| 3 | The presence of phosphorylation form of D1 protein in its cross-linked aggregates in high light treated spinach leaves in vivo显示文摘In the present study, using specific antibody against D1 protein, we detected four ag- gregates of D1 protein in thylakoid membranes from spinach leaves illuminated at high light (800―2500 μmol photons·m?2·s?1) for 3 h. Their accumulations were dependent on the light intensity to which the leaves had been subjected. Further immunoblot analysis indicated that 70 kD aggregate was a prod- uct of D1 protein cross-linked with CP43, 65 and 60 kD aggregate were two cross-linked products be- tween D1 and D2 proteins, and 41 kD aggregate was one cross-linked D1 with α-subunit of cytochrome b559 (Cyt b559). This result provided the evidence for the existence of the aggregation of the D1 protein in vivo. The maximal level of D1/Cyt b559 aggregate occurred at 1000 μmol photons·m?2·s?1 but drastically decreased with further increasing light intensity. Im- munoblot analysis with phosphothreonine (Thr (P)) antibody indicated that D1/CP43 and D1/Cyt b559 aggregates contained the phosphorylated protein(s). In vitro dephosphorylation experiment also showed that D1/Cyt b559 and D1/CP43 aggregates lost the immunoreactivity with Thr (P) antibody after the phosphatase treatment of the membranes from high-light-illuminated leaves. In such applications, wide-baseline feature matching is a critical problem. In this paper, we apply randomized tree method to match key points extracted from the input image to those key frames as a classification problem. Extended Kalman filter is also utilized for jitter correction. A video see-through mobile augmented reality system is built for the on-site digital reconstruction of Yuanmingyuan Garden. | 6 | Recent developments in multimodality fluorescence imaging probes显示文摘Multimodality optical imaging probes have emerged as powerful tools that improve detection sensitivity and accuracy, important in disease diagnosis and treatment. In this review, we focus on recent developments of optical fluorescence imaging(OFI) probe integration with other imaging modalities such as X-ray computed tomography(CT), magnetic resonance imaging(MRI), positron emission tomography(PET), single-photon emission computed tomography(SPECT), and photoacoustic imaging(PAI). The imaging technologies are briefly described in order to introduce the strengths and limitations of each techniques and the need for further multimodality optical imaging probe development. The emphasis of this account is placed on how design strategies are currently implemented to afford physicochemically and biologically compatible multimodality optical fluorescence imaging probes. | 11 | PEPT1-mediated prodrug strategy for oral delivery of peramivir显示文摘Peramivir was a novel and highly potent neuraminidase(NA) inhibitor for the treatment of influenza A and B. However, it exhibited a very low oral bioavailability(only 3%) due to the high polarity(log P of-1.4) and the low membrane permeability across the intestine. To utilize the PEPT1-mediated prodrug strategy to improve the oral absorption and develop the oral alternative, seven amino acid ester prodrugs and seven amino acid amide prodrugs have been synthesized. The permeability of these prodrugs across Caco-2 cells were screened. Peramivr-(CH_2)_2-l-Val and Peramivir-l-Ile were of the highest permeability in ester prodrugs and amide prodrugs, respectively, and then they were selected for further studies. Glycylsarcosine(gly-sar) uptake by Caco-2 could be inbihited by Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile in a concentration-dependent manner, and the IC 50 was 1.34 ± 0.31 m M and 1.78 ± 0.48 m M, respectively. The direct uptake of Peramivir-(CH_2)_2-l-Val and Peramivirl-Ile in MDCK-PEPT1 cells were significantly higher than in MDCK mock cells, and could be markedly inhibited by gly-sar. The uptake of Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile(0.01 to 50 m M) in MDCK-hPEPT1 cells conformed to Michaelis–Menten Equation. The oral bioavailability of peramivir was 65.3% and 37.3% after the oral administration of Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile to rats, respectively. The oral absorption and bioactivation of Peramivir-(CH_2)_2-l-Val was rapid and extensive, and no Peramivir-(CH_2)_2-l-Val was found in plasma. Because the amide bond was relatively stable, Peramivir-l-Ile could not be totally converted to the parent drug in vivo. Peramivir-(CH_2)_2-l-Val with good oral profiles and rapid bioactivation might be a promising prodrug for the further clinic development. Cloud computing offers better stability and lower price than traditional IT facilities. Huge storage capacity is essential for video streaming service. More and more cloud providers appear so there are increasing cloud platforms to choose. A better choice is to use more than one data center, which is called multi-cloud. In this paper a closed-loop approach is proposed for optimizing Quality of Service (QoS) and