| 1 | One-step synthesis of fluorescent hydroxyls-coated carbon dots with hydrothermal reaction and its application to optical sensing of metal ions显示文摘Carbon dots (CDs) with average diameter of 3.1 ± 0.5 nm were facilely synthesized with candle soot through hydrothermal reaction in sodium hydroxide aqueous solution. The as-prepared CDs were covered with a lot of hydroxyls, possessed properties of good water-solubility, anti-photobleaching, salt tolerance, and low cytotoxicity, and had a fluorescence quantum yield (QY) of about 5.5%. The fluorescence of the hydroxyls-coated CDs could be selectively quenched by metal ions such as Cr3+, Al3+ and Fe3+, which is because these metals can easily combine with the hydroxyl groups on the surface of CDs and induce aggregation of hydroxyls-coated CDs. | 3 | Petroleum hydrocarbons and their effects on fishery species in the Bohai Sea, North China显示文摘Systematic studies on the changes in concentrations of petroleum hydrocarbons (PHs) and their effects on fishery species in the Bohai Sea during 1974-2004 are presented. Changes in PHs concentrations were closely related to Yellow River runoff. Concentrations of PHs accumulated in fish and shrimp increased by about 0.712 mg/kg dry weight when trophic level of fish and shrimps increased by 1. Attention should also be paid to the high PHs concentrations in mollusks along the coastal waters of the Bohai Sea. Average concentration of PHs in the adjacent coastal waters of Tianjin City during 1996-2005 decreased the population growth rates of fish, crustaceans and mollusks in the Bohai Sea by 2.58%, 6.59% and 2.33%, respectively. | 4 | Mouse macrophage specific knockout of SIRT1 influences macrophage polarization and promotes angiotensin Ⅱ-induced abdominal aortic aneurysm formation显示文摘Abdominal aortic aneurysm(AAA) is a vascular degenerative disease. Macrophage polarization and the balance between classically activated macrophages(M1) and alternatively activated macrophages(M2)are crucial for AAA pathogenesis. The present study aims to investigate the roles of macrophage SIRT1 in AAA formation and macrophage polarization. We found that in mouse peritoneal macrophages, SIRT1 expression was decreased after M1 stimulation, but was enhanced after M2 stimulation. Results from SIRT1^(flox/flox) mice and macrophage specific SIRT1 knockout mice with treatment of angiotensin II(Ang II)for 4 weeks showed that macrophage specific deficiency of SIRT1 increased the incidence of AAA and exacerbated the severity, including more severe aneurysm types, enlarged diameter of the aneurysm and increased degradation of elastin. In mouse aortas, SIRT1 deficiency increased the proinflammatory M1 molecule inducible nitric oxide synthase(i NOS), and decreased M2 molecules such as arginase 1(Arg1) and mannose receptor(MR). Furthermore, in peritoneal macrophages, SIRT1 deficiency increased the expression of M1 inflammatory molecules, but decreased the expression of M2 molecules. Overexpression of SIRT1 had the opposite effects. | 8 | Carbon sinks/sources in the Yellow and East China Seas Air-sea interface exchange, dissolution in seawater, and burial in sediments显示文摘The sinks/sources of carbon in the Yellow Sea(YS) and East China Sea(ECS), which are important continental shelf seas in China, could exert a great influence on coastal ecosystem dynamics and the regional climate change process. The CO_2 exchange process across the seawater-air interface, dissolved and particulate carbon in seawater, and carbon burial in sediments were studied to understand the sinks/sources of carbon in the continental shelf seas of China. The YS and the ECS generally have different patterns of seasonal air-sea CO_2 exchange. In the YS, regions west of 124°E can absorb CO_2 from the atmosphere during spring and winter, and release CO_2 to the atmosphere during summer and autumn. The entire YS is considered as a CO_2 source throughout the year with respect to the atmosphere, but there are still uncertainties regarding the exact air-sea CO_2 exchange flux. Surface temperature and phytoplankton production were the key controlling factors of the air-sea CO_2 exchange flux in the offshore region and nearshore region of the YS, respectively. The ECS can absorb CO_2 during spring, summer, and winter and release CO_2 to the atmosphere during autumn. The annual average exchange rate in the ECS was-4.2±3.2 mmol m^(-2) d^(-1) and it served as an obvious sink for atmospheric CO_2 with an air-sea exchange flux of 13.7×10~6 t. The controlling factors of the air-sea CO_2 exchange in the ECS varied significantly seasonally. Storage of dissolved inorganic carbon(DIC) and dissolved organic carbon(DOC) in the YS and the ECS were 425×10~6 t and 1364×10~6 t, and 28.2×10~6 t and 54.1×10~6 t,respectively. Long-term observation showed that the DOC content in the YS had a decreasing trend, indicating that the 'practical carbon sink' in the YS was decreasing. The total amount of particulate organic carbon(POC) stored in the YS and ECS was10.6×10~6 t, which was comparable to the air-sea CO_2 flux in these two continental shelf seas. The amounts of carbon sequestered by phytoplankton in the YS and the ECS were 60.42×10~6 t and 153.41×10~6 t, respectively. Artificial