Laboratory pull-out tests on fully grouted rock bolts and cable bolts:Results and lessons learned显示文摘Laboratory pull-out tests were conducted on the following rock bolts and cable bolts:steel rebars, smooth steel bars, fiberglass reinforced polymer threaded bolts, flexible cable bolts,IR5/IN special cable bolts and Mini-cage cable bolts. The diameter of the tested bolts was between 16 mm and 26 mm.The bolts were grouted in a sandstone sample using resin or cement grouts. The tests were conducted under either constant radial stiffness or constant confining pressure boundary conditions applied on the outer surface of the rock sample. In most tests, the rate of displacement was about 0.02 mm/s.The tests were performed using a pull-out bench that allows testing a wide range of parameters. This paper provides an extensive database of laboratory pull-out test results and confirms the influence of the confining pressure and the embedment length on the pull-out response(rock bolts and cable bolts). It also highlights the sensitivity of the results to the operating conditions and to the behavior of the sample as a whole, which cannot be neglected when the test results are used to assess the bolt-grout or the grouterock interface. | Isabelle Thenevin Laura Blanco-Martín Faouzi Hadj-Hassen Jacques Schleifer Zbigniew Lubosik Aleksander Wrana | 2017 | Journal of Rock Mechanics and Geotechnical Engineering2017,9,5: | 14
ApoB-100, ApoE and CYP7A1 gene polymorphisms in Mexican patients with cholesterol gallstone disease显示文摘AIM: To determine the possible association of the ApoB100 (Xba Ⅰ ), ApoE (Hha Ⅰ ) and CYP7A1 (Bsa Ⅰ ) gene polymorphisms, with the development of cholesterol gallstone disease (GD) in a Mexican population. METHODS: The polymorphisms were analyzed by polymerase chain reaction followed by restriction fragment length polymorphism, in two groups matched by ethnicity, age and sex: patients with GD (n = 101) and stone-free control subjects (n = 101). RESULTS: Allelic frequencies in patients and controls were: 34.16% vs 41.58% (P = 0.124) for X+of ApoB-100; 4.46% vs 5.94% (P = 0.501) for E2, 85.64% vs 78.22% (P = 0.052) for E3, 9.90% vs 15.84% (P = 0.075) for E4 of ApoE; and 25.74% vs 27.72% (P = 0.653) for C of CYP7A1. Differences in genotypic frequencies between the studied groups were not significant (P < 0.05). CONCLUSION: These results demonstrated that no association exists between the studied polymorphisms and cholelithiasis in this high prevalent population. | Sánchez-Cuén Jaime Aguilar-Medina Maribel Arámbula-Meraz Eliakym Romero-Navarro José Granados Julio Sicairos-Medina Laura Ramos-Payán Rosalío | 2010 | World Journal of Gastroenterology2010,16,37: | 14
An integrated approach for increasing breeding efficiency in apple and peach in Europe显示文摘Despite the availability of whole genome sequences of apple and peach, there has been a considerable gap between genomics and breeding. To bridge the gap,the European Union funded the FruitBreedomics project(March 2011 to August 2015)involving 28 research institutes and private companies.Three complementary approaches were pursued:(i)tool and software development,(ii)deciphering genetic control of main horticultural traits taking into account allelic diversity and(iii)developing plant materials, tools and methodologies for breeders.Decisive breakthroughs were made including the making available of ready-to-go DNA diagnostic tests for Marker Assisted Breeding, development of new, dense SNP arrays in apple and peach, new phenotypic methods for some complex traits, software for gene/QTL discovery on breeding germplasm via Pedigree Based Analysis(PBA).This resulted in the discovery of highly predictive molecular markers for traits of horticultural interest via PBA and via Genome Wide Association Studies(GWAS)on several European genebank collections.FruitBreedomics also developed pre-breeding plant materials in which multiple sources of resistance were pyramided and software that can support breeders in their selection activities.Through FruitBreedomics, significant progresses were made in the field of apple and peach breeding, genetics, genomics and bioinformatics of which advantage will be made by breeders, germplasm curators and scientists.A major part of the data collected during the project has been stored in the FruitBreedomics database and has been made available to the public. This review covers the scientific discoveries made in this major endeavour, and perspective in the apple and peach breeding and genomics in Europe and beyond. | Francois Laurens Maria JoséAranzana Pere Arus Daniele Bassi Marco Bink Joan Bonany Andrea Caprera Luca Corelli-Grappadelli Evelyne Costes Charles-Eric Durel Jehan-Baptiste Mauroux Hélène Muranty Nelson Nazzicari Thierry Pascal Andrea Patocchi Andreas Peil Bénédicte Quilot-Turion Laura Rossini Alessandra Stella Michela Troggio Riccardo Velasco Eric van de Weg | 2018 | Horticulture Research2018,5,1: | 10
