1Production of proinflammatory cytokines in the human THP-1 monocyte cell line following induction by Tp0751,a recombinant protein of Treponema pallidum显示文摘The tissue destruction characteristic of syphilis infection may be caused by inflammation due to Treponema pallidum and the ensuing immune responses to the pathogen.T.pallidum membrane proteins are thought to be potent inducers of inflammation during the early stages of infection.However,the actual membrane proteins that induce inflammatory cytokine production are not known,nor are the molecular mechanisms responsible for triggering and sustaining the inflammatory cascades.In the present study,Tp0751 recombinant protein from T.pallidum was found to induce the production of proinflammatory cytokines,including TNF-α,IL-1βand IL-6,in a THP-1 human monocyte cell line.The signal transduction pathways involved in the production of these cytokines were then further investigated.No inhibition of TNF-a,IL-1β,or IL-6 production was observed following treatment with the SAPK/JNK specific inhibitor SP600125 or with an ERK inhibitor PD98059.By contrast,anti-TLR2 mAb,anti-CD14 mAb,and the p38 inhibitor SB203580 significantly inhibited the production of all three cytokines.In addition,pyrrolidine dithiocarbamate (PDTC),a specific inhibitor of NF-κB,profoundly inhibited the production of these cytokines.Tp0751 treatment strongly activated NF-κB,as revealed by Western blotting.However,NF-κB translocation was significantly inhibited by treatment with PDTC.These results indicated that TLR2,CD14,MAPKs/p38,and NF-κB might be implicated in the inflammatory reaction caused by T.pallidum infection.LIU ShuangQuan1,2,WANG ShiPing1,WU YiMou3,ZHAO FeiJun3,ZENG TieBing3,ZHANG YueJun3,ZHANG QiuGui2 & GAO DongMei1 1 Department of Parasitology,Xiangya School of Medicine,Central South University,Changsha 410078 ,China 2 The First Affiliated Hospital,University of South China,Hengyan 421001,China 3 Department of Microbiology and Immunology,University of South China,Hengyan 421001,China 2010Science China(Life Sciences)2010,53,2:28
2Total alkyl dibenzothiophenes content tracing the filling pathway of condensate reservoir in the Fushan Depression,South China Sea显示文摘The condensates are generally characterized by high maturity,low concentration of steranes and ter-panes biomarkers and low content of non-hydrocarbon fraction. In this case commonly used steranes,terpanes and carbazoles parameters cannot be effectively applied in the reservoir-filling tracing. The hydrogen bond formed by sulfur atom in the dibenzothiophenes (DBTs) results in molecule adsorption and fractionation during oil migration in reservoir. Like carbazoles,total DBTs content decreases with the increasing of oil migration distance. Therefore,a new parameter——total DBTs content is proposed to be used to trace the oil migration orientation and filling pathway. In present study,total DBTs con-tents of condensates and light oils are obtained by adding internal standard——eight deuterium atoms substituted DBT during Gas Chromatography-Mass Spectrometry analysis of aromatic fraction. Except for a few samples with much lower content of non-hydrocarbon fraction,the total DBTs content shows a fine positive correlation with that of carbazoles. Large errors can be caused in the process of pyrrolic nitrogen compounds separation. The application of this new parameter in the Fushan Depression of Beibu Gulf Basin,South China Sea indicates that this parameter is a reliable one to trace filling pathway in condensate reservoirs. Combined with other DBTs-related parameters,such as 4-/1-methydibenzo-thiophene and 2,4-/1,4-dimethyldibenzothiophene,oil migration orientation and filling pathway of the Fushan Depression was determined. The accumulations of Huachang oil field in the Fushan Depres-sion are mainly migrated and charged from northeast to southeast along the Huachang uplift. It can be predicated that the light oil and condensates in the Huachang oil field should be sourced from the source kitchen at the Bailian Sag. 3AtHsfA2 modulates expression of stress responsive genes and enhances tolerance to heat and oxidative stress in Arabidopsis显示文摘There is increasing evidence for considerable interlinking between the responses to heat stress and oxidative stress, and recent researches suggest heat shock transcription factors (Hsfs) play an important role in linking heat shock with oxidative stress signals. In this paper, we present evidence that AtHsfA2 modulated