Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further, there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients. Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer. Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks. The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response, time to progression, disease control rate, and safety. Results:Of 179 patients screened,125 were eligible and received RC48 treatment. The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively. The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety, suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
A new solution for topology optimization problems with multiple loads:The guide-weight method显示文摘The guide-weight method is introduced to solve two kinds of topology optimization problems with multiple loads in this paper. The guide-weight method and its Lagrange multipliers' solution methods are presented first, and the Lagrange multipliers' soution method of problems with multiple constraints is improved by the dual method. Then the iterative formulas of the guide-weight method for topology optimization problems of minimum compliance and minimum weight are derived and coresponding numerical examples are calculated. The results of the examples exhibits that when the guide-weight method is used to solve topology optimization problems with multiple loads,it works very well with simple iterative formulas, and has fast convergence and good solution. After comparison with the results calculated by the SCP method in Ansys, one can conclude that the guide-weight method is an effective method and it provides a new way for solving topology optimization problems.LIU XinJun LI ZhiDong CHEN Xiang 2011Science China(Technological Sciences)2011,54,6:22
Impacts of changing scale on Getis-Ord Gi* hotspots of CPUE:a case study of the neon flying squid (Ommastrephes bartramii)in the northwest Pacific Ocean显示文摘We examined the scale impacts on spatial hot and cold spots of CPUE for Ommastrephes bartramii in the northwest Pacific Ocean. The original fishery data were tessellated to 18 spatial scales from 5′×5′ to 90′×90′ with a scale interval of 5′ to identify the local clusters. The changes in location, boundaries, and statistics regarding the Getis-Ord Gi* hot and cold spots in response to the spatial scales were analyzed in detail. Several statistics including Min, mean, Max, SD, CV, skewness, kurtosis, first quartile(Q1), median, third quartile(Q3), area and centroid were calculated for spatial hot and cold spots. Scaling impacts were examined for the selected statistics using linear, logarithmic, exponential, power law and polynomial functions. Clear scaling relations were identified for Max, SD and kurtosis for both hot and cold spots. For the remaining statistics, either a difference of scale impacts was found between the two clusters, or no clear scaling relation was identified. Spatial scales coarser than 30′ are not recommended to identify the local spatial patterns of fisheries because the boundary and locations of hot and cold spots at a coarser scale are significantly different from those at the original scale.FENG Yongjiu CHEN Xinjun GAO Feng LIU Yang 2018Acta Oceanologica Sinica2018,37,5:23
Type Synthesis of 4-DOF Parallel Kinematic Mechanisms Based on Grassmann Line Geometry and Atlas Method显示文摘Many methods are proposed to deal with the type synthesis of parallel kinematic mechanisms(PKMs),but most of them are less intuitive to some extent. Thus,to propose a concise and intuitive type synthesis method for engineering application is a very challenging issue, which should be further studied in the field.Grassmann line geometry, which can investigate the dimensions of spatial line-clusters in a concise way,is taken as the mathematic foundation.Atlas method is introduced to visually describe the degrees of freedom(DOFs)and constraints of a mechanism, and the dual rule is brought in to realize the mutual conversion of the freedom-space and constraint-space.Consequently,a systematic method based on Grassmann line geometry and Atlas method is generated and the entire type synthesis process is presented.Three type 4-DOF PKMs,i.e.,1T3R,2T2R and 3T1R(T:translational DOF;R:rotational DOF),are classified according to the different combinations of the translational DOFs and rotational DOFs.The type synthesis of4-DOF PKMs is carried out and the possible configurations are thoroughly investigated. Some new PKMs with useful functions are generated during this procedure. The type synthesis method based on Grassmann line geometry and Atlas method is intuitive and concise, and can reduce the complexity of the PKMs’type synthesis. Moreover, this method can provide theoretical guidance for other PKMs’type synthesis and engineering application.A novel type synthesis method is proposed, which solves the existing methods’problems in terms of complicated, not intuitive and unsuitable for practical application.XIE Fugui LI Tiemin LIU Xinjun 2013Chinese Journal of Mechanical Engineering2013,26,6:21