MicroRNAs dysregulation in hepatocellular carcinoma: Insights in genomic medicine显示文摘Hepatocellular carcinoma(HCC) is the leading primary liver cancer and its clinical outcome is still poor. MicroRNAs(mi RNAs) have demonstrated an interesting potential to regulate gene expression at post-transcriptional level. Current findings suggest that mi RNAs deregulation in cancer is caused by genetic and/or epigenetic, transcriptional and post-transcriptional modifications resulting in abnormal expression and hallmarks of malignant transformation: aberrant cell growth, cell death, differentiation, angiogenesis, invasion and metástasis. The important role of mi RNAs in the development and progression of HCC has increased the efforts to understand and develop mechanisms of control overt this single-stranded RNAs. Several studies have analyzed tumoral response to the regulation and control of deregulated mi RNAs with good results in vitro and in vivo, proving that targeting aberrant expression of mi RNAs is a powerful anticancer therapeutic. Identification of up and/or down regulated mi RNAs related to HCC has led to the discovery of new potential application for detection of their presence in the affected organism. Mi RNAs represent a relevant new target for diagnosis, prognosis and treatment in a wide variety of pathologic entities, including HCC. This manuscript intends to summarize current knowledge regarding mi RNAs and their role in HCC development. | Iván Lyra-González Laura E Flores-Fong Ignacio González-García David Medina-Preciado Juan Armendáriz-Borunda | 2015 | World Journal of Hepatology2015,7,11: | 9
Counter-regulatory phosphatases TNAP and NPP1 temporally regulate tooth root cementogenesis显示文摘Cementum is critical for anchoring the insertion of periodontal ligament fibers to the tooth root. Several aspects of cementogenesis remain unclear, including differences between acellular cementum and cellular cementum, and between cementum and bone.Biomineralization is regulated by the ratio of inorganic phosphate(Pi) to mineral inhibitor pyrophosphate(PPi), where local Piand PPi concentrations are controlled by phosphatases including tissue-nonspecific alkaline phosphatase(TNAP) and ectonucleotide pyrophosphatase/phosphodiesterase 1(NPP1). The focus of this study was to define the roles of these phosphatases in cementogenesis. TNAP was associated with earliest cementoblasts near forming acellular and cellular cementum. With loss of TNAP in the Alpl null mouse, acellular cementum was inhibited, while cellular cementum production increased, albeit as hypomineralized cementoid. In contrast, NPP1 was detected in cementoblasts after acellular cementum formation, and at low levels around cellular cementum. Loss of NPP1 in the Enpp1 null mouse increased acellular cementum, with little effect on cellular cementum.Developmental patterns were recapitulated in a mouse model for acellular cementum regeneration, with early TNAP expression and later NPP1 expression. In vitro, cementoblasts expressed Alpl gene/protein early, whereas Enpp1 gene/protein expression was significantly induced only under mineralization conditions. These patterns were confirmed in human teeth, including widespread TNAP, and NPP1 restricted to cementoblasts lining acellular cementum. These studies suggest that early TNAP expression creates a low PPienvironment promoting acellular cementum initiation, while later NPP1 expression increases PPi, restricting acellular cementum apposition. Alterations in PPihave little effect on cellular cementum formation, though matrix mineralization is affected. | Laura E Zweifler Mudita K Patel Francisco H Nociti Jr Helen F Wimer Jose L Milln Martha J Somerman Brian L Foster | 2015 | International Journal of Oral Science2015,7,1: | 5
