| 2 | Sialic acid-engineered mesoporous polydopamine nanoparticles loaded with SPIO and Fe3+as a novel theranostic agent for T1/T2 dual-mode MRI-guided combined chemo-photothermal treatment of hepatic cancer显示文摘diagnostic and therapeutic capability are highly needed for the treatment of hepatic cancer.Herein,we aimed to develop a novel mesoporous polydopamine(MPDA)-based theranostic agent for T1/T2 dual magnetic resonance imaging(MRI)-guided cancer chemo-photothermal therapy.Superparamagnetic iron oxide(SPIO)-loaded MPDA NPs(MPDA@SPIO)was firstly prepared,followed by modifying with a targeted molecule of sialic acid(SA)and chelating with Fe^(3+)(SA-MPDA@SPIO/Fe^(3+) NPs).After that,doxorubicin(DOX)-loaded SA-MPDA@SPIO/Fe^(3+) NPs(SA-MPDA@SPIO/DOX/Fe^(3+))was prepared for tumor theranostics.The prepared SAPEG-MPDA@SPIO/Fe^(3+) NPs were water-dispersible and biocompatible as evidenced by MTT assay.In vitro photothermal and relaxivity property suggested that the novel theranostic agent possessed excellent photothermal conversion capability and photostability,with relaxivity of being r1=4.29 mM��1s��1 and r2=105.53 mM��1s��1,respectively.SAPEG-MPDA@SPIO/Fe^(3+) NPs could effectively encapsulate the DOX,showing dual pH-and thermal-triggered drug release behavior.In vitro and in vivo studies revealed that SA-MPDA@SPIO/DOX/Fe^(3+) NPs could effectively target to the hepatic tumor tissue,which was possibly due to the specific interaction between SA and the overexpressed E-selectin.This behavior also endowed SA-MPDA@SPIO/DOX/Fe^(3+)NPs with a more precise T1-T2 dual mode contrast imaging effect than the one without SA modification.In addition,SAPEG-MPDA@SPIO/DOX/Fe^(3+) NPs displayed a superior therapeutic effect,which was due to its active targeting ability and combined effects of chemotherapy and photothermal therapy.These results demonstrated that SAPEG-MPDA@SPIO/DOX/Fe^(3+) NPs is an effective targeted nanoplatform for tumor theranostics,having potential value in the effective treatment of hepatic cancer. | Gaofeng Shu Minjiang Chen Jingjing Song Xiaoling Xu Chenying Lu Yuyin Du Min Xu Zhongwei Zhao Minxia Zhu Kai Fan Xiaoxi Fan Shiji Fang Bufu Tang Yiyang Dai Yongzhong Du Jiansong Ji | 2021 | Bioactive Materials2021,6,5: | 4 |
| 3 | Interleukin-5 promotes ATP-binding cassette transporter A1 expression through miR-211/JAK2/STAT3 pathways in THP-1-dervied macrophages显示文摘Interleukin-5 (IL-5) is manifested as its involvement in the process of atherosclerosis, but the mechanism is still unknown. In this study, we explored the effect of IL-5 on lipid metabolism and its underlying mechanisms in THP-1-derived macrophages. The quantitative polymerase chain reaction (qPCR) and western blot analysis results showed that IL-5 significantly up-regulated ATP-binding cassette transporter A1 (ABCA1) expression in a dose-dependent and time-dependent manner. [3H]-labeled cholesterol was used to assess the levels of cholesterol efflux, and the results showed that IL-5 increased ABCA1-mediated cholesterol efflux. A high-performance liquid chromatography assay indicated that cellular cholesterol content was decreased by IL-5 treatment in THP-1-derived macrophages. The selective inhibitor and small interfering RNA were used to block the Janus kinase (JAK)/signal transducer and activator of transcription 3 (STAT3) pathway. The results of the qPCR and western blot analysis showed that IL-5 activated JAK2/STAT3 pathway to up-regulate ABCA1 expression. Meanwhile, IL-5 reduced the expression level of miR-211. Furthermore, we found that JAK2 is a target gene of miR-211 and miR-211 mimic inhibited the expression of JAK2 and reduced the levels of p-STAT3 and ABCA1 as revealed by luciferase reporter assay, qPCR and western blot analysis. | 6 | Tuning charge density of chimeric antigen receptor optimizes tonic signaling and CAR-T cell fitness显示文摘Tonic signaling of chimeric antigen receptor(CAR),i.e.,the spontaneous CAR activation in the absence of tumor antigen stimulation,is considered to be a pivotal event controlling CAR-T efficacy.However,the molecular mechanism underlying the spontaneous CAR signals remains elusive.Here,we unveil that positively charged patches(PCPs)on the surface of the CAR