Stereotactic aspiration for hypertensive intracerebral haemorrhage in a Chinese population:a retrospective cohort study显示文摘Objective We aimed to compare the therapeutic effects of stereotactic aspiration and best medical management in patients who developed supratentorial hypertensive intracerebral haemorrhage(HICH)with a volume of haemorrhage between 20 and 40 mL.Methods The clinical data of 220 patients with supratentorial HICH with a volume between 20 and 40 mL were retrospectively analysed.Among them,142 received stereotactic aspiration surgery(stereotactic aspiration group)and 78 received best medical management(conservative group). All were followed up for 6 months.Multivariate logistic regression and Kaplan-Meier survival curves were used to compare the outcome between the two groups.results The rebleeding rate was lower in the group that had stereotactic aspiration when compared with the group with medical treatment(6[4.2%]vs 9[11.5%],χ2=4.364,p=0.037).After 6 months, although the mortality rate did not differ significantly between the two groups(8 cases[5.6%]vs 10 cases[12.8%],χ2=3.461,p=0.063),the rate of a favourable outcome was higher in the group who received stereotactic aspiration(χ2=15.870,p=0.000).Logistic regression identified that medical treatment(OR=1.64,p=0.000)was an independent risk factor for an unfavourable outcome. The Kaplan-Meier curves indicated that the median favourable outcome time in the stereotactic aspiration group was 59.5 days compared with that in the medically treated group(87.0 days). The log-rank test indicated that the prognosis at 6 months was better for those treated with stereotactic haematoma aspiration(χ2=29.866,p=0.000).However,the 6-month survival rate was similar between the two groups(χ2=3.253,p=0.068).Conclusions Stereotactic haematoma aspiration significantly improved the quality of life, although did not effectively reduce the rate of mortality. When selected appropriately, patients with HICH may benefit from this type of surgical intervention.Xuyang Zhang Shaolong Zhou Qiang Zhang Xudong Fu Yuehui Wu Jiasheng Liu Bo Liang Zhuo Yang Xinjun Wang 2019Stroke & Vascular Neurology2019,4,1:19
Error Modeling and Sensitivity Analysis of a Parallel Robot with SCARA(Selective Compliance Assembly Robot Arm) Motions显示文摘Parallel robots with SCARA(selective compliance assembly robot arm) motions are utilized widely in the field of high speed pick-and-place manipulation. Error modeling for these robots generally simplifies the parallelogram structures included by the robots as a link. As the established error model fails to reflect the error feature of the parallelogram structures, the effect of accuracy design and kinematic calibration based on the error model come to be undermined. An error modeling methodology is proposed to establish an error model of parallel robots with parallelogram structures. The error model can embody the geometric errors of all joints, including the joints of parallelogram structures. Thus it can contain more exhaustively the factors that reduce the accuracy of the robot. Based on the error model and some sensitivity indices defined in the sense of statistics, sensitivity analysis is carried out. Accordingly, some atlases are depicted to express each geometric error's influence on the moving platform's pose errors. From these atlases, the geometric errors that have greater impact on the accuracy of the moving platform are identified, and some sensitive areas where the pose errors of the moving platform are extremely sensitive to the geometric errors are also figured out. By taking into account the error factors which are generally neglected in all existing modeling methods, the proposed modeling method can thoroughly disclose the process of error transmission and enhance the efficacy of accuracy design and calibration.CHEN Yuzhen XIE Fugui LIU Xinjun ZHOU Yanhua 2014Chinese Journal of Mechanical Engineering2014,27,4:17
