Enzyme activities and gene expression of starch metabolism provide insights into grape berry development显示文摘Grapes are categorized as a non-climacteric type of fruit which its ripening is not associated to important rises in respiration and ethylene synthesis. The starch metabolism shares a certain role in the carbohydrate metabolic pathways during grape berry development, and is regarded as an important transient pool in the pathway of sugar accumulation. However, the comprehensive role of starch and its contribution to the quality and flavor of grape berry have not been explored thoroughly. In this study, the expression levels of genes enzyme activities and carbohydrate concentrations related to starch metabolism, were analyzed to understand the molecular mechanism of starch accumulation during grape berry development. The results indicated that starch granules in grape berry were located at the chloroplast in the sub-epidermal tissues, acting as the temporary reserves of photosynthetic products to meet the needs for berry development, and relatively high starch contents could be detected at véraison stage. Moreover, both ADP-glucose pyrophosphorylase(EC 2.7.7.27)and sucrose phosphate synthase(EC 2.3.1.14)involved in starch synthesis displayed elevated gene expression and enzymes activities in the sub-epidermal tissue,whileα-andβ-amylases involved in its degradation were highly transcribed and active in the central flesh, explaining the absence of starch in this last tissue.Change in the gene expression and activities of ADP-glucose pyrophosphorylase,β-amylase and sucrose phosphate synthase revealed that they were regulated by the circadian rhythms in the fruitlets compared with those in the leaves. Both the morphological, enzymological and transcriptional data in this study provide advanced understandings on the function of starch during berry development and ripening that are so important for berry quality. This study will further facilitate our understanding of the sugar metabolism in grape berry as well as in other plant species. | Xudong Zhu Chaobo Zhang Weimin Wu Xiaopeng Li Chuan Zhang Jinggui Fang | 2017 | Horticulture Research2017,4,1: | 4
Mutation of the PHYTOENE DESATURASE 3 gene causes yellowish-white petals in Brassica napus显示文摘Oilseed rape (Brassica napus) with yellow flowers is an attractive ornamental landscape plant during the flowering period, and the development of different petal colors has become a breeding objective.Although yellowish flower color is a common variant observed in field-grown oilseed rape, the genetics behind this variation remains unclear. We obtained a yellowish-white flower (ywf) mutant from Zhongshuang 9 (ZS9) by ethyl methanesulfonate mutagenesis (EMS) treatment.Compared with ZS9,ywf exhibited a lower carotenoid content with a reduced and defective chromoplast ultrastructure in the petals.Genetic analysis revealed that the yellowish-white trait was controlled by a single recessive gene.Using bulked-segregant analysis sequencing (BSA-seq) and kompetitive allele-specific PCR(KASP),we performed map-based cloning of the ywf locus on chromosome A08 and found that ywf harbored a C-to-T substitution in the coding region, resulting in a premature translation termination.YWF,encoding phytoene desaturase 3 (PDS3),was highly expressed in oilseed rape petals and involved in carotenoid biosynthesis.Pathway enrichment analysis of the transcriptome profiles from ZS9 and ywf indicated the carotenoid biosynthesis pathway to be highly enriched.Further analyses of differentially expressed genes and carotenoid components revealed that the truncated Bna A08.PDS3 resulted in decreased carotenoid biosynthesis in the mutant. These results contribute to an understanding of the carotenoid biosynthesis pathway and manipulation of flower-color variation in B.napus. | Chuanji Zhao Luqman Bin Safdar Meili Xie Meijuan Shi Zhixue Dong Li Yang Xiaohui Cheng Yueying Liu Zetao Bai Yang Xiang Chaobo Tong Junyan Huang Lijiang Liu Shengyi Liu | 2021 | The Crop Journal2021,9,5: | 3