cost. Modules of monitoring and controlling data centers are required as well as the application feedback such as video streaming services. An algorithm is proposed to help choose cloud providers and data centers in a multi-cloud environment as a video service manager. Performance with different video service workloads are evaluated. Compared with using only one cloud provider, dynamically deploying services in multi-cloud is better in aspects of both cost and QoS. | 13 | Mapping Human Pluripotent Stem Cell-derived Erythroid Differentiation by Single-cell Transcriptome Analysis显示文摘There is an imbalance between the supply and demand of functional red blood cells(RBCs)in clinical applications.This imbalance can be addressed by regenerating RBCs using several in vitro methods.Induced pluripotent stem cells(iPSCs)can handle the low supply of cord blood and the ethical issues in embryonic stem cell research,and provide a promising strategy to eliminate immune rejection.However,no complete single-cell level differentiation pathway exists for the iPSC-derived erythroid differentiation system.In this study,we used iPSC line BC1 to establish a RBC regeneration system.The 10X Genomics single-cell transcriptome platform was used to map the cell lineage and differentiation trajectory on day 14 of the regeneration system.We observed that iPSC differentiation was not synchronized during embryoid body(EB)culture.The cells(on day 14)mainly consisted of mesodermal and various blood cells,similar to the yolk sac hematopoiesis.We identified six cell classifications and characterized the regulatory transcription factor(TF)networks and cell-cell contacts underlying the system.iPSCs undergo two transformations during the differentiation trajectory,accompanied by the dynamic expression of cell adhesion molecules and estrogen-responsive genes.We identified erythroid cells at different stages,such as burst-forming unit erythroid(BFU-E)and orthochromatic erythroblast(ortho-E)cells,and found that the regulation of TFs(e.g.,TFDP1 and FOXO3)is erythroid-stage specific.Immune erythroid cells were identified in our system.This study provides systematic theoretical guidance for optimizing the iPSC-derived erythroid differentiation system,and this system is a useful model for simulating in vivo hematopoietic development and differentiation. | Zijuan Xin Wei Zhang Shangjin Gong Junwei Zhu Yanming Li Zhaojun Zhang Xiangdong Fang | 2021 | Genomics, Proteomics & Bioinformatics2021,19,3: | 2 |

Herein, we wish to report a coumarin-thiazolidine masked D-penicillamine based dual-modality fluorescent probe COU-DPA-1 for selective detection, differentiation, and detoxification of multiple heavy metal ions(Ag^(+), Hg^(2+), Cu^(2+)). The probe shows divergent fluorescence(FL)/circular dichroism(CD) responses via divergent bond-cleavage cascade reactions(metal ion promoted C-S cleavage and hydrolysis at two distinctive cleavage sites): FL “turn-off” and CD “turn-on” for Ag+(no hydrolysis), FL “turn-on” and CD “turn-off” for Hg^(+)(imine hydrolysis), and FL “self-threshold ratiometric” and CD “turn-off” for excess Cu^(2+)(lactone and imine hydrolysis), providing the first example of a fluorescence/CD dual-modality probe for multiple species with complimentary responses. | 18 | Resampling Factor Estimation via Dual-Stream Convolutional Neural Network显示文摘The estimation of image resampling factors is an important problem in image forensics.Among all the resampling factor estimation methods,spectrumbased methods are one of the most widely used methods and have attracted a lot of research interest.However,because of inherent ambiguity,spectrum-based methods fail to discriminate upscale and downscale operations without any prior information.In general,the application of resampling leaves detectable traces in both spatial domain and frequency domain of a resampled image.Firstly,the resampling process will introduce correlations between neighboring pixels.In this case,a set of periodic pixels that are correlated to their neighbors can be found in a resampled image.Secondly,the resampled image has distinct and strong peaks on spectrum while the spectrum of original image has no clear peaks.Hence,in this paper,we propose a dual-stream convolutional neural network for image resampling factors estimation.One of the two streams is gray stream whose purpose is to extract resampling traces features directly from the rescaled images.The other is frequency stream that discovers the differences of spectrum between rescaled and original images.The features from two streams are then fused to construct a feature representation including the resampling traces left in spatial and frequency domain,which is later fed into softmax layer for resampling factor estimation.Experimental results show that the proposed method is effective on resampling factor estimation and outperforms some CNN-based methods. | Shangjun Luo Junwei Luo Wei Lu Yanmei Fang Jinhua Zeng Shaopei Shi Yue Zhang | 2021 | Computers, Materials & Continua2021,,1: | 1 |
