expression profile of mi RNAs was significantly associated with GBC prognosis and prediction, and forced over-expression/ inhibition of these mi RNAs was shown to affect tumor growth and development. Further, differential expression of mi RNAs in the blood samples of GBC patients suggest mi RNAs as promising noninvasive biomarker. 4Coronary artery anomalies overview: The normal and the abnormal显示文摘The aim of this review is to give a comprehensive and concise overview of coronary embryology and normal coronary anatomy, describe common variants of normal and summarize typical patterns of anomalous coronary artery anatomy. Extensive iconography supports the text, with particular attention to images obtained in vivo using non-invasive imaging. We have divided this article into three groups, according to their frequency in the general population: Normal, normal variant and anomaly. Although congenital coronary artery anomalies are relatively uncommon, they are the second most common cause of sudden cardiac death among young athletes and therefore warrant detailed review. Based on the functional relevance of each abnormality, coronary artery anomalies can be classified as anomalies with obligatory ischemia, without ischemia or with exceptional ischemia. The clinical symptoms may include chest pain, dyspnea, palpitations, syncope, cardiomyopathy, arrhythmia, myocardial infarction and sudden cardiac death. 5Bile acids and their effects on diabetes显示文摘Diabetes is a widespread,rapidly increasing metabolic disease that is driven by hyperglycemia.Early glycemic control is of primary importance to avoid vascular complications including development of retinal disorders leading to blindness,end-stage renal disease,and accelerated atherosclerosis with a higher risk of myocardial infarction,stroke and limb amputations.Even after hyperglycemia has been brought under control, 'metabolic memory,'a cluster of irreversible metabolic changes that allow diabetes to progress,may persist depending on the duration of hyperglycemia.Manipulation of bile acid (BA)receptors and the BA pool have been shown to be useful in establishing glycemic control in diabetes due to their ability to regulate energy metabolism by binding and activating nuclear transcription factors such as farnesoid X receptor (FXR)in liver and intestine as well as the G-protein coupled receptor,TGR5,in enteroendocrine cells and pancreatic p-cells.The downstream targets of BA activated FXR,FGF15/21,are also important for glucose/insulin homeostasis.In this review we will discuss the effect of BAs on glucose and lipid metabolism and explore recent research on establishing glycemic control in diabetes through the manipulation of BAs and their receptors in the liver,intestine and pancreas, alteration of the enterohepatic circulation,bariatric surgery and alignment of circadian rhythms.Cynthia Rajani Wei Jia 2018Frontiers of Medicine2018,12,6:8
6Expression profile of cholecystokinin type-A receptor in gallbladder cancer and gallstone disease显示文摘BACKGROUND:Regulatory peptide receptors have attracted the interest of oncologists as a new promising approach for cancer pathology,imaging and therapy.Although cholecystokinin (CCK) is a potent modulator of gallbladder contractility and plays a potential role in pancreatic carcinogenesis through CCK type-A receptor (CCKAR),its role in gallbladder cancer (GBC) is still unknown and immunohistochemical detection of CCKAR in the gallbladder has not yet been reported.This novel case-control study aimed to investigate the expression profile of CCKAR in GBC and gallstone disease (GSD).METHODS:This study included 162 samples of gallbladder:94 from GBC and 68 from GSD.Expression of CCKAR was analyzed by immunohistochemistry and immunoblotting.The results were statistically correlated with disease history including age,sex,presence of gallstone,stage and differentiation.RESULTS:CCKAR was positive in 30/68 (44.1%) of GSD and 72/94 (76.6%) of GBC samples.Fifty-one of the 72 (70.8%) CCKAR-positive GBC samples showed over-expression.Interestingly,consistent results also appeared in the immunoblotting study.CONCLUSIONS:CCKAR expression was significantly increased in GBC compared to GSD.Moreover,CCKAR expression was associated with the degree of tumor differentiation,i.e.,less expression in poorly-differentiated tumors.Thus,it has future prognostic and therapeutic implications in the management of GBC.Rajani Rai Mallika Tewari Mohan Kumar Tej Bali Singh Hari S Shukla 2011Hepatobiliary & Pancreatic Diseases International2011,10,4:7