A real-time abnormal operation pattern detection method for building energy systems based on association rule bases显示文摘Expert systems are effective for anomaly detection in building energy systems. However,it is usually inefficient to establish comprehensive rule bases manually for complex building energy systems.Association rule mining is available to accelerate the establishment of the rule bases due to its powerful capability of discovering rules from numerous data. This paper proposes a real-time abnormal operation pattern detection method towards building energy systems. It can benefit from both expert systems and association rule mining.Association rules are utilized to establish association rule bases of abnormal and normal operation patterns. The established rule bases are then utilized to develop an expert system for real-time detection of abnormal operation patterns. The proposed method is applied to an actual chiller plant for evaluating its performance. Results show that 15 types of known abnormal operation patterns and 11 types of unknown abnormal operation patterns are detected successfully by the proposed method. | Chaobo Zhang Yang Zhao Yangze Zhou Xuejun Zhang Tingting Li | 2022 | Building Simulation2022,15,1: | 3

Wearable battery-free theranostic dental patch for wireless intraoral sensing and drug delivery显示文摘Dental caries caused by oral microbiome dysbiosis with the elevation of acidogenic bacteria is the most prevalent noncommunicable disease worldwide.Early prevention and timely fluoride treatment are crucial for caries lesion management.Herein,to address the challenges of in situ sensing and topical drug delivery within the oral cavity,a miniaturized,battery-free,and wearable dental patch system was developed for microenvironment monitoring and controlled fluoride treatment. With the integration of near-field communication, the dental patch realizes wireless energy harvesting and data transmission with mobile terminals like smartphones when attached conformally to the tooth surface. The acidic microenvironment caused by bacterial metabolism are electrochemically detected, while fluorides can be delivered locally from the electric-responsive drug delivery electrode for on-demand treatment. As flexible electronics armed to the teeth, this intraoral theranostic wearable system provides an inspiring platform for point-of-care monitoring and treatment of dental caries and oral diseases. | Zhenghan Shi Yanli Lu Shuying Shen Yi Xu Chang Shu Yue Wu Jingjiang Lv Xin Li Zupeng Yan Zijian An Chaobo Dai Lingkai Su Fenni Zhang Qingjun Liu | 2022 | npj Flexible Electronics2022,6,1: | 1
Mapping loci controlling fatty acid profiles, oil and protein content by genome-wide association study in Brassica napus显示文摘Optimizing the profile and quantity of fatty acids in rapeseed(Brassica napus L.) is critical for maximizing the value of edible oil and biodiesel. However, selection of these complex seed quality traits is difficult before haplotypes controlling their contents are identified. To efficiently identify genetic loci influencing these traits and underlying candidate genes and networks, we performed a genome-wide association study(GWAS) of eight seed quality traits(oil and protein content, palmitic, stearic, oleic, linoleic, eicosenoic and erucic acids content). The GWAS population comprised 370 diverse accessions, which were phenotyped in five environments and genotyped using 60K SNP arrays. The results indicated that oil and protein contents generally showed negative correlations, while fatty acid contents showed positive or negative correlations, with palmitic and erucic acid contents directly affecting oil content. Seven SNPs on five chromosomes were associated with both seed oil and protein content, and five genes orthologous to genes in Arabidopsis thaliana were predicted as candidates. From resequencing data, besides known haplotypes in Bna A.FAE1.a and Bna C.FAE1.a, three accessions harboring a new haplotype conferring moderate erucic acid content were identified. Interestingly, in a haplotype block, one haplotype was associated with high palmitic acid content and low oil content, while the others showed the reverse effects. This finding was consistent with a negative correlation between palmitic acid and oil contents, suggesting historical selection for high oil content. The identification by this study of genetic variation and complex correlations of eight seed quality traits may be beneficial for crop selection strategies. | Minqiang Tang Yuanyuan Zhang Yueying Liu Chaobo Tong Xiaohui Cheng Wei Zhu Zaiyun Li Junyan Huang Shengyi Liu | 2019 | The Crop Journal2019,7,2: | 1