breeding of macroalgae could effectively enhance blue carbon sequestration, which could fix 0.36×10~6–0.45×10~6 t of carbon annually. Organic carbon(OC) buried in the sediments of the YS was estimated to be 4.75×10~6 t, and OC of marine origin was 3.03×10~6 t, accounting for5.0% of the TOC fixed by phytoplankton primary production. In the ECS, the corresponding depositional flux of OC in the sediment was estimated to be 7.4×10~6 t yr^(-1), and the marine-origin OC was 5.5×10~6 t, accounting for 5.4% of the phytoplankton primary production. | 9 | Physiological and Molecular Features of Puccinellia tenuiflora Tolerating Salt and Alkaline-Salt Stress显示文摘Saline-alkali soil seriously threatens agriculture productivity; therefore, understanding the mechanism of plant tolerance to alkaline-salt stress has become a major challenge. Halophytic Puccinellia tenuiflora can tolerate salt and alkaline-salt stress, and is thus an ideal plant for studying this tolerance mechanism. In this study, we examined the salt and alkaline-salt stress tolerance of P. tenuiflora, and analyzed gene expression profiles under these stresses. Physiological experiments revealed that P. tenuiflora can grow normally with maximum stress under 600 mmol/L NaCl and 150 mmol/L Na 2 CO 3 (pH 11.0) for 6 d. We identified 4,982 unigenes closely homologous to rice and barley. Furthermore, 1,105 genes showed differentially expressed profiles under salt and alkaline-salt treatments. Differentially expressed genes were overrepresented in functions of photosynthesis, oxidation reduction, signal transduction, and transcription regulation. Almost all genes downregulated under salt and alkaline-salt stress were related to cell structure, photosynthesis, and protein synthesis. Comparing with salt stress, alkaline-salt stress triggered more differentially expressed genes and significantly upregulated genes related to H + transport and citric acid synthesis. | 10 | New records of Jurassic petrified wood in Jianchang of western Liaoning,China and their palaeoclimate implications显示文摘Diverse and rich fossil wood records have been documented from the Mesozoic of Liaoning Province, NE China. The Tiaojishan Formation(mainly distributed in Beipiao, Chaoyang, Fuxin and Jianchang regions) is one of the most significant horizons for the Jurassic petrified wood in Liaoning Province. Previously, wood fossils of this formation were mainly reported in Beipiao and Chaoyang regions, whereas fossil wood record was merely known in Jianchang region. Here we describe new fossil wood specimens from the Tiaojishan Formation in Jianchang County, western Liaoning. Two fossil wood taxa, i.e., Protaxodioxylon jianchangense Tian et Wang sp. nov. and Xenoxylon peidense Zheng et Zhang were recognized on the basis of anatomical features. These fossil wood records add new data for understanding the fossil wood diversity, floral composition and palaeoclimate of the Tiaojishan Formation. | 12 | Environmental significance of biogenic elements in surface sediments of the Changjiang Estuary and its adjacent areas显示文摘Biogenic elements and six phosphorus(P) fractions in surface sediments from the Changjiang Estuary and adjacent waters were determined to investigate the governing factors of these elements, and further to discuss their potential uses as paleo-environment proxies and risks of P release from sediment. Total organic carbon(TOC) and leachable organic P(Lea-OP) showed high concentrations in the estuary, Zhejiang coast and ofshore upwelling area. They came from both the Changjiang River and marine biological input.Biogenic silicon(BSi) exhibited a high concentration band between 123 and 124°E. BSi mainly came from diatom production and its concentration in the inshore area was diluted by river sediment. Total nitrogen(TN) was primarily of marine biogenic origin. Seaward decreasing trends of Fe-bound P and Al-bound P revealed their terrestrial origins. Influenced by old Huanghe sediment delivered by the Jiangsu coastal current, the maximum concentration of detrital P(Det-P) was observed in the area north of the estuary. Similar high concentrations of carbonate fluorapatite(CFA-P) and CaCO3 in the southern study area suggested marine calcium-organism sources of CFA-P. TOC, TN and non-apatite P were enriched in fine sediment, and Det-P partially exhibited coarse-grain enrichment, but BSi had no correlation with sediment grain size. Diferent sources and governing factors made biogenic elements and P species have distinct potential uses in indicating environmental conditions. Transferable P accounted for 14%-46% of total P. | 14 | Dissolved barium as a tracer of Kuroshio incursion in the Kuroshio region east of Taiwan Island and the adjacent East China Sea显示文摘From May to June 2014, the geochemical characteristics of dissolved barium(Ba) in sea water and its influx from the Kuroshio into the East China Sea(ECS) were studied by investigation of the Kuroshio mainstream east of Taiwan Island and the adjacent ECS. This allowed for the scope and extent of the Kuroshio incursion to be quantitatively described for the first time by using Ba as a tracer. The concentration of Ba in the Kuroshio mainstream increased gradually downward from the surface in the range 4.91–19.2 