antigenbinding domain mediate CAR clustering and result in CAR tonic signaling.For CARs with high tonic signaling(e.g.,GD2.CAR and CSPG4.CAR),reducing PCPs on CARs or boosting ionic strength in the culture medium during ex vivo CAR-T cell expansion minimizes spontaneous CAR activation and alleviates CAR-T cell exhaustion.In contrast,introducing PCPs into the CAR with weak tonic signaling,such as CD19.CAR,results in improved in vivo persistence and superior antitumor function.These results demonstrate that CAR tonic signaling is induced and maintained by PCP-mediated CAR clustering.Notably,the mutations we generated to alter the PCPs maintain the antigen-binding affinity and specificity of the CAR.Therefore,our findings suggest that the rational tuning of PCPs to optimize tonic signaling and in vivo fitness of CAR-T cells is a promising design strategy for the nextgeneration CAR. | Jian Chen Shizhen Qiu Wentao Li Kun Wang Yu Zhang Han Yang Baichuan Liu Guangfei Li Li Li Min Chen Junjie Lan Jiahua Niu Peijie He Lei Cheng Gaofeng Fan Xin Liu Xianmin Song Chenqi Xu Haitao Wu Haopeng Wang | 2023 | Cell Research2023,33,5: | 2 |
| 7 | Tet2 Regulates Osteoclast Differentiation by Interacting with Runx1 and Maintaining Genomic 5-Hydroxymethylcytosine(5hmC)显示文摘As a dioxygenase, Ten-Eleven Translocation 2(TET2) catalyzes subsequent steps of 5-methylcytosine(5 mC) oxidation. TET2 plays a critical role in the self-renewal, proliferation,and differentiation of hematopoietic stem cells, but its impact on mature hematopoietic cells is not well-characterized. Here we show that Tet2 plays an essential role in osteoclastogenesis. Deletion of Tet2 impairs the differentiation of osteoclast precursor cells(macrophages) and their maturation into bone-resorbing osteoclasts in vitro. Furthermore, Tet2^(-/-) mice exhibit mild osteopetrosis, accompanied by decreased number of osteoclasts in vivo. Tet2 loss in macrophages results in the altered expression of a set of genes implicated in osteoclast differentiation, such as Cebpa, Mafb, and Nfkbiz. Tet2 deletion also leads to a genome-wide alteration in the level of 5-hydroxymethylcytosine(5 hmC) and altered expression of a specific subset of macrophage genes associated with osteoclast differentiation. Furthermore, Tet2 interacts with Runx1 and negatively modulates its transcriptional activity. Our studies demonstrate a novel molecular mechanism controlling osteoclast differentiation and function by Tet2, that is, through interactions with Runx1 and the maintenance of genomic 5 hmC. | 9 | In situ formation of ionically conductive nanointerphase on Si particles for stable battery anode显示文摘Silicon(Si)is a promising anode candidate for next-generation lithium-ion batteries(LIBs)due to its high theoretical capacity.Solar Si photovoltaic waste possesses good purity and high output.Using it as the raw material for battery anodes can synchronously solve the problem of solid waste pollution and enable high energy density LIBs.A critical issue impeding the practical application of Si is the undesirable side reactions at the electrolyte-particle interface and the resulting increase in impedance during cycling.Herein,a Si-P core shell structure with chemical bonding at the Si-P interface is fabricated through a simple mechanical alloying reaction between red P and solar Si photovoltaic waste.The P nanoshell with thickness within 15 nm converts to Li3P during the initial lithiation process and maintains its phase on cycling.The as-formed Li3P nanolayer functions as a stable,ionically conductive protective layer that reduces the direct contact between Si and electrolytes,and thus suppresses undesired side reactions.The Si-P nanocomposite exhibits stable electrochemical cycling with a high reversible capacity of 1,178 mAh g^(−1)after 500 cycles at 1,200 mA g^(−1),as well as excellent rate capability(912 mAh g^(−1)at 2 C).With 15 wt%addition to graphite,a graphite/Si-P hybrid electrode shows a high overall reversible specific capacity of 447 mAh g^(−1)and 88.3%capacity retention after 100 cycles at high areal capacity of 2.64 mAh cm^(−2) at 100 mA g^(−1),indicating its promise as a drop-in anode in practical LIBs. | Xiaoxue Chen Gaofeng Ge Wenyu Wang Bao Zhang Jianjun Jiang Xuelin Yang Yuzhang Li Li Wang Xiangming He Yongming Sun | 2021 | Science China Chemistry2021,64,8: | 2 |