expression of stress responsive genes and enhanced tolerance to heat and oxidative stress in Arabidopsis. Using Northern blot and quantitative RT-PCR analysis, we demonstrated that the expression of AtHsfA2 was induced by not only HS but also oxidative stress. By functional analysis of AtHsfA2 knockout mutants and AtHsfA2 overexpressing transgenic plants, we also demonstrated that the mutants displayed reduced the basal and acquired thermotolerance as well as oxidative stress tolerance but the overexpression lines displayed increased tolerance to these stress. The phenotypes correlated with the expres-sion of some Hsps and APX1, ion leakage, H2O2 level and degree of oxidative injuries. These results showed that, by modulated expression of stress responsive genes, AtHsfA2 enhanced tolerance to heat and oxidative stress in Arabidopsis. 4Study of ozone “weekend effect” in Shanghai显示文摘Analysis of observed ozone data in 2006 from five monitoring sites (Xujiahui, Chongming, Baoshan, Pudong, Jinshan) in Shanghai reveals that ozone (O_3) concentrations in Xujiahui are higher at week-ends than those on weekdays, despite the fact that emissions of ozone precursor substances, such as oxides of nitrogen (NOx), carbon monoxide (CO) and volatile organic compounds (VOCs) are lower at weekends than those on weekdays. The possible chemical cause of ozone 'weekend effect' is that NO_2/NO ratio increases at weekends by 25.61% compared with those on weekdays. In addition, because of an average 12.13% reduction in NOx (NO + NO_2) in the early morning (05:00-09:00) at weekends compared with that on weekdays, the ozone inhibition period ends 0.5 h earlier at weekends resulting in the longer duration of ozone accumulation and the higher ozone production rate. The rate of ozone production is a function of VOCs and NOx in the atmosphere. VOCs/NOx ratio in Xujiahui is 4.55 at weekends, and 4.37 on weekdays, belonging to the 'NOx-limited'. The increasing VOCs/NOx ratio at weekends leads to ozone enhancement from 73 ppbv to 80 ppbv, which are consistent with ozone 'weekend effect' in Xujiahui. 5Geochemistry of high-Mg andesites from the early Cretaceous Yixian Formation, western Liaoning: Implications for lower crustal delamination and Sr/Y variations显示文摘85 volcanic rocks of the Yixian Formation from the Sihetun type section were collected and analyzed for geochemical and isotopic compositions. Major element compositions indicate that the Sihetun volcanic rocks are high magnesium andesites with some basalts occurring at the bottom of section. The Sihetun high magnesium andesites (SiO2 = 52.82―59.31wt%, Al2O3 = 14.15―16.35wt%) show many characteristics of adakites such as depletion in heavy rare-earth elements (HREE; Yb = 1.03―1.88 μg/g) and Y(12―20 μg/g) and high Sr (620―1323 μg/g) and Sr/Y(32―88), with high LaN/YbN ratio (10―25). They share similar major and trace element characteristics to volcanic rocks from the Xinglonggou Formation except their lower Nd isotope ratios (143Nd/144Nd (130Ma) = 0.5118―0.5119, ε Nd (130Ma) = ?11.6―?13.8, 87Sr/86Sr (130 Ma) = 0.7058―0.7064. They were in- terpreted in a way that eclogite that formed at the base of thickened Archean lower crust of the North China craton foundered into the convecting mantle and subsequently melted and interacted with peridotite. However, compared to the Xinglonggou volcanic rocks, the source of the Sihetun magma contained more ancient continental crustal material in order to explain its evolved Nd isotopes. The age of the Sihetun Formation was 120 to 130 Ma, and this indicates that delamination lasted to the early Cretaceous period. The Sr contents and Sr/Y ratios of the Sihetun high-Mg andesites show sig- nificant negative correlations with SiO2 for samples with SiO2 > 56%. These suggest that the Sr and Sr/Y values were reduced due to fractional crystallization of plagioclase. 