μg L.1. In the surface layer of the ECS, the Ba concentration was highest in coastal water and gradually decreased seaward, while it was higher in coastal and offshore water but lowest in middle shelf for bottom layer. The influx of Ba from Kuroshio into the ECS during May to October was calculated to be 2.19×108 kg by a water exchange model, in which the subsurface layer had the largest portion. The distribution of Ba indicated that Kuroshio upwelled in the sea area northeast of Taiwan Island. The north-flowing water in the Taiwan Strait restrained the incursion of Kuroshio surface water onto the ECS shelf, while Kuroshio subsurface water gradually affected the bottom of the ECS from outside. The results of end member calculation, using Ba as a parameter, showed that the Kuroshio surface water had little impact on the ECS, while the Kuroshio subsurface water formed an intrusion current by climbing northwest along the bottom of the middle shelf from the sea area northeast of Taiwan Island into the Qiantang Estuary, of which the volume of Kuroshio water was nearly 65%. Kuroshio water was the predominant part of the water on the outer shelf bottom and its proportion in areas deeper than the 100 m isobath could reach more than 95%. In the DH9 section(north of Taiwan Island), Kuroshio subsurface water intruded westward along the bottom from the shelf edge and then rose upward(in lower proportion). Kuroshio water accounted for 95% of the ocean volume could reach as far as 122°E. | 15 | Synergistic defensive mechanism of phytochelatins and antioxidative enzymes in Brassica chinensis L. against Cd stress显示文摘Brassica chinensis L. was chosen and exposed to different concentrations of Cd exposure to evaluate its Cd-accumulating capacity and its potential cellular defensive mechanisms. Cd accumulation in the shoots and roots of B. chinensis was up to 1348.3±461.8 and 3761.0±795.0 mg per killogram of dry weight, respectively, under 200 μmol/L of Cd exposure. Increasing Cd accumulation in the plant was accompanied by rapid accumulation of phytochelatins (PCs), and the sequestration of Cd by PCs provided a primary cellular mechanism for Cd detoxification and tolerance of B. chinensis. Furthermore, malondialdehyde formation, hydrogen peroxide content and antioxidative enzyme activities such as superoxide dismutase, catalase, guaiacol peroxidase and ascorbate peroxidase were observed in the shoots of Cd-stressed B. chinensis. Increasing enzyme activities in response to concentrations of 5 to 50 μmol/L Cd showed an efficient defense against oxidative stress, suggesting that the antioxidative system was a secondary defensive mechanism. These resulted in reduced free Cd damage and enhanced Cd accumulation and tolerance. Glutathione plays a pivotal role in these two detoxification pathways. | 16 | SAD2 in Arabidopsis Functions in Trichome Initiation through Mediating GL3 Function and Regulating GL1,TTG1 and GL2 Expression显示文摘Most genes identified that control Arabidopsis trichome initiation and formation are transcription factors or regulatory com- ponents in transcriptional networks and include GLABROUS1 (GL1), GLABRA2 (GL2), GLABRA3 (GL3) and TRANSPARENT TESTA GLABRA1 (TTG1). Herein, we report that an importin β-like protein, SENSITIVE TO ABA AND DROUGHT2 (SAD2), is required for trichome initiation. Mutations in SAD2 disrupted trichome initiation resulting in reduced trichome number, but had no effect on trichome development or root hair number and development. Expression levels of GL1, MYB23, GL2 and TTG1 were reduced in shoots of sad2 mutants while expression levels of GL3 and ENHANCER OF GLABRA3 (EGL3) were enhanced. Overexpression of GL3 increased trichome numbers in wild type but not in sad2 mutants, indicating that the function of the GL3 protein is altered in the sad2 mutants. In contrast, overexpression of GFP-GL1 decreased trichome number in both wild type and sad2. Double mutant analysis of gl1 sad2 and gl3 sad2 indicated that SAD2 functions genetically, at least in part, in the same pathway with these two genes. Co-immunoprecipitation indicated that the sad2 mutation does not disrupt formation of the TTG1-GL3-GL1 complex. Analysis of GFP fusions of GL1, GL2, GL3 and TTG1 suggested that these proteins are most likely not direct cargo of SAD2. Then,the nanofiltration(NF) membrane fouling experiments were carried out using the above fractionated water to investigate the effect of MW distribution and hydrophilic/hydrophobic characteristics of EfOM on the membrane flux decline using the fractionated water samples. The experimental results have shown that 45.61% of the total organics belongs to the low MW one,among which the percentage of the hydrophilic organics with low MW(less than 2 kDa) was up to 28.07%,while that of the hydrophobic organics was 17.54%. In particular,the hydrophilic fraction was found to be the most abundant fraction in the effluents. MW distribution has a significant effect on the membrane fouling. When the MW was less than 30 kDa,the lower the MW,the larger was the specific flux decline,while in the case of MW higher than 30 kDa,the higher the MW,the larger was the specific flux decline,and the decline degree of low MW organics was larger than the high MW one.