Optimal design of a new spatial 3-DOF parallel robot with respect to a frame-free index显示文摘Optimal design is one of the most important issues in robots. Since the very beginning, the concepts of the Jacobian matrix, manipulability and condition number, which are used successfully in the field of serial robots, have been applied to parallel robots. Unlike serial robots, parallel robots are good for motion/force transmission. Their performance evaluation and design should be correspondingly dif- ferent. This paper is an attempt to optimally design a novel spatial three-degree-of-freedom (3-DOF) parallel robot by using the concept of motion/force transmission. Accordingly, three indices are defined. The suggested indices are independent of any coordinate frame and could be applied to the analysis and design of a parallel robot whose singularities can be identified wholly by using the relative angle between the output and adjacent links, and by using the relative angle between the input and adjacent links.JinSong Wang XinJun Liu Chao Wu 2009Science China(Technological Sciences)2009,52,4:12
Development and validation of an endoscopic images-based deep learning model for detection with nasopharyngeal malignancies显示文摘Background:Due to the occult anatomic location of the nasopharynx and frequent presence of adenoid hyperpla-sia,the positive rate for malignancy identification during biopsy is low, thus leading to delayed or missed diagnosis for nasopharyngeal malignancies upon initial attempt. Here,we aimed to develop an artificial intelligence tool to detect nasopharyngeal malignancies under endoscopic examination based on deep learning. Methods:An endoscopic images-based nasopharyngeal malignancy detection model(eNPM-DM)consisting of a fully convolutional network based on the inception architecture was developed and fine-tuned using separate training and validation sets for both classification and segmentation.Briefly,a total of 28,966 qualified images were collected.Among these images,27,536 biopsy-proven images from 7951 individuals obtained from January 1st,2008,to December 31st,2016,were split into the training, validation and test sets at a ratio of 7:1:2 using simple randomiza-tion.Additionally,1430 images obtained from January 1st,2017,to March 31st,2017,were used as a prospective test set to compare the performance of the established model against oncologist evaluation. The dice similarity coef-ficient(DSC)was used to evaluate the efficiency of eNPM-DM in automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images,by comparing automatic segmentation with manual segmenta-tion performed by the experts. Results:All images were histopathologically confirmed, and included 5713(19.7%)normal control,19,107(66.0%)nasopharyngeal carcinoma(NPC),335(1.2%)NPC and 3811(13.2%)benign diseases. The eNPM-DM attained an overall accuracy of 88.7%(95%confidence interval(CI)87.8%-89.5%)in detecting malignancies in the test set.In the prospective comparison phase,eNPM-DM outperformed the experts:the overall accuracy was 88.0%(95%CI 86.1%-89.6%)vs.80.5%(95%CI 77.0%-84.0%).The eNPM-DM required less time(40 s vs.110.0±5.8 min)and exhibited encouraging performance in automatic segmentation of nasopharyngeal malignant area from the background, with an average DSC of 0.78±0.24 and 0.75±0.26 in the test and prospective test sets, respectively. Conclusions:The eNPM-DM outperformed oncologist evaluation in diagnostic classification of nasopharyngeal mass into benign versus malignant, and realized automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images.Chaofeng Li Bingzhong Jing Liangru Ke Bin Li Weixiong Xia Caisheng He Chaonan Qian Chong Zhao Haiqiang Mai Mingyuan Chen Kajia Cao Haoyuan Mo Ling Guo Qiuyan Chen Linquan Tang Wenze Qiu Yahui Yu Hu Liang Xinjun Huang Guoying Liu Wangzhong Li Lin Wang Rui Sun Xiong Zou Shanshan Guo Peiyu Huang Donghua Luo Fang Qiu Yishan Wu Yijun Hua Kuiyuan Liu Shuhui Lv Jingjing Miao Yanqun Xiang Ying Sun Xiang Guo Xing Lv 2018Cancer Communications2018,38,1:9
