| 2 | Clinicopathological, immunohistochemical, and ultrastructural study of 13 cases of melanotic schwannoma显示文摘Background Melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cellswith ultrastructural features of schwann cells.The description of the course of the tumors differs somewhat, but itis generally considered as a benign lesion.We investigated the clinicopathologic features, immunophenotypes,and ultrastructural features of 13 patients with nonpsammomatous melanotic schwannoma (NPMS).Methods Tumor specimens of each patient were sectioned and stained with hematoxylin-eosin, Fontana-Masson, Prussian blue, and periodic acid-Schiff (PAS).Immunohistochemical markers such as S-100, Leu-7,HMB-45, Melan-A, CK, EMA, vimentin, GFAP, laminin, collagenⅣ and MIB-1 were detected with theEnvision immunohistochemical staining method.Four of the cases were observed by electron microscopy.Results Of the 13 patients, 8 were male and 5 female, aged from 11 to 92 years (mean, 38.6 years).Thetumor sites included the spinal nerve root (5 patients), cranial nerve (1), greater omentum (1), subcutaneoustissue (3), mesentery (1), bone (1) and mediastinum(1).Eleven patients were followed up for over2 years,with a mean of 5.9 years.One patient (9.1%) with a primary tumor in the greater omentum developed anotherprimary tumor of the same type in the subcutaneous tissue of the abdominal wall after the first operation.Localrecurrence of the tumor was seen in2 patients (18.2%).One patient (9.1%) showed the local recurrence andmetastasis.Seven patients (63.6%) showed no evidence of the recurrence or metastasis.Grossly, all tumorswere well-circumscribed and the gross findings were suggestive of melanin-containing tumors.The tumor wascomposed of spindled and epithelioid cells with abundant intracytoplasmic melanin pigments.Nuclei were roundand contained delicate, evenly distributed chromatins as well as small, distinct nucleoli.In some areas, thenucleoli were large and prominent.Rare mitoses were seen in mostlesions exceptthe larger omentumlesion.Thepigment was shown to be positive for the Fontana-Masson and negative for Prussian blue and PAS.Immunohistochemical staining for S-100, Leu-7, HMB-45, Melan-A, and vimentin were strongly positive.Linear immunoreactions of both laminin and collagenⅣ was detected in all patients.Ultrastructurally, numerouselongated tumor-cell processes, duplicated basement membrane and melanosomes were observed in alldevelopmental stages.Conclusions Histologically, melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cells with ultrastructural features ofschwann cells.Distinguishing between this tumor andmalignant melanoma is of paramount importance inplanning of management. Immunohistochemically,combined use of laminin and collagenⅣ is valuablein distinguishing melanotic schwannoma frommalignant melanoma. Wide local resection andadditional radiotherapy should be advocated.Furtherstudies including cytogenetic or molecular biology arestill required to better delineate melanoticschwannoma from malignant melanoma. | 3 | A gene expression estimator of intramuscular fat percentage for use in both cattle and sheep显示文摘Background:The expression of genes encoding proteins involved in triacyglyceride and fatty acid synthesis and storage in cattle muscle are correlated with intramuscular fat(IMF)%.Are the same genes also correlated with IMF%in sheep muscle,and can the same set of genes be used to estimate IMF%in both species?Results:The correlation between gene expression(microarray) and IMF%in the longissimus muscle(LM) of twenty sheep was calculated.An integrated analysis of this dataset with an equivalent cattle correlation dataset and a cattle differential expression dataset was undertaken.A total of 30 genes were identified to be strongly correlated with IMF%in both cattle and sheep.The overlap of genes was highly significant,8 of the 13 genes in the TAG gene set and 8 of the 13 genes in the FA gene set were in the top 100 and 500 genes respertively most correlated with IMF%in sheep,P-value = 0.Of the 30 genes,CIDEA,THRSP,ACSM1,DGAT2 and FABP4 had the highest average rank in both species.Using the data from two small groups of Brahman cattle(control and Hormone growth promotant-treated[known to decrease IMF%in muscle]) and 22 animals in total,the utility of a direct measure and different estimators of IMF%(ultrasound and gene expression) to differentiate between the two groups were examined.Directly measured IMF%and IMF%estimated from ultrasound scanning could not discriminate between the two groups.However,using gene expression to estimate IMF%discriminated between