7Computational Identification of Sweet Wormwood (Artemisia annua) microRNA and Their mRNA Targets显示文摘Despite its efficacy against malaria, the relatively low yield (0.01%-0.8%) of artemisinin in Artemisia annua is a serious limitation to the commercialization of the drug. A better understanding of the biosynthetic pathway of artemisinin and its regulation by both exogenous and endogenous factors is essential to improve artemisinin yield. Increasing evidence has shown that microRNAs (miRNAs) play multiple roles in various biological processes. In this study, we used previously known miRNAs from Arabidopsis and rice against expressed sequence tag (EST) database of A. annua to search for potential miRNAs and their targets in A. annua. A total of six potential miRNAs were predicted, which belong to the miR414 and miR1310 families. Furthermore, eight potential target genes were identified in this species. Among them, seven genes encode proteins that play important roles in ar- temisinin biosynthesis, including HMG-CoA reductase (HMGR), amorpha-4,11-diene synthase (ADS), farnesyl pyrophosphate synthase (FPS) and cytochrome P450. In addition, a gene coding for putative AINTEGUMENTA, which is involved in signal transduction and development, was also predicted as one of the targets. 8Optoelectronic properties and Seebeck coefficient in SnSe thin films显示文摘SnSe thin films of thickness 180 nm have been deposited on glass substrates by reactive evaporation at an optimized substrate temperature of 523 ± 5 K and pressure of 10^(-5) mbar.The as-prepared SnSe thin films are characterized for their structural,optical and electrical properties by various experimental techniques.The p-type conductivity,near-optimum direct band gap,high absorption coefficient and good photosensitivity of the SnSe thin film indicate its suitability for photovoltaic applications.The optical constants,loss factor,quality factor and optical conductivity of the films are evaluated.The results of Hall and thermoelectric power measurements are correlated to determine the density of states,Fermi energy and effective mass of carriers and are obtained as 2.8×10^(17)cm^(-3),0.03 eV and 0.05m_0 respectively.The high Seebeck coefficient ≈ 7863 μV/K,reasonably good power factor ≈7.2×10^(-4) W/(m·K^2) and thermoelectric figure of merit ≈1.2 observed at 42 K suggests that,on further work,the prepared SnSe thin films can also be considered as a possible candidate for cryogenic thermoelectric applications.K S Urmila T A Namitha J Rajani R R Philip B Pradeep 2016Journal of Semiconductors2016,37,9:2
9Maximum Power Enhancement Under Partial Shadings Using a Modified Sudoku Reconfiguration显示文摘Partial shading is one of the important factors in reducing maximum power generation from PV(Photovoltaic)arrays.Maximum power generation can be improved by selecting a PV array through a Total-Cross-Tied(T-C-T)connection.However,maximum power generated from T-C-T can still be improved by distribution of shading over various rows.Due to distribution of shading,the current entering the node increases and results in improved maximum power generation.This can be done effectively by using Sudoku reconfiguration techniques.These techniques are economical,since they don’t require any sensors and switching networks.This technique only changes the physical location of the PV panel but the electrical connection between the panels remains the same.This paper proposes a Modified Sudoku reconfiguration pattern which enhances the maximum power from the T-C-T connected PV array.Furthermore,the theoretical calculation of row current and power output have been done for existing and proposed topologies under various shading patterns.The performance of the proposed pattern has been analyzed and compared using specifications,such as Global Maximum Power(GMP),Fill Factor(FF),mismatch losses,and efficiency.From the results,it can be concluded that the Modified Sudoku reconfiguration enhances the GMP under all shading conditions.Kandipati Rajani Tejavathu Ramesh 2021CSEE Journal of Power and Energy Systems2021,7,6:2