Step kinematic calibration of a 3-DOF planar parallel kinematic machine tool显示文摘This paper presents a novel step kinematic calibration method for a 3 degree-of-freedom(DOF) planar parallel kinematic machine tool, based on the minimal linear combinations(MLCs) of error parameters. The method using mapping of linear combinations of parameters in error transfer multi-parameters coupling system changes the modeling, identification and error compensation of geometric parameters in the general kinematic calibration into those of linear combinations of parameters.By using the four theorems of the MLCs,the sets of the MLCs that are respectively related to the relative precision and absolute precision are determined. All simple and feasible measurement methods in practice are given, and identification analysis of the set of the MLCs for each measurement is carried out.According to the identification analysis results,a step calibration including step measurement, step identification and step error compensation is determined by taking into account both measurement costs and observability. The experiment shows that the proposed method has the following merits:(1) the parameter errors that cannot influence precision are completely avoided;(2) it reflects the mapping of linear combinations of parameters more accurately and enhances the precision of identification;and(3) the method is robust, efficient and effective,so that the errors in position and orientation are kept at the same order of the measurement noise.Due to these merits, the present method is attractive for the 3-DOF planar parallel kinematic machine tool and can be also applied to other parallel kinematic machine tools with weakly nonlinear kinematics.CHANG Peng,WANG JinSong,LI TieMin,LIU XinJun & GUAN LiWen Department of Precision Instruments and Mechanology,Tsinghua University,Beijing 100084,China 2008Science China(Technological Sciences)2008,51,12:9
Relationship Between Beak Morphological Variables and Body Size and Mantle Length of Male and Female Argentine Shortfin Squid(Illex argentinus)显示文摘Beak of cephalopod is an important hard tissue.Understanding the morphology of beak can yield critical information on the role of cephalopods in the ecosystem. The south patagonic stock of the Argentine shortfin squid,Illex argentinus,is not only one of the most important fishing targets, but also one of the most important species in the marine eco-system of the southwest Atlantic.A total of 430 samples of I.argentinus, including 229 females 103-346mm in mantle length(ML) and 201 males 140-298mm in ML,were collected from the area off the Exclusive Economic Zone of Argentinean waters by Chinese squid jigging vessels during February to May 2007.The morphology of their beaks was evaluated. The relationships between beak morphological variables and ML differed significantly among males and females. They could be best described by logarithmic functions for females and linear functions for males except for upper wing length(UWL) and lower rostrum length(LRL),which followed exponential functions in their relationships with ML.The results showed the sexual dimorphism in the relationship between ML and beak morphology for the south patagonic stock of I.argentinus.However,no significant difference was found between males and females in the relationships of beak morphological variables(except for UWL) versus body weight(BW),suggesting that the relationship between beak morphological variables and BW can be used for estimating the biomass consumed by their predators.CHEN Xinjun LU Huajie LIU Bilin FANG Zhou 2012Journal of Ocean University of China2012,11,4:7
Optimal Design of a Main Driving Mechanism for Servo Punch Press Based on Performance Atlases显示文摘The servomotor drive turret punch press is attracting more attentions and being developed more intensively due to the advantages of high speed, high accuracy, high flexibility, high productivity, low noise, cleaning and energy saving. To effectively improve the performance and lower the cost,it is necessary to develop new mechanisms and establish corresponding optimal design method with uniform performance indices.A new patented main driving mechanism and a new optimal design method are proposed. In the optimal design, the performance indices,i.e.,the local motion/force transmission indices ITI,OTI,good transmission workspace good transmission workspace(GTW) and the global transmission indices GTIs are defined. The non-dimensional normalization method is used to get all feasible solutions in dimensional synthesis.Thereafter, the performance atlases, which can present all possible design solutions, are depicted. As a result, the feasible solution of the mechanism with good motion/force transmission performance is obtained. And the solution can be flexibly adjusted by designer according to the practical design requirements. The proposed mechanism is original, and the presented design method provides a feasible solution to the optimal design of the main driving mechanism for servo punch press.ZHOU Yanhua XIE Fugui LIU Xinjun 2013Chinese Journal of Mechanical Engineering2013,26,5:7