| 15 | EAT-NAS: elastic architecture transfer for accelerating large-scale neural architecture search显示文摘Neural architecture search(NAS) methods have been proposed to relieve human experts from tedious architecture engineering. However, most current methods are constrained in small-scale search owing to the issue of huge computational resource consumption. Meanwhile, the direct application of architectures searched on small datasets to large datasets often bears no performance guarantee due to the discrepancy between different datasets. This limitation impedes the wide use of NAS on large-scale tasks. To overcome this obstacle, we propose an elastic architecture transfer mechanism for accelerating large-scale NAS(EATNAS).In our implementations, the architectures are first searched on a small dataset, e.g., CIFAR-10. The best one is chosen as the basic architecture. The search process on a large dataset, e.g., ImageNet, is initialized with the basic architecture as the seed. The large-scale search process is accelerated with the help of the basic architecture. We propose not only a NAS method but also a mechanism for architecture-level transfer learning. In our experiments, we obtain two final models EATNet-A and EATNet-B, which achieve competitive accuracies of 75.5% and 75.6%, respectively, on ImageNet. Both the models also surpass the models searched from scratch on ImageNet under the same settings. | 18 | lncRNA ELFN1-AS1 upregulates TRIM29 by suppressing miR-211-3p to promote gastric cancer progression显示文摘Long noncoding RNA(lncRNA)extracellular leucine rich repeat and fibronectin type III domain containing 1-antisense RNA 1(ELFN1-AS1)has been found to be upregulated in various tumors.However,the biological functions of ELFN1-AS1 in gastric cancer(GC)are not entirely understood.In the present study,the expression levels of ELFN1-AS1,miR-211-3p,and TRIM29 are determined using reverse transcription-quantitative PCR.Subsequently,CCK8,EdU,and colony formation assays are performed to determine GC cell vitality.The migratory and invasive capabilities of GC cells are further evaluated using transwell invasion and cell scratch assays.Western blot analysis is performed to quantify the levels of proteins associated with GC cell apoptosis and epithelialmesenchymal transition(EMT).The competing endogenous RNA(ceRNA)activity of ELFN1-AS1 on TRIM29 through miR-211-3p is confirmed by pull-down,RIP,and luciferase reporter assays.Our study proves that ELFN1-AS1 and TRIM29 are highly expressed in GC tissues.ELFN1-AS1 silencing inhibits GC cell proliferation,migration,invasion and EMT,and induces cell apoptosis.Rescue experiments reveal that the oncogenicity of ELFN1-AS1 is modulated by acting as a sponge for miR-211-3p,thereby increasing the expression of the target gene of miR-211-3p,TRIM29.In summary,ELFN1-AS1 maintains GC cell tumorigenicity via the ELFN1-AS1/miR-211-3p/TRIM29 axis,indicating that this axis can be directed for GC treatment in the future. | Jinxi Huang Weiwei Yuan Beibei Chen Gaofeng Li Xiaobing Chen | 2023 | Acta Biochimica et Biophysica Sinica2023,55,3: | 1 |
| 19 | Terahertz metalens of hyper-dispersion显示文摘Chromatic optical lenses have promising applications in three-dimensional imaging, which allows fast spectral tomography without mechanical moving parts. The scanning range of current chromatic optical lenses is limited by their dispersion ability. The recent development in metasurfaces provides ideal blocks for optical wavefront manipulation and dispersion engineering of artificial materials at sub-wavelength scales. Hyper-dispersive metalenses can be realized by utilizing dispersive meta-atoms, which have enhanced dispersion compared to regular diffractive lenses. This is critical for increasing the imaging depth of fast spectral tomography. In this work, a hyper-dispersive metalens is realized with a chromatic dispersion 1.76 times greater than that of a regular diffractive metalens in the THz frequency range of 2.40–2.61 THz by simultaneously controlling the frequencydependent phase, group delay(GD), and GD dispersion of the metalens.