Cloning and sequence analysis of Sox genes in a tetraploid cyprinid fish,Tor douronensis显示文摘A PCR survey for Sox genes in a young tetraploid fish Tor douronensis (Teleostei:Cyprinidae) was per-formed to access the evolutionary fates of important functional genes after genome duplication caused by polyploidization event. Totally 13 Sox genes were obtained in Tor douronensis, which represent SoxB,SoxC and SoxE groups. Phylogenetic analysis of Sox genes in Tor douronensis provided evidence for fish-specific genome duplication, and suggested that Sox19 might be a teleost specific Sox gene member. Sequence analysis revealed most of the nucleotide substitutions between duplicated copies of Sox genes caused by tetraploidization event or their orthologues in other species are silent substitutions. It would appear that the sequences are under purifying selective pressure, strongly suggesting that they repre-sent functional genes and supporting selection against all null allele at either of two duplicated loci of Sox4a,Sox9a and Sox9b. Surprising variations of the intron length and similarities of two duplicated copies of Sox9a and Sox9b,suggest that Tor douronensis might be an allotetraploidy. | GUO BaoCheng Li JunBing TONG ChaoBo HE ShunPing | 2008 | Chinese Science Bulletin2008,53,13: | 1
Federated learning-based short-term building energy consumption prediction method for solving the data silos problem显示文摘Transfer learning is an effective method to predict the energy consumption of information-poor buildings by learning transferable knowledge from operational data of information-rich buildings. However,it is not recommended to directly use the operational data without protection due to the risk of leaking occupants’privacy. To address this problem, this study proposes a federated learning-based method to learn transferable knowledge from building operational data without privacy leaking. It trains a transferable federated model based on the operational data from the buildings similar to the target building with limited data.An advanced secure aggregation algorithm is adopted in the training process to ensure that no one can infer private information from the training data. The federated model obtained is evaluated by comparing it with the standalone model without federated learning based on 13 similar office buildings from the Building Data Genome Project. The results show that the federated model outperforms the standalone model concerning the prediction accuracy and training time.On average, the federated model achieves a 25.4%decrease in CV-RMSE when the target building has limited operational data.Even if the target building has no operational data, the federated model still achieves acceptable accuracy(CV-RMSE is 22.2%).Meanwhile,the training time of the federated model is 90%less than that of the standalone model. The research insights can help develop federated learning-based methods for solving the data silos problem in building energy management. The methodology and analysis procedures are reproducible and all codes and data sets are available on Github. | Junyang Li Chaobo Zhang Yang Zhao Weikang Qiu Qi Chen Xuejun Zhang | 2022 | Building Simulation2022,15,6: | 1

Design of a novel mitochondria targetable turn-on fluorescence probe for hydrogen peroxide and its two-photon bioimaging applications显示文摘Considering that hydrogen peroxide(H2O2)plays significant roles in oxidative stress, the cellular signal transduction and essential biological process regulation, the detection and imaging of H2O2 in living systems undertakes critical responsibility.Herein,we have developed a novel two-photon fluorescence turn on probe, named as Pyp-B for mitochondria H2O2 detection in living systems.Selectivity studies show that probe Pyp-B exhibit highly sensitive response toward H2O2 than other reactive oxygen species(ROS)and reactive nitrogen species(RNS)as well as biologically relevant species. The fluorescence colocalization studies demonstrate that the probe can localize in the mitochondria solely.Furthermore,as a bio-compatibility molecule, the highly selective and sensitive of fluorescence probe Pyp-B have been confirmed by its cell imaging application of H2O2 in living A549 cells and zebrafishes under the physiological conditions. | Kangnan Wang Wen Ma Yuchun Xu Xin Liu Gui Chen Meng Yu Qiling Pan Chaobo Huang Xiaochuan Li Qingchun Mu Yongbing Sun Zhiqiang Yu | 2020 | Chinese Chemical Letters2020,31,12: | 1