Seasonal Variation of Biochemical Components in Clam (Saxidomuspurpuratus Sowerby 1852) in Relation to Its Reproductive Cycle and the Environmental Condition of Sanggou Bay, China显示文摘Seasonal variation of biochemical components in clam(Saxidomus purpuratus Sowerby 1852) was investigated from March 2012 to February 2013 in relation to environmental condition of Sanggou Bay and the reproductive cycle of clam.According to the histological analysis, the reproductive cycle of S.purpuratus includes two distinctive phases:a total spent and inactive stage from November to January, and a gametogenesis stage, including ripeness and spawning, during the rest of the year.Gametes were generated at a low temperature(2.1℃) in February.Spawning took place once a year from June to October. The massive spawning occurred in August when the highest water temperature and chlorophyll a level could be observed. The key biochemical components(glycogen, protein and lipid) in five tissues(gonad, foot, mantle, siphon and adductor muscle) were analyzed. The glycogen content was high before gametogenesis, and decreased significantly during the gonad development in the gonad, mantle and foot of both females and males, suggesting that glycogen was an important energy source for gonad development. The protein and lipid contents increased in the ovary during the gonad development, demonstrating that they are the major organic components of oocytes. The lipid and protein contents decreased in the testis, implying that they can provide energy and material for spermatogenesis. The results also showed that protein stored in the mantle and foot could support the reproduction after the glycogen was depleted.BI Jinhong LI Qi ZHANG Xinjun ZHANG Zhixin TIAN Jinling XU Yushan LIU Wenguang 2016Journal of Ocean University of China2016,15,2:6
Design and experimental study of the SPKM165, a five-axis serial-parallel kinematic milling machine显示文摘A five-axis serial-parallel kinematic milling machine, the SPKM 165, is introduced. This machine consists of a three-degree of-freedom parallel module and a two-degree-of-freedom serial table. The SPKM 165 is capable of five-face machining. A discussion of the inverse kinematics of the five-axis control is provided. A dimensional synthesis procedure is presented in terms of motion/force transmissibility. Finite-element analysis was used to evaluate the stiffness of a CAD model before the machine was manufactured. Kinematic calibration was implemented to improve the accuracy of the end effector. 14Modeling Monthly Spatial Distribution of Ommastrephes bartramii CPUE in the Northwest Pacific and Its Spatially Nonstationary Relationships with the Marine Environment显示文摘There are substantial spatial variations in the relationships between catch-per-unit-effort(CPUE) and oceanographic conditions with respect to pelagic species. This study examines the monthly spatiotemporal distribution of CPUE of the neon flying squid, Ommastrephes bartramii, in the Northwest Pacific from July to November during 2004–2013, and analyzes the relationships with oceanographic conditions using a generalized additive model(GAM) and geographically weighted regression(GWR) model. The results show that most of the squids were harvested in waters with sea surface temperature(SST) between 7.6 and 24.6℃, chlorophyll-a(Chl-a) concentration below 1.0 mgm^(-3), sea surface salinity(SSS) between 32.7 and 34.6, and sea surface height(SSH) between-12.8 and 28.4 cm. The monthly spatial distribution patterns of O. bartramii predicted using GAM and GWR models are similar to observed patterns for all months. There are notable variations in the local coefficients of GWR, indicating the presence of spatial non-stationarity in the relationship between O. bartramii CPUE and oceanographic conditions. The statistical results show that there were nearly equal positive and negative coefficients for Chl-a, more positive than negative coefficients for SST, and more negative than positive coefficients for SSS and SSH. The overall accuracies of the hot spots predicted by GWR exceed 60%(except for October), indicating a good performance of this model and its improvement over GAM. Our study provides a better understanding of the ecological dynamics of O. bartramii CPUE and makes it possible to use GWR to study the spatially nonstationary characteristics of other pelagic species.FENG Yongjiu LIU Yang CHEN Xinjun 2018Journal of Ocean University of China2018,17,3:5