the two groups.Increasing the number of genes used to estimate IMF%from one to five significantly increased the discrimination power;but increasing the number of genes to 15 resulted in little further improvement.Conclusion:We have demonstrated the utility of a comparative approach to identify robust estimators of IMF%in the LM in cattle and sheep.We have also demonstrated a number of approaches(potentially applicable to much smaller groups of animals than conventional methods) to using gene expression to rank animals for IMF%within a single farm/treatment,or to estimate differences in IMF%between two farms/treatments. | Bing Guo Kritaya Kongsuwan Paul L Greenwood Guanghong Zhou Wangang Zhang Brian P Dalrymple | 2014 | Journal of Animal Science and Biotechnology2014,5,4: | 6 |

| 5 | Molecular dynamics simulation and continuum modelling of granular surface flow in rotating drums显示文摘Numerical simulations of granular flows in rotating drums operated at medium to high rates (Fr=0.1― 0.2) have been carried out by using a Molecular Dynamics (MD) algorithm that incorporates inelastic particle interactions, sliding friction and rolling friction. The results indicate that the behavior of granular flow in rotating drums can be classified into two distinct zones: a shear active layer at the bed surface and a quasi-static plug flow region adjacent to the wall. The residence time of a tracer particle in the active layer is approximately a third or a half of that in the plug flow region. The thickness of the active layer at mid-chord is about 0.57―0.61 times that of the plug flow region. It is found that all cases simulated in this work are in the rolling-cascading intermediate regime instead of the pure rolling re-gime. The simulated tangential velocity at the mid-chord is also compared with experimental results reported in the literature and good agreement has been obtained. Based on the MD simulations and experimental results, a continuum approach has also been developed. It is shown that the behavior of granular solids in the plug flow region experiences plastic deformation along the radial direction from the wall with the velocity profiles well described by an exponential function, whereas the active layer velocity follows a simple expression for the Couette shear flow. | 6 | Spatial and historical distribution of organic phosphorus driven by environment conditions in lake sediments显示文摘The chemistry of sedimentary organic phosphorus(OP) and its fraction distribution in sediments are greatly influenced by environmental conditions such as terrestrial inputs and runoffs. The linkage of OP with environmental conditions was analyzed on the basis of OP spatial and historical distributions in lake sediments. The redundancy analysis and OP spatial distribution results suggested that both Na OH-OP(OP extracted by Na OH) and Re-OP(residual OP) in surface sediments from the selected 13 lakes reflected the gradient effects of environmental conditions and the autochthonous and/or allochthonous inputs driven by latitude zonality in China. The lake level and salinity of Lake Hulun and the runoff and precipitation of its drainage basin were reconstructed on the basis of the geochemistry index. This work showed that a gradient in weather conditions presented by the latitude zonality in China impacts the OP accumulation through multiple drivers and in many ways.The drivers are mainly precipitation and temperature, governing organic matter(OM)production, degradation rate and transportation in the watershed. Over a long temporal dimension(4000 years), the vertical distributions of Re-OP and Na OH-OP based on a dated sediment profile from HLH were largely regulated by the autochthonous and/or allochthonous inputs, which depended on the environmental and climate conditions and anthropogenic activities in the drainage basin. | 7 | The comparison of ensemble or deterministic dispersion modeling on global dispersion during Fukushima Dai-ichi nuclear accident显示文摘Ensemble forcasting,originally developed for weather prediction,is lately being extended to atmospheric dispersion applications,which is a new,effective methodology for improving the atmospheric dispersion numerical modeling.In March 2011,due to the massive 9.0 earthquakes and ensuing tsunami that struck off the northern coast of the island of Honshu,the Fukushima Nuclear Plant I had the substantial leak of radioactive materials into surrounding environment and atmosphere.To aim at the global dispersion modeling of atmospheric radionuclides from Fukushima Nuclear Accident,this paper presents two approaches of atmospheric dispersion forecasting:