A multi agent-based optimal control method for combined cooling and power systems with thermal energy storage显示文摘Combined cooling, heating and power(CCHP)systems have been considered as a potential energy saving technology for buildings due to their high energy efficiency and low carbon emission.Thermal energy storage(TES)can improve the energy efficiency of CCHP systems, since they reduce the mismatch between the energy supply and demand. However,it also increases the complexity of operation optimization of CCHP systems. In this study,a multi-agent system(MAS)-based optimal control method is proposed to minimize the operation cost of CCHP systems combined with TES.Four types of agents,i.e.,coordinator agents, building agents, energy management agents and optimization agents, are implemented in the MAS to cooperate with each other. The operation optimization problem is solved by the genetic algorithm.A simulated system is utilized to validate the performance of the proposed method. Results show that the operation cost reductions of 10.0%on a typical summer day and 7.7%on a typical spring day are achieved compared with a rule-based control method.A sensitivity analysis is further performed and results show that the optimal operation cost does not change obviously when the rated capacity of TES exceeds a threshold. | Zihao Wang Chaobo Zhang Hongbo Li Yang Zhao | 2021 | Building Simulation2021,14,6: | 0
MoS_(2)nanosheets and bulk materials altered lipid profiles in 3D Caco-2 spheroids显示文摘MoS_(2)nanosheets(NSs)are novel 2D nanomaterials(NMs)with potential uses in many areas, and therefore oral exposure route to MoS_(2)NSs is plausible.Currently,MoS_(2)NSs are considered as biocompatible NMs,but there is lacking of systemic investigations to study the interactions of MoS_(2)NSs with intestinal cells. In this study,we exposed the 3D Caco-2 spheroids to MoS_(2)NSs or MoS_(2)powders(denoted as MoS_(2)-bulk),and investigated the potential adverse effects of MoS_(2)-materials based on transcriptomics and lipidomics analysis. As expected, both MoS_(2)NSs and MoS_(2)-bulk were dose-dependently internalized into 3D Caco-2 spheroids but did not induce cytotoxicity, membrane disruption or decrease of thiols. However, the Gene Ontology(GO)and Kyoto Encyclopedia of Gene and Genomes(KEGG)analysis indicated that nutrient absorption and metabolism was decreased. One of the most significantly decreased KEGG pathways is fat digestion and absorption(map04975),and Western blotting analysis further showed that fatty acid binding protein 1 and apolipoprotein A1,key proteins involved in fat digestion and absorption, were down-regulated by MoS_(2)NSs or MoS_(2)-bulk.In addition,BODIPY 493/503 staining suggested that exposure to MoS_(2)NSs and MoS_(2)-bulk decreased lipid levels in the spheroids. However, lipidomics data indicated that MoS_(2)materials only decreased 8 lipid classes, including lysophosphatidylcholine,lysodimethylphosphatidylethanolamine,N-acylethanolamine,ceramide phosphoethanolamines, gangliosides, lysosphingomyelin and sulfatide, whereas most of the lipid classes were indeed increased. In addition,MoS_(2)NSs was more potent to decrease the lipid classes compared with MoS_(2)-bulk.Combined,the results from this study showed that MoS_(2)NSs and bulk materials were non-cytotoxic but altered lipid profiles in 3D Caco-2 spheroids. | Min Xie Chaobo Huang Yongqi Liang Shuang Li Liping Sheng Yi Cao | 2022 | Chinese Chemical Letters2022,33,1: | 0
Wind farm active power dispatching algorithm based on Grey Incidence显示文摘This study proposes a wind farm active power dispatching(WFAPD) algorithm based on the grey incidence method, which does not rely on an accurate mathematical model of wind turbines. Based on the wind turbine start-stop data at different wind speeds, the weighting coefficients, which are the participation degrees of a variable speed system and a variable pitch system in power regulation, are obtained using the grey incidence method. The incidence coefficient curve is fitted by the B-spline function at a full range of wind speeds, and the power regulation capacity of all wind turbines is obtained. Finally, the WFAPD algorithm, which is based on the regulating capacity of each wind turbine, is compared with the wind speed weighting power dispatching(WSWPD) algorithm in MATLAB. The simulation results show that the active power fluctuation of the wind farm is smaller, the rotating speed of wind turbines is smoother, and the fatigue load of highspeed turbines is effectively reduced. | Binbin Zhang Mengxin Jia Chaobo Chen Kun Wang Jichao Li | 2023 | Global Energy Interconnection2023,6,2: | 0
