4Transition of autophagy and apoptosis in fibroblasts depends on dominant expression of HIF-1αor p53显示文摘It has been revealed that hypoxia is dynamic in hypertrophic scars;therefore,we considered that it may have different effects on hypoxia-inducible factor-1α(HIF-1α)and p53 expression.Herein,we aimed to confirm the presence of a teeterboard-like conversion between HIF-1αand p53,which is correlated with scar formation and regression.Thus,we obtained samples of normal skin and hypertrophic scars to identify the differences in HIF-1αand autophagy using immunohistochemistry and transmission electron microscopy.In addition,we used moderate hypoxia in vitro to simulate the proliferative scar,and silenced HIF-1αor p53 gene expression or triggered overexpression to investigate the changes of HIF-1αand p53 expression,autophagy,apoptosis,and cell proliferation under this condition.HIF-1α,p53,and autophagy-related proteins were assayed using western blotting and immunofluorescence,whereas apoptosis was detected using flow cytometry analysis,and cell proliferation was detected using cell counting kit-8(CCK-8)and 5-bromo-2′-deoxyuridine(BrdU)staining.Furthermore,immunoprecipitation was performed to verify the binding of HIF-1αand p53 to transcription cofactor p300.Our results demonstrated that,in scar tissue,HIF-1αexpression increased in parallel with autophagosome formation.Under hypoxia,HIF-1αexpression and autophagy were upregulated,whereas p53 expression and apoptosis were downregulated in vitro.HIF-1αknockdown downregulated autophagy,proliferation,and p300-bound HIF-1α,and upregulated p53 expression,apoptosis,and p300-bound p53.Meanwhile,p53 knockdown induced the opposite effects and enhanced HIF-1α,whereas p53 overexpression resulted in the same effects and reduced HIF-1α.Our results suggest a teeterboard-like conversion between HIF-1αand p53,which is linked with scar hyperplasia and regression.Min LI Yidan SU Xiaoyuan GAO Jiarong YU Zhiyong WANG Xiqiao WANG 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,3:2
5Potential therapeutic effects and applications of Eucommiae Folium in secondary hypertension显示文摘Eucommiae Folium(EF),a traditional Chinese medicine,has been used to treat secondary hypertension,including renal hypertension and salt-sensitive hypertension,as well as hypertension caused by thoracic aortic endothelial dysfunction,a high-fat diet,and oxidized low-density lipoprotein.The antihypertensive components of EF are divided into four categories:flavonoids,iridoids,lignans,and phenylpropanoids,such as chlorogenic acid,geniposide acid and pinoresinol diglucoside.EF regulates the occurrence and development of hypertension by regulating biological processes,such as inhibiting inflammation,regulating the nitric oxide synthase pathway,reducing oxidative stress levels,regulating endothelial vasoactive factors,and lowering blood pressure.However,its molecular antihypertensive mechanisms are still unclear and require further investigation.In this review,by consulting the relevant literature on the antihypertensive effects of EF and using network pharmacology,we summarized the active ingredients and pharmacological mechanisms of EF in the treatment of hypertension to clarify how EF is associated with secondary hypertension,the related components,and underlying mechanisms.The results of the network pharmacology analysis indicated that EF treats hypertension through a multicomponent,multi-target and multi-pathway mechanism.In particular,we discussed the role of EF targets in the treatment of hypertension,including epithelial sodium channel,heat shock protein70,rhoassociated protein kinase 1,catalase,and superoxide dismutase.The relevant signal transduction pathways,the ras homolog family member A(RhoA)/Rho-associated protein kinase(ROCK)and nicotinamide adenine dinucleotide phosphate(NADPH)oxidase/eNOS/NO/Ca^(2+)pathways,are also discussed.Mengyuan Li Yanchao Zheng Sha Deng Tian Yu Yucong Ma Jiaming Ge Jiarong Li Xiankuan Li Lin Ma 2022Journal of Pharmaceutical Analysis2022,12,5:2

11Gas accumulation conditions and key technologies for exploration & development of Sinian and Cambrian gas reservoirs in Anyue gasfield显示文摘Exploration of the Sinian-Lower Paleozoic formations in the Sichuan Basin starts from the 1950s,and is tortuous and complex.To summarize the exploration experiences is of great significance of reference for exploration of oil and gas in deep carbonate rocks of the Sichuan Basin.Gas accumulation in the Anyue oversize gasfield is chiefly controlled by the Deyang-Anyue intra-platform rift and central Sichuan paleouplift.The intra-platform rift controls the scale hydrocarbon generation center and effective hydrocarbon plays.The Anyue gasfield is characterized by near-source hydrocarbon accumulation.The marginal zone of intra-platform rift and high part of the paleouplift control the scale distribution of favorable facies belts of reservoir development,and overlapped by paleokarstic reformation,the reservoirs are widely distributed.The intra-platform rift and paleouplift jointly control formation of the scale traps and hydrocarbon enrichment.Gas reservoir features and major controlling factors for hydrocarbon accumulation have some differences in the main play strata,such as Longwangmiao Formation,and Member 4 and Member 2 of Dengying Formation.The gas reservoir in the Longwangmiao Formation is a lithologic gas reservoir under the tectonic setting,and the inherited paleouplift is a key for reservoir formation and hydrocarbon accumulation.The gas reservoir in the Member 4 of Dengying Formation is mainly controlled by large structural-stratigraphic composite traps,and the gas reservoir in the Member 2 of Dengying Formation is the structural trap.Distribution of the large gasfields in the Dengying Formation is controlled by the intra-platform rift,near-source hydrocarbon-reservoir configuration controlled by the ancient erosion plane and stratigraphic sealing in the updip direction.For exploration and development of the Anyue gasfield,four series of technologies are developed,namely,reservoir seismic fine description technology,well logging fine evaluation technology of gas layers,fast drilling technology of complex strata,and stimulation technology for high temperature and high pressure gas layers.Technical progresses provide powerful guarantee for efficient exploration and development of the oversize gasfields.Yueming Yang Yu Yang Guang Yang Jiarong Song LongWen Chuanguang Deng Maolong Xia Qi Ran Guobin Duan Bing Luo Bing Xie 2018Petroleum Research2018,3,3:1