Evaluation of the Ex-PRESS~ P-50 implant under scleral flap in combined cataract and glaucoma surgery显示文摘AIM: To evaluate the efficacy and safety of glaucoma drainage device Ex-PRESS~ P-50 for combined cataract surgery and glaucoma.·METHODS: Patients having cataract and open angle glaucoma or patients with open advanced glaucoma which needed two or more antiglaucoma medications were included. Combined cataract surgery and glaucoma with Ex-PRESS~ P-50 model placed under scleral flap was performed.·RESULTS: Out of 40 eyes of 40 patients(55% male and45% female) completed the study during one-year follow-up. The mean of age was 76.6 ±11.02 y. The intraocular pressure(IOP) decreased significantly during the 12-month follow-up from 23.5 mm Hg to 16.8 mm Hg(Wilcoxon signed ranks test, P <0.001). A 59.5% of patients did not need any topical treatment, 10.8% of them needed one active principle, 27% needed two active principles, and 2.7% of them needed three active principles for successful IOP control(<21 mm Hg).·CONCLUSION:Combinedsurgeryofphacoemulsification with Ex PRESS~ P-50 lowers IOP from the preoperative baseline and reduces significantly the number of antiglaucoma active principles for IOP control after the operation. | Valentín Huerva Jordi Soldevila Francisco J.Ascaso Laura Lavilla M.Jesús Muniesa M.Carmen Sánchez | 2016 | International Journal of Ophthalmology(English edition)2016,9,4: | 5
Tolerance in liver transplantation: Biomarkers and clinical relevance显示文摘Transplantation is the optimal treatment for end-stage organ failure, and modern immunosuppression has allowed important progress in short-term outcomes. However, immunosuppression poorly influences chronic rejection and elicits chronic toxicity in current clinical practice. Thus,a major goal in transplantation is to understand and induce tolerance. It is well established that human regulatory T cells expressing the transcription factor Fox P3 play important roles in the maintenance of immunological self-tolerance and immune homeostasis. The major regulatory T cell subsets and mechanisms of expansion that are critical for induction and long-term maintenance of graft tolerance and survival are being actively investigated. Likewise, other immune cells, such as dendritic cells,monocyte/macrophages or natural killer cells, have been described as part of the process known as 'operational tolerance'. However, translation of these results towards clinical practice needs solid tools to identify accurately and reliably patients who are going to be tolerant. In this way,a plethora of genetic and cellular biomarkers is raising and being validated worldwide in large multicenter clinical trials. Few of the studies performed so far have provided a detailed analysis of the impact of immunosuppression withdrawal on pre-existing complications derived from the long-term administration of immunosuppressive drugs and the side effects associated with them. The future of liver transplantation is aimed to develop new therapies which increase the actual low tolerant vs non-tolerant recipients ratio. | Alberto Baroja-Mazo Beatriz Revilla-Nuin Pascual Parrilla Laura Martínez-Alarcón Pablo Ramírez José Antonio Pons | 2016 | World Journal of Gastroenterology2016,22,34: | 4
Diagnosis in bile acid-CoA:Amino acid N-acyltransferase deficiency显示文摘Cholate-CoA ligase(CCL) and bile acid-CoA:amino acid N-acyltransferase(BAAT) sequentially mediate bile-acid amidation.Defects can cause intrahepatic cholestasis.Distinction has required gene sequencing. We assessed potential clinical utility of immunostaining of liver for CCL and BAAT.Using commercially available antibodies against BAAT and CCL,we immunostained liver from an infant with jaundice, deficiency of amidated bile acids, and transcription-terminating mutation in BAAT.CCL was normally expressed.BAAT expression was not detected.Immunostaining may facilitate diagnosis in bileacid amidation defects. | Nedim Hadzi Laura N Bull Peter T Clayton AS Knisely | 2012 | World Journal of Gastroenterology2012,18,25: | 3
Downregulation of the Basic Peroxidase Isoenzyme from Zinnia elegans by Gibberellic Acid显示文摘Hypocotyl formation during the epigeal germination of seedlings is under strict hormonal regulation. In a 3 d old Zinnia elegans seedling system, gibberellic acid (GA3) exerts an opposite effect to that exerted by light on hypocotyl photomorphogenesis because GA3 promotes an etiolated-like growth with an inhibition of radial (secondary) growth. For this reason, the effect of GA3 on the basic peroxidase isoenzyme from Z. elegans (ZePrx), an enzyme involved in hypocotyl lignin biosynthesis, was studied. The results showed that GA3 reduces ZePrx activity, similarly to the way in which it reduces seedling secondary growth. This hormonal response is supported by the analysis of the ZePrx promoter, which contains four types of GA3-responsive cis-elements: the W Box/O2S; the Pyr Box; the GARE; and the Amy Box. Taken together, these results suggest that ZePrx is directly regulated by GA3, with this effect matching the inhibitory effect of GA on the hypocotyl secondary growth. | María Josefa López Núez-Flores Jorge Gutirrez Laura V.Gómez-Ros Esther Novo Uzal Mariana Sottomayor Alfonso Ros Barceló | 2010 | Journal of Integrative Plant Biology2010,52,2: | 3