6Forecasting the summer rainfall in North China using the year-to-year increment approach显示文摘A new approach to forecasting the year-to-year increment of rainfall in North China in July-August (JA) is proposed. DY is defined as the difference of a variable between the current year and the preceding year (year-to-year increment). NR denotes the seasonal mean precipitation rate over North China in JA. After analyzing the atmospheric circulation anomalies associated with the DY of NR, five key predictors for the DY of NR have been identified. The prediction model for the DY of NR is established by using multi-linear regression method and the NR is obtained (the current forecasted DY of NR added to the preceding observed NR). The prediction model shows a high correlation coefficient (0.8) between the simulated and the observed DY of NR throughout period 1965-1999, with an average relative root mean square error of 19% for the percentage of precipitation rate anomaly over North China. The prediction model makes a hindcast for 2000-2007, with an average relative root mean square error of 21% for the percentage of precipitation rate anomaly over North China. The model reproduces the downward trend of the percentage of precipitation rate anomaly over North China during 1965-2006. Because the current operational prediction models of the summer precipitation have average forecast scores of 60%-70%, it has been more difficult to forecast the summer rainfall over North China. 10Soil organic carbon storage and soil CO_2 flux in the alpine meadow ecosystem显示文摘High-resolution sampling,measurements of organic carbon contents and 14C signatures of selected four soil profiles in the Haibei Station situated on the northeast Tibetan Plateau,and application of 14C tracing technology were conducted in an attempt to investigate the turnover times of soil organic car-bon and the soil-CO2 flux in the alpine meadow ecosystem. The results show that the organic carbon stored in the soils varies from 22.12×104 kg C hm-2 to 30.75×104 kg C hm-2 in the alpine meadow eco-systems,with an average of 26.86×104 kg C hm-2. Turnover times of organic carbon pools increase with depth from 45 a to 73 a in the surface soil horizon to hundreds of years or millennia or even longer at the deep soil horizons in the alpine meadow ecosystems. The soil-CO2 flux ranges from 103.24 g C m-2 a-1 to 254.93 gC m-2 a-1,with an average of 191.23 g C m-2 a-1. The CO2 efflux produced from microbial decomposition of organic matter varies from 73.3 g C m-2 a-1 to 181 g C m-2 a-1. More than 30% of total soil organic carbon resides in the active carbon pool and 72.8%―81.23% of total CO2 emitted from or-ganic matter decomposition results from the topsoil horizon (from 0 cm to 10 cm) for the Kobresia meadow. 11Primary scientific results of Chang'E-1 lunar mission显示文摘The strategic plan for the development of the unmanned Chinese Lunar Exploration Program is characterized by three distinct stages: 'orbiting around', 'landing on' and 'returning from' the Moon. The first Chinese lunar probe, Chang'E-1, which was successfully launched on October 24th, 2007 at Xichang Satellite Launch Center, and guided to crash on the Moon on March 1st, 2009, at 52.36°E, 1.50°S, in the north of Mare Fecunditatis, is the first step towards the 'orbiting around' stage. The Chang'E-1 mission lasted 495 days, exceeding the expected life-span by about four months. A total of 1.37 TB raw data was received from Chang'E-1. It was then processed into 4 TB scientific data products at various levels. Many scientific results have been obtained by analyzing these data, including especially the 'global lunar image from the first Chinese lunar explora- tion mission'. All scientific goals of Chang'E-1 have been achieved. It provides much useful materials for further advances of lunar sciences and planetary chemistry. 12Characteristics of pCO_(2) in surface water of the Bering Abyssal Plain and their effects on carbon cycle in the western Arctic Ocean显示文摘Characteristics of the pCO2 distribution in surface water of the Bering Abyssal Plain and their relationships with the ambient hydrological conditions were discussed using variations of the partial pressure of CO2 in surface water of the Bering Abyssal Plain and the Chukchi Sea. Data in this study are from a field investigation during the First Chinese National Arctic Research Expedition in 1999. Compared to the high productivity in the Bering Continental Shelf, much lower levels of chlorophyll a were observed in the Bering Abyssal Plain. The effect of hydrological factors on the pCO2 distribution in surface seawater of the Plain in summer has become a major driving force and dominated over biological factors. The Plain also presents a High Nutrient Low Chlorophyll (HNLC). In addition, the pCO2 distribution in the Bering Abyssal Plain has also been found to be influenced from the Bering Slope Current which would transform to the Anadyr Cur- rent when it inflows northwestward over the Plain. The Anadyr Current would bring a high nutrient water to the western Arctic Ocean where local nutrients are almost depleted in the surface water during the summer time. Resupplying nutrients would stimulate the growth of phytoplankton and enhance capacity of absorbing atmospheric CO2 in the surface water. Otherwise, in the Bering Sea the dissolved inorganic carbon brought from freshwater are not deposited down to the deep sea water but most of them would be transported into the western Arctic Ocean by the Alaska Coastal Current to form a carbon sink there. 