12晚期宫颈癌和盆腔淋巴结中高危HPV状态的预后意义显示文摘Objective. In this study, we investigated the presence of high- risk (HR) HPV types most prevalent in the Hungarian population in surgically removed cervical cancers and pelvic lymph nodes. The aim of our work was to determine the prognostic significance of HPV status in the lymph nodes draining the tumor. Methods. Primary tumor specimens from 150 patients and 900 lymph node samples (six per case) were studied. Fifty- six/150 were early (FIGO IA- IB) stage, while 94/150 were advanced (FIGO IIA- IIIB) stage cancers. Beside histopathological evaluation, DNA extracted from the tissue samples was subjected to nested PCR to detect characteristic type-specific sequences of HPVs 16, 18 and 33. Moreover, clinicopathological data were collected for an average 48- month postoperative follow-up period for the purposes of statistical analysis. Results. The presence of HR- HPV types in the lymph nodes shows no correlation with disease- free survival, whereas the presence of lymph node metastases significantly decreases life expectancy (P = 0.002). Lymph nodes with metastases more frequently carry HR- HPV than nodes with no evidence of tumorous infiltration (65% versus 36% , P < 0.001); however, a high number of metastases surrounding HR- HPV-positive tumors were found negative for the viruses (42/120). Conclusions. HR- HPV status of pelvic lymph nodes draining cervical cancers has no noticeable influence on the life expectancy of the patients. HR- HPV- positive tumor cells do not necessarily have a selective advantage in forming metastases. Presumably, a number of alterations in cellular genes rather than the presence of papillomavirus DNA may have a decisive role in the progression of cervical cancers.Fü le T. Csapó Z. Má thé M. I. 13Abscisic Acid Connects Phytohormone Signaling with RNA Metabolic Pathways and Promotes an Antiviral Response that Is Evaded by a Self- Controlled RNA Virus显示文摘A complex network of cellular receptors,RNA targeting pathways,and small-molecule signaling provides robust plant immunity and tolerance to viruses.To maximize their fitness,viruses must evolve control mechanisms to balance host immune evasion and plant-damaging effects.The genus Potyvirus comprises plant viruses characterized by RNA genomes that encode large polyproteins led by the P1 protease.A P1 autoinhibitory domain controls polyprotein processing,the release of a downstream functional RNAsilencing suppressor,and viral replication.Here,we show that P1Pro,a plum pox virus clone that lacks the P1 autoinhibitory domain,triggers complex reprogramming of the host transcriptome and high levels of abscisic acid(ABA)accumulation.A meta-analysis highlighted ABA connections with host pathways known to control RNA stability,turnover,maturation,and translation.Transcriptomic changes triggered by P1Pro infection or ABA showed similarities in host RNA abundance and diversity.Genetic and hormone treatment assays showed that ABA promotes plant resistance to potyviral infection.Finally,quantitative mathematical modeling of viral replication in the presence of defense pathways supported self-control of polyprotein processing kinetics as a viral mechanism that attenuates the magnitude of the host antiviral response.Overall,our findings indicate that ABA is an active player in plant antiviral immunity,which is nonetheless evaded by a self-controlled RNA virus.Fabio Pasin Hongying Shan Beatriz García Maren Müller David San León Márta Ludman David H.Fresno Károly Fátyol Sergi Munné-Bosch Guillermo Rodrigo Juan Antonio García 2020Plant Communications2020,1,5:1
14Prevalence of Schistosoma mono- and co-infections with multiple common parasites and associated risk factors and morbidity profile among adults in the Taabo health and demographic surveillance system, South-Central Côte d’Ivoire显示文摘Background Schistosomiasis remains an important public health problem,also among adults,and infected individuals not treated serve as a reservoir for continued transmission.Despite this fact,evidence on the epidemiology of schistosomiasis in adults in Côte d’Ivoire is scanty.This study aimed to determine the prevalence and risk factors of Schistosoma infection and co-infection with other helminth species and Plasmodium among adults in the Taabo region in the south-central part of Côte d’Ivoire.Methods A cross-sectional survey was carried out in April and May 2017 in the frame of the“Côte d’Ivoire Dual Burden of Disease Study”(CoDuBu).A total of 901 randomly selected individuals,aged 18–90 years,provided blood,stool and urine samples for the diagnosis of malaria and helminth infections.Stool samples were subjected to the Kato-Katz technique for detection of Schistosoma mansoni and soil-transmitted helminth eggs,while urine samples were examined for eggs of Schistosoma haematobium and circulating cathodic antigen of S.mansoni.Risk factors and morbidity profiles were assessed using health examination and questionnaires.Multinomial logistic regressions were employed to identify risk factors and morbidity patterns associated with S.mansoni mono-and co-infections.Results The prevalence of S.mansoni and S.haematobium was 23.2%and 1.0%,respectively.Most S.mansoni were mono-infections(81.3%).Independent determinants of S.mansoni infection were young age,low socioeconomic status(mono-and co-infection)and poor hygiene practices(co-infection)(P<0.05).S.mansoni infection was independently associated with higher pain and symptom scores(mono-infection),poor self-rated health and low healthcare use(co-infection)(P<0.05).Conclusions This study showed that adults represent a substantial reservoir of S.mansoni.To sustain schistosomiasis control and improve people’s wellbeing,it is important to expand preventive chemotherapy from school-aged children to adults,coupled with hygiene and health education.Fidèle K.Bassa Ikenna C.Eze Rufin K.Assaré Clémence Essé Siaka Koné Félix Acka Véronique Laubhouet-Koffi Dinard Kouassi Bassirou Bonfoh Jürg Utzinger Eliézer K.N’Goran 2022Infectious Diseases of Poverty2022,11,1:0