Extracellular vesicles in liver disease and beyond显示文摘Extracellular vesicles(EVs) are membrane-derived vesicles which can be released by different cell types, including hepatocytes, hepatic stellate cells and immune cells in normal and pathological conditions. EVs carry lipids, proteins, coding and non-coding RNAs and mitochondrial DNA causing modifications on the recipient cells. These vesicles are considered potential biomarkers and therapeutic agents for human diagnostic and prognostic due to their function as intercellular mediators of cell-cell communication within the liver and between other organs. However, the development and optimization of methods for EVs isolation is required to characterize their biological functions as well as their potential as a treatment option in the clinic. Nevertheless, many questions remain unanswered related to the function of EVs under physiological and pathological conditions. In the current editorial, the results obtained in different studies that investigated the role of intrahepatic EVs during liver diseases, including drug-induced liver injury, non-alcoholic fatty liver, nonalcoholic steatohepatitis, alcoholic liver disease and hepatocellular carcinoma and extrahepatic EVs in remote organs during pathological events such as pulmonary disease, cardiovascular diseases, neurodegenerative disorders e.g., Alzheimer's disease, Parkinson's disease and multiple sclerosis as well as in immunopathological processes, are discussed. Although much light needs to be shed on the mechanisms of EVs, these membranederived vesicles represent both a novel promising diagnostic, and a therapeutic tool for clinical use that we emphasize in the current editorial. | Laura Morán Francisco Javier Cubero | 2018 | World Journal of Gastroenterology2018,24,40: | 3
Paclobutrazol and Its Use in Fruit Production:A Review显示文摘There are documentary records referring to paclobutrazol(PBZ)as a growth bioregulator that inhibits the gibberellin synthesis and its application increases yields in fruit and vegetable crop productions. Its agronomic management includes it as an emerging technology to reduce vigour, promote flower induction and flower development in fruit trees with increased economic returns. Its use is banned in some countries because of concerns about residues that can cause harmful effects on the environment. Therefore, the aim of this article was to collect, analyse and summarise relevant information on the use of PBZ in fruit tree production and its possible risks to the environment. The results obtained indicated that the application of PBZ can be effective in solving some problems related to flowering if it is applied in the right amount and at the right time. However,it is necessary to elucidate the physiological processes with which it is associated and its response to be taken into account to increase yield.PBZ is currently used in fruit trees such as mango, lime, apple and guava, increasing their productivity. However, some studies have shown its residual effect on the environment. Therefore,PBZ is a viable strategy, because it presents a series of advantages in the production of fruit trees. However,it is vital to generate protocols that seek its regulation with a rational and sustainable approach. | Laura Raquel Orozco-Meléndez Ofelia Adriana Hernández-Rodríguez Oscar Cruz-Álvarez Loreto Robles-Hernández Graciela Dolores Avila-Quezada Esteban Sánchez Chavez DamiánAarón Porras-Flores Dámaris Leopoldina Ojeda-Barrios | 2022 | Phyton-International Journal of Experimental Botany2022,91,1: | 2

Efficacy of bevacizumab-containing chemotherapy in metastatic colorectal cancer and CXCL5 expression: Six case reports显示文摘BACKGROUND In metastatic colorectal cancer(mCRC),the anti-vascular endothelial growth factor drug bevacizumab(BVZ)plus chemotherapy significantly improves progression-free survival compared to chemotherapy(CT)alone. This benefit is not, however, observed in all patients. While increased chemokine CXCL5 gene expression promoting angiogenesis has been proposed as a prognostic mCRC biomarker, few studies have examined its relationship with drug efficacy. This study sought to analyze tumor CXCL5 gene expression in six patients with different efficacy of BVZ-containing CT in terms of the tumor response to treatment.CASE SUMMARY We report six cases of stage IV KRAS-mutated mCRC.Patients were given first line treatment with BVZ-containing chemotherapy in University Hospital of Fuenlabrada. The six patients differed in terms of primary tumor location(right/left side),tumor burden(mostly hepatic and peritoneal disease)and clinical disease course. Before treatment onset, total RNA was isolated from paraffinated tumor biopsy specimens and CXCL5 gene expression quantified through conventional RT-qPCR procedures. Our main finding was that CXCL5 expression levels were several times higher in three patients with lower progression free survival(under 6 mo)from the start of treatment.CONCLUSION A higher expression of CXCL5 was observed in the three patients showing worse tumor response to treatment. | Apolonia Novillo María Gaibar Alicia Romero-Lorca María Fuencisla Gilsanz Laura Beltrán Miguel Galán Beatriz Antón Diego Malón Amalia Moreno Ana Fernández-Santander | 2020 | World Journal of Gastroenterology2020,26,16: | 2
The first pterosaur 3-D egg:Implications for Pterodaustro guinazui nesting strategies,an Albian filter feeder pterosaur from central Argentina显示文摘The increasing number of fossil pterosaur eggs sheds light on nesting environments and breeding behaviors of these extinct flying reptiles. Here we report the first partial three-dimensional egg of the pterosaur,Pterodaustro guinazui, from central Argentina. The specimen was discovered from the same Albian deposits as the exceptional P.guinazui embryo described in 2004.Microscopic characterizations indicate a pristine preservation of the 50 mm thick calcium carbonate, which differs significantly from the soft shell of Chinese pterosaur eggs.Estimate of the eggshell conductance implies that the nest had a minimum moisture content of 75%.This moisture estimate, combined with geological and taphonomical data, suggests that P.guinazui may have adopted a nesting strategy similar to those of grebes and flamingos rather than being buried on land,as previously hypothesized. Moreover, our results demonstrate that the nesting paleoenvironment of this pterosaur species was closely linked to a mesohaline lacustrine ecosystem in a basin governed by regional tectonic subsidence,a setting characteristic for the feeding and reproduction of modern flamingos. | Gerald Grellet-Tinner Michael B.Thompson Lucas E.Fiorelli Eloísa Argaaraz Laura Codorniú E.Martín Hechenleitner | 2014 | Geoscience Frontiers2014,5,6: | 2
Telecentres in Mountain Regions-A Peruvian Case Study of the Impact of Information and Communication Technologies on Remoteness and Exclusion显示文摘Communities in developing country mountain areas,in part due to their remoteness, find themselves excluded from social, political and economic systems; and excluded from access to resources. This paper aims to study the impact of information and communication technologies (ICTs) on remoteness and exclusion. It utilises two models - the resource movement framework, and the 'information chain' - to analyse a telecentre in one district of mountainous Huancavelica,Peru's poorest region, set in the high Andes. It finds ICTs enabling new and positive resource flows for the two key user groups:teenaged school students and young farmers. These help to maintain social networks. They also support information searches that have improved agricultural practice where other information chain resources have been available. But non-use and ineffective use of the telecentre are found where information chain resources are lacking. ICTs have some impacts on intangible elements of remoteness. In this particular example, they also offer access to some previously-excluded resources. But they have not really addressed the systematic exclusions faced by mountain communities. And they so far appear to be a technology of inequality; favouring those residents who begin with better resource endowments. The paper concludes by offering some recommendations for mountain ICT project practice. | Richard Heeks Laura León Kanashiro | 2009 | Journal of Mountain Science2009,6,4: | 2