15HJ-1A HSI on-orbit radiometric calibration and validation research显示文摘The calibration experiment data at Dunhuang radiometric calibration site in October, 2008 were used to achieve the on-orbit radiometric calibration for HJ-1A hyper spectral imager (HSI). Two other field experiments data were used to validate the Dunhuang calibration results. One field experiment took place in Inner-Mongolia, China in September, 2008, and the other field experiment took place in Lake Frome, Australia in February, 2009. Finally, the ‘confidence interval of calibration error’ concept was put forward for quantitatively computing the calibration coefficient error confidence interval. The results showed that the Dunhuang calibration results in 2008 had high reliability. 16Recombinant expression of rt-PA gene (encoding Reteplase) in gametophytes of the seaweed Laminaria japonica (Laminariales,Phaeophyta)显示文摘The life cycle of seaweed Laminaria japonica involves a generation alternation between diploid sporophyte and haploid gametophte. The expression of foreign genes in sporophte has been proved. In this research, the recombinant expression in gametophyte was investigated by particle bombardment with the rt-PA gene encoding the recombinant human tissue-type plasminogen activator (Reteplase), which is a thrombolytic agent for acute myocardial infarction (AMI). Transgenic gametophytes were selected by their resistance to herbicide phosphiothricin (PPT), and proliferated in an established bubble column photo-bioreactor. 19Geochemistry of ophiolite cherts from the Qinling orogenic belt and implications for their tectonic settings显示文摘Paleozoic cherts from the Mianl黣 and the Erlangping ophiolite zones of the Qinling orogenic belt are characterized by low Si/Al ratios (52.14-683.52 in the Mianle cherts, 12.29-58.62 in the Erlangping cherts), Fe2O3 (0.01-0.35 and 0.02-1.24) and high Al2O3/(Al2O3+Fe2O3) ratios (0.82-0.99 and 0.83-0.99). The negative correlation between Si2O and Al2O3 in the cherts reflects the important role of terrigenous components. The Erlangping cherts have Lan/Cen=0.9-1.15 and Ce/Ce*=0.95-1.15 with low contents of V, Ni and Cu, consistent with those of cherts forming on the continental margin. In contrast, the Ce/Ce* ratios of the Mianle cherts range from 0.71 to 1.18 and Lan/Cen from 0.88 to 1.43 with slightly high V, Ni and Cu, which are similar to cherts found in the mid-ocean ridges and pelagic basins. 20Arabian Peninsula-North Pacific Oscillation and its association with the Asian summer monsoon显示文摘Using correlation and EOF analyses on sea level pressure from 57-year NCEP-NCAR reanalysis data, the Arabian Peninsula-North Pacific Oscillation (APNPO) is identified. The APNPO reflects the co-variability between the North Pacific high and South Asian summer monsoon low. This teleconnec- tion pattern is closely related to the Asian summer monsoon. On interannual timescale, it co-varies with both the East Asian summer monsoon (EASM) and South Asian summer monsoon (SASM); on decadal timescale, it co-varies with the EASM: both exhibit two abrupt climate changes in the middle 1960s and the late 1970s respectively. The possible physical process for the connections between the APNPO and Asian summer monsoon is then explored by analyzing the APNPO-related atmospheric circulations. The results show that with a strong APNPO, the Somali Jet, SASM flow, EASM flow, and South Asian high are all enhanced, and an anomalous anticyclone is produced at the upper level over northeast China via a zonal wave train. Meanwhile, the moisture transportation to the Asian monsoon regions is also strengthened in a strong APNPO year, leading to a strong moisture convergence over India and northern China. All these changes of circulations and moisture conditions finally result in an anoma- lous Asian summer monsoon and monsoon rainfall over India and northern China. In addition, the APNPO has a good persistence from spring to summer. The spring APNPO is also significantly corre- lated with Asian summer monsoon variability.