Optimal Design and Force Control of a Nine-Cable-Driven Parallel Mechanism for Lunar Takeoff Simulation显示文摘Traditional simulation methods are unable to meet the requirements of lunar takeo simulations, such as high force output precision, low cost, and repeated use. Considering that cable-driven parallel mechanisms have the advantages of high payload to weight ratio, potentially large workspace, and high-speed motion, these mechanisms have the potential to be used for lunar takeo simulations. Thus, this paper presents a parallel mechanism driven by nine cables. The purpose of this study is to optimize the dimensions of the cable-driven parallel mechanism to meet dynamic workspace requirements under cable tension constraints. The dynamic workspace requirements are derived from the kinematical function requests of the lunar takeo simulation equipment. Experimental design and response surface methods are adopted for building the surrogate mathematical model linking the optimal variables and the optimization indices. A set of dimensional parameters are determined by analyzing the surrogate mathematical model. The volume of the dynamic workspace increased by 46% after optimization. Besides, a force control method is proposed for calculating output vector and sinusoidal forces. A force control loop is introduced into the traditional position control loop to adjust the cable force precisely, while controlling the cable length. The e ectiveness of the proposed control method is verified through experiments. A 5% vector output accuracy and 12 Hz undulation force output can be realized. This paper proposes a cable-driven parallel mechanism which can be used for lunar takeo simulation.Wangmin Yi Yu Zheng Weifang Wang XiaoqiangTang Xinjun Liu Fanwei Meng 2019Chinese Journal of Mechanical Engineering2019,32,4:5
Fishery Biology of Jumbo Flying Squid Dosidicus gigas off Costa Rica Dome显示文摘The jumbo flying squid(Dosidicus gigas) population was surveyed with the help of Chinese squid jigging vessels off the Costa Rica Dome(4°–11°N, 90°–100°W) in 2009 and 2010. The daily catch of D. gigas in the two survey cruises ranged from 0 to 5.5 t and was mostly obtained from the areas bounded by 6°–9°N and 91°–94°W and by 6°30′–7°30′N and 96°–97°W. The sea surface temperature in the areas yielding the most catch ranged from 27.5 to 29℃. The sex ratio of the total catch was 3.75:1(female: male). The mantle length of the squid ranged from 211 to 355 mm(male) and from 204 to 429 mm(female) with an average of 297.9 and 306.7 mm, respectively. In the relationship of the mantle length(mm) and body weight(g) of the squid, there was no significant difference between sexes. The female and male were at a similar maturity, and most individuals are maturing or have matured with a few females being spent. The size(mantle length) and age at the first sexual maturity were 297 mm and 195 d in females, and less than 211 mm and 130 d in males, respectively. Most of the sampled stomachs(70.6%) had no food remains. The major preys of the squids were fish, cephalopods and crustaceans, with the most abundant Myctophum orientale and D. gigas. The preys in more than 65% of the non-empty sampled stomachs evidenced the cannibalism of D. gigas. The results improved current understanding of the fishery biology of D. gigas off the Costa Rica Dome, which may facilitate the assessment and management of relative fishery resources.CHEN Xinjun LI Jianghua LIU Bilin LI Gang LU Huajie 2014Journal of Ocean University of China2014,13,3:4
Identification of Three Common Loliginidae Squid Species in the South China Sea by Analyzing Hard Tissues with Geometric Outline Method显示文摘The hard tissues of squid can provide important information for species identification. In this study, we used statolith and beak to identify three squid species including Uroteuthis duvaucelii, Loliolus beka, and U. edulis in the South China Sea. Because of the highly overlapping habitat and similar body morphology of the three squid species, we explored four different ways to identify them, by using statolith, upper beak, lower beak and a combination of statolith and beak. An outline geometric morphometric method and stepwise discriminant analysis were used to evaluate the most suitable method for the identification. We found that the combination of statolith and beak had the highest cross validation rate that was 75.0%, 87.5% and 88.7% for U. duvaucelii, L. beka and U. edulis, respectively. Using two beaks