1miR-200 family expression is downregulated upon neoplastic progression of Barrett's esophagus显示文摘AIM: To investigate miR-200 family expression in Barrett's epithelium, gastric and duodenal epithelia, and esophageal adenocarcinoma. METHODS: Real-time reverse transcriptase-polymerase chain reaction was used to measure miR-200, ZEB1 and ZEB2 expression. Ingenuity Pathway Analysis of miR-200 targets was used to predict biological outcomes. RESULTS: Barrett's epithelium expressed lower levels of miR-141 and miR-200c than did gastric and duodenal epithelia (P < 0.001). In silico analysis indicated roles for the miR-200 family in molecular pathways that distinguish Barrett's epithelium from gastric and duodenalepithelia, and which control apoptosis and proliferation. All miR-200 members were downregulated in adenocarcinoma (P < 0.02), and miR-200c expression was also downregulated in non-invasive epithelium adjacent to adenocarcinoma (P < 0.02). The expression of all miR-200 members was lower in Barrett's epithelium derived high-grade dysplastic cell lines than in a cell line derived from benign Barrett's epithelium. We observed signif icant inverse correlations between miR-200 family expression and ZEB1 and ZEB2 expression in Barrett's epithelium and esophageal adenocarcinoma (P < 0.05). 2The spatial distribution of major and trace elements of surface sediments in the northeastern Beibu Gulf of the South China Sea显示文摘A multi-index analysis including grain size, major and trace elements is performed on the surface sediments from the northeastern Beibu Gulf to trace the sources of the sediments and to understand the controlling factors for elements distribution. The mean grain size exhibits a wide variation ranging from 0.09Φ to 8.05Φ with an average value of 5.33Φ. The average contents of major elements descend in an order of c(SiO_2)>c(Al_2O_3)>c(Fe_2O_3)>c(CaO)>c(MgO)>c(K_2O)>c(Na_2O)>c(TiO_2)>c(P_2O_5)>c(MnO), while those of trace elements exhibit a descending order of c(Sr)>c(Rb)>c(V)>c(Zn)>c(Cr)>c(Pb)>c(Ni)>c(Cu)>c(As). On the basis of elementary distribution characteristics and statistical analyses, the study area is divided into the four zones: Zone I is located in the northeastern coastal area of the gulf, which receives large amount of fluvial materials from local rivers in Guangxi and Guangdong, China, and the Qiongzhou Strait; Zone Ⅱ is located in the center of the study area, where surface sediments exhibits a multiple source; Zone Ⅲ is located in the Qiongzhou Strait, where surface sediments are dominated by materials from the Zhujiang River and Hainan; Zone IV is located in the southwest of the study area, where surface sediments are mainly originated from the Red River and Hainan. 3Point-of-care testing in the diagnosis of gastrointestinal cancers: Current technology and future directions显示文摘Point-of-care(POC) tests enable rapid results and are well established in medical practice.Recent advances in analytical techniques have led to a new generation of POC devices that will alter gastrointestinal diagnostic pathways.This review aims to identify current and newtechnologies for the POC diagnosis of gastrointestinal cancer.A structured search of the Embase and Medline databases was performed.Papers reporting diagnostic tests for gastrointestinal cancer available as a POC device or containing a description of feasibility for POC application were included.Studies recovered were heterogeneous and therefore results are presented as a narrative review.Six diagnostic methods were identified(fecal occult blood, fecal proteins, volatile organic compounds, pyruvate kinase isoenzyme type M2, tumour markers and DNA analysis).Fecal occult blood testing has a reported sensitivity of 66%-85% and specificity greater than 95%.The others are at a range of development and clinical application.POC devices have a proven role in the diagnosis of gastrointestinal cancer.Barriers to their implementation exist and the transition from experimental to clinical medicine is currently slow.New technologies demonstrate potential to provide accurate POC tests and an ability to diagnose gastrointestinal cancer at an early stage with improved clinical outcome and survival.Jeremy R Huddy Melody Z Ni Sheraz R Markar George B Hanna 2015World Journal of Gastroenterology2015,21,14:2
4Muscle wasting in rheumatoid arthritis:The role of oxidative stress显示文摘Rheumatoid arthritis(RA), the commonest inflammatory arthritis, is a debilitating disease leading to functional and social disability. In addition to the joints, RA affects several other tissues of the body including the muscle. RA patients have significantly less muscle mass compared to the general population. Several theories have been proposed to explain this. High grade inflammation, a central component in the pathophysiology of the disease, has long been proposed as the key driver of muscle wasting. More recent findings however, indicate that inflammation on its own cannot fully explain the high prevalence of muscle wasting in RA. Thus, thecontribution of other potential confounders, such as nutrition and physical activity, has also been studied. Results indicate that they play a significant role in muscle wasting in RA, but again neither of these factors seems to be able to fully explain the condition. Oxidative stress is one of the major mechanisms thought to contribute to the development and progression of RA but its potential contribution to muscle wasting in these patients has received limited attention. Oxidative stress has been shown to promote muscle wasting in healthy populations and people with several chronic conditions. Moreover, all of the aforementioned potential contributors to muscle wasting in RA(i.e., inflammation, nutrition, and physical activity) may promote pro- or antioxidative mechanisms.
