| 1 | Significant decrease in prevalence ofHelicobacter pylori in the Czech Republic显示文摘AIM:To study possible decrease in prevalence of Helicobacter pylori(H.pylori) infection in the Czech Republic within a 10-year period.METHODS:A total of 22 centres entered the study.The catchment areas of these centres covered cities and towns with more than 20 000 inhabitants,smaller towns(≤ 20 000 inhabitants) with surrounding villages and rural areas,and were spread over the whole country,corresponding well to the geographical distribution of the Czech population.A total of 1 837 subjects(aged 5-98 years) took part in the study,randomly selected out of 38 147 people from the general population.H.pylori infection was investigated by means of a 13 C-urea breath test.Breath samples in duplicates were analysed using isotope ratio mass spectrometry.The cut-off point was 3.5.Social and demographic characteristics were based on data from self-completed questionnaires.RESULTS:The overall prevalence of H.pylori infection was 23.5%(430/1826),and 4.8%(20/420) in children aged 15 or less.There was no statistically significant difference in prevalence between males(24.3%;208/857) and females(22.9%,222/969,P = 0.494).H.pylori infection was strongly associated with higher age,among subjects aged 55+ years,prevalence of H.pylori infection was 39.8%(252/633,P < 0.001).The highest prevalence of H.pylori infection was found among persons aged 55-64 years(43.9%,97/221) and 75+ years(37.9%,58/153).Among study subjects aged 15+ years,prevalence of H.pylori infection was significantly increased in those with lowest education(odds risk 3.19,95% CI 1.87-5.47).Compared to never married(14.1%),the prevalence of H.pylori infection was statistically significantly higher among married(35.4%,246/694,P < 0.001),divorced(36.8%,49/133,P < 0.001) and widowed study subjects(40.2%,45/112,P < 0.001),both in minimally and fully adjusted analysis.There was no significant difference in the prevalence of H.pylori infection between married and widowed subjects(35.4%,246/694 vs 40.2%,45/112,P = 0.389).There was little variation in smoking prevalence across categories of smoking and there was no evidence of an increased risk ofH.pylori infection among current or past smokers in our data(odds risk 1.04 with 95% CI 0.78-1.40 for current smokers;odds ratio 0.83 with 95% CI 0.60-1.16 for former smokers).The current prevalence of H.pylori in 2011 was significantly lower compared to the prevalence reported from identical geographical areas in 2001(23.5%vs 41.7%,P < 0.001).CONCLUSION:The overall prevalence of H.pylori infection in the general population has fallen substantially in the Czech Republic over the past 10 years. | Jan Bure Marcela Kopáová Ilona Koupil Bohumil Seifert Miluka kodová Fendrichová Jana pirková Viktor Voíek Stanislav Rejchrt TomáDouda Norbert Král Ilja Tachecí | 2012 | World Journal of Gastroenterology2012,18,32: | 8 |
| 2 | Reallocation of foliicolous species of the genus Strigula into six genera(lichenized Ascomycota,Dothideomycetes,Strigulaceae)显示文摘Strigula sensu lato has been previously defined based on phenotype characters as a rather broad genus including tropical to temperate species growing on a wide array of substrata.In this study,based on a multilocus phylogenetic approach,we show that foliicolous species form six well-delimited clades that correlate with diagnostic phenotype features,including thallus morphology,carbonization of the involucrellum and excipulum,ascospore dimensions,and type of macroconidia.Given the topology,with five of the six clades emerging on long stem branches,and the strong phenotypical differentiation between the clades,we recognize these at the genus level,making mostly use of previously established genus names.Four genera,namely Phylloporis,Puiggariella,Raciborskiella,and Racoplaca,are resurrected for the S.phyllogena,S.nemathora,S.janeirensis,and S.subtilissima groups,respectively,whereas one new genus,Serusiauxiella gen.nov.,is introduced for a novel lineage with peculiar macroconidia.The only sequenced non-foliicolous species,S.jamesii,is not closely related to these six foliicolous lineages but clusters with Flavobathelium and Phyllobathelium,revealing it as an additional undescribed genus-level lineage being treated elsewhere.Within the new genus Serusiauxiella,three new species are described:Serusiauxiella filifera sp.nov.,S.flagellata sp.nov.,and S.sinensis sp.nov.In addition,ten new combinations are proposed:Phylloporis austropunctata comb.nov.,P.radiata comb.nov.,P.vulgaris comb.nov.,Puiggariella confluens comb.et stat.nov.,P.nemathora comb.nov.,P.nigrocincta comb.nov.,Racoplaca maculata comb.nov.,R.melanobapha comb.nov.,R.transversoundulata,and R.tremens comb.nov.We also report on a peculiar,previously unrecognized growth behaviour of the macroconidial appendages in Strigula s.lat. | Shu‑Hua Jiang Robert Lücking Amanda Barreto Xavier‑Leite Marcela E.S.Cáceres AndréAptroot Carlos Vinas Portilla Jiang‑Chun Wei | 2020 | Fungal Diversity2020,,3: | 5 |

| 6 | HP0953-hypothetical virulence factor overexpresion and localization during Helicobacter pylori infection of gastric epithelium显示文摘BACKGROUND The high prevalence and persistence of Helicobacter pylori(H. pylori) infection, as well as the diversity of pathologies related to it, suggest that the virulence factors used by this microorganism are varied. Moreover, as its proteome contains 340hypothetical proteins, it is important to investigate them to completely understand the mechanisms of its virulence and survival. We have previously reported that the hypothetical protein HP0953 is overexpressed during the first hours of adhesion to inert surfaces, under stress conditions, suggesting its role in the environmental survival of this bacterium and perhaps as a virulence factor.AIM To investigate the expression and localization of HP0953 during adhesion to an inert surface and against gastric(AGS) cells.METHODS Expression analysis was performed for HP0953 during H. pylori adhesion. HP0953 expression at 0,3, 12, 24, and 48 h was evaluated and compared using the Kruskal-Wallis equality-of-populations rank test. Recombinant protein was produced and used to obtain polyclonal antibodies for immunolocalization. Immunogold technique was performed on bacterial sections during adherence to inert surfaces and AGS cells, which was analyzed by transmission electron microscopy. HP0953 protein sequence was analyzed to predict the presence of a signal peptide and transmembrane helices, both provided by the ExPASy platform, and using the GLYCOPP platform for glycosylation sites. Different programs, via, I-TASSER, RaptorX, and HHalign-Kbest, were used to perform three-dimensional modeling.RESULTS HP0953 exhibited its maximum expression at 12 h of infection in gastric epithelium cells.Immunogold technique revealed HP0953 localization in the cytoplasm and accumulation in some peripheral areas of the bacterial body, with greater expression when it is close to AGS cells.Bioinformatics analysis revealed the presence of a signal peptide that interacts with the transmembrane region and then allows the release of the protein to the external environment. The programs also showed a similarity with the Tip-alpha protein of H. pylori. Tip-alpha is an exotoxin that penetrates cells and induces tumor necrosis factor alpha production, and HP0953 could have a similar function as posttranslational modification sites were found;modifications in turn require enzymes located in eukaryotic cells.