had similar results and the lowest cross validation rate was 60.0%, 50.0%, and 73.7% for the upper beak, 46.9%, 58.5% and 75.3% for the lower beak of U. duvaucelii, L. beka and U. edulis, respectively. Analyzing with the statolith had moderate cross validation which was 72.2%, 80.0%, and 87.7% for U. duvaucelii, L. beka and U. edulis, respectively. From the results it is suggested when the entire body of a squid is available, a combination of statolith and beak should be used for the identification. When only one hard tissue is available, species identification can be subjected to large errors.JIN Yue LIU Bilin LI Jianhua CHEN Xinjun 2017Journal of Ocean University of China2017,16,5:4

Using Different Standardized Methods for Species Identification:A Case Study Using Beaks from Three Ommastrephid Species显示文摘The cephalopod beak is a vital hard structure with a stable configuration and has been widely used for the identification of cephalopod species. This study was conducted to determine the best standardization method for identifying different species by measuring 12 morphological variables of the beaks of Illex argentinus, Ommastrephes bartramii, and Dosidicus gigas that were collected by Chinese jigging vessels. To remove the effects of size, these morphometric variables were standardized using three methods. The average ratios of the upper beak morphological variables and upper crest length of O. bartramii and D. gigas were found to be greater than those of I. argentinus. However, for lower beaks, only the average of LRL(lower rostrum length)/LCL(lower crest length), LRW(lower rostrum width)/LCL, and LLWL(lower lateral wall length)/LCL of O. bartramii and D. gigas were greater than those of I. argentinus. The ratios of beak morphological variables and crest length were found to be all significantly different among the three species(P < 0.001). Among the three standardization methods, the correct classification rate of stepwise discriminant analysis(SDA) was the highest using the ratios of beak morphological variables and crest length. Compared with hood length, the correct classification rate was slightly higher when using beak variables standardized by crest length using an allometric model. The correct classification rate of the lower beak was also found to be greater than that of the upper beak. This study indicates that the ratios of beak morphological variables to crest length could be used for interspecies and intraspecies identification. Meanwhile, the lower beak variables were found to be more effective than upper beak variables in classifying beaks found in the stomachs of predators.HU Guanyu FANG Zhou LIU Bilin CHEN Xinjun STAPLES Kevin CHEN Yong 2018Journal of Ocean University of China2018,17,2:4
Ontogenetic difference of beak elemental concentration and its possible application in migration reconstruction for Ommastrephes bartramii in the North Pacific Ocean显示文摘The migration route of oceanic squid provides critical information for us to understand their spatial and temporal variations.Mark-recapture and electronic tags tend to be problematic during processing.Cephalopod hard structures such as the beak, containing abundant ecological information with stable morphology and statolithlike sequences of growth increments, may provide information for studying spatio-temporal distribution. In this study,we developed a method, which is based on elemental concentration of beaks at different ontogenetic stages and sampling locations,to reconstruct the squid migration route. We applied this method to Ommastrephes bartramii in the North Pacific Ocean.Nine trace elements were detected in the rostrum sagittal sections(RSS)of the beak using laser ablation inductively coupled plasma mass spectrometry(LA-ICP-MS).For those elements, significant differences were found between the different ontogenetic stages for phosphorus(P),copper(Cu)and zinc(Zn).Sodium(Na),P and Zn were chosen as indicators of sea surface temperature(SST)and a regression model was estimated. The high probability of occurrence in a particular area represented the possible optimal squid location based on a Bayesian model.A reconstructed migration route in this study, combining all the locations at different ontogenetic stages, was consistent with that hypothesized in previous studies. This study demonstrates that the beak can provide useful information for identifying the migration routes of oceanic squid.Zhou Fang Bilin Liu Xinjun Chen Yong Chen 2019Acta Oceanologica Sinica2019,38,10:4
