| 1 | Capacity of Old Trees to Respond to Environmental Change显示文摘Atmospheric carbon dioxide CO2 has increased dramatically within the current life spans of long-lived trees and old forests.Consider that a 500-year-old tree in the early twenty-first century has spent 70% of its life growing under pre-industrial levels of CO2,which were 30% lower than current levels.Here we address the question of whether old trees have already responded to the rapid rise in CO2 occurring over the past 150 years.In spite of limited data,aging trees have been shown to possess a substantial capacity for increased net growth after a period of post-maturity growth decline.Observations of renewed growth and physiological function in old trees have,in some instances,coincided with Industrial Age increases in key environmental resources,including CO2,suggesting the potential for continued growth in old trees as a function of continued global climate change. | Nathan G.Phillips Thomas N.Buckley David T.Tissue | 2008 | Journal of Integrative Plant Biology2008,50,11: | 2 |
| 2 | An optimized protocol for stepwise optimization of real-time RT-PCR analysis显示文摘Computational tool-assisted primer design for real-time reverse transcription(RT)PCR(qPCR)analysis largely ignores the sequence similarities between sequences of homologous genes in a plant genome.It can lead to false confidence in the quality of the designed primers,which sometimes results in skipping the optimization steps for qPCR.However,the optimization of qPCR parameters plays an essential role in the efficiency,specificity,and sensitivity of each gene’s primers.Here,we proposed an optimized approach to sequentially optimizing primer sequences,annealing temperatures,primer concentrations,and cDNA concentration range for each reference(and target)gene.Our approach started with a sequence-specific primer design that should be based on the single-nucleotide polymorphisms(SNPs)present in all the homologous sequences for each of the reference(and target)genes under study.By combining the efficiency calibrated and standard curve methods with the 2−ΔΔCt method,the standard cDNA concentration curve with a logarithmic scale was obtained for each primer pair for each gene.As a result,an R 2≥0.9999 and the efficiency(E)=100±5% should be achieved for the best primer pair of each gene,which serve as the prerequisite for using the 2^(−ΔΔCt) method for data analysis.We applied our newly developed approach to identify the best reference genes in different tissues and at various inflorescence developmental stages of Tripidium ravennae,an ornamental and biomass grass,and validated their utility under varying abiotic stress conditions.We also applied this approach to test the expression stability of six reference genes in soybean under biotic stress treatment with Xanthomonas axonopodis pv.glycines(Xag).Thus,these case studies demonstrated the effectiveness of our optimized protocol for qPCR analysis. | Fangzhou Zhao Nathan A.Maren Pawel Z.Kosentka Ying-Yu Liao Hongyan Lu James R.Duduit Debao Huang Hamid Ashrafi Tuanjie Zhao Alejandra I.Huerta Thomas G.Ranney Wusheng Liu | 2021 | Horticulture Research2021,8,1: | 2 |
| 3 | Appendectomy and Clostridium difficile colitis:Relationships revealed by clinical observations and immunology显示文摘Advances in understanding the interaction between the human immune system and the microbiome have led to an improved understanding of the function of the vermiform appendix as a safe-house for beneficial bacteria in the colon.These advances have been made despite long standing clinical observations that the appendectomy is a safe and effective procedure.However,more recent clinical data show that an appendectomy puts patients at increased risk for recurrent Clostridium difficile(C.difficile)-associated colitis,and probably other diseases associated with an altered microbiome.At the same time,appendectomy does not apparently put patients at risk for an initial onset of C.difficile-associated colitis.These clinical observations point toward the idea that the vermiform appendix might not effectively protect the microbiome in the face of broad spectrum antibiotics,the use of which precedes the initial onset of C.difficile-associated colitis.Further,these observations point to the idea that historically important threats to the microbiome such as infectious gastrointestinal pathogens have been supplanted by other threats,particularly the use of broad spectrum antibiotics. | Nathan L Sanders R Randal Bollinger Ryan Lee Steven Thomas William Parker | 2013 | World Journal of Gastroenterology2013,19,34: | 2 |

| 14 | Maize Carbohydrate Partitioning Defective33 Encodes an MCTP Protein and Functions in Sucrose Export from Leaves显示文摘To sustain plant growth,development,and crop yield,sucrose must be transported from leaves to distant parts of the plant,such as seeds and roots.To identify genes that regulate sucrose accumulation and transport in maize(Zea mays),we isolated carbo/iydrafe part/f/ofi/ngf defecf/Ve33(cpd33),a recessive mutant that accumulated excess starch and soluble sugars in mature leaves.The cpd33 mutants also exhibited chlorosis in the leaf blades,greatly diminished plant growth,and reduced fertility.Cpd33 encodes a protein containing multiple C2 domains and transmembrane regions.Subcellular localization experiments showed the CPD33 protein localized to plasmodesmata(PD),the plasma membrane,and the endoplasmic reticulum.We also found that a loss-of-function mutant of the CPD33 homolog in Arabidopsis,QUIRKY,had a similar carbohydrate hyperaccumulation phenotype.Radioactively labeled sucrose transport assays showed that sucrose export was significantly lower in cpd33 mutant leaves relative to wild-type leaves.However,PD transport in the adaxial-abaxial direction was unaffected in cpd33 mutant leaves.Intriguingly,transmission electron microscopy revealed fewer PD at the companion cell-sieve element interface in mutant phloem tissue,providing a possible explanation for the reduced sucrose export in mutant leaves.Collectively,our results suggest that CPD33 functions to promote symplastic transport into sieve elements. | Thu M.Tran Tyler J.McCubbin Saadia Bihmidine Benjamin T.Julius R.Frank Baker Martin Schauflinger Clifford Weil Nathan Springer Paul Chomet Ruth Wagner Jeff Woessner Karen Grote Jeanette Peevers Thomas L.Slewinski David M.Braun | 2019 | Molecular Plant2019,12,9: | 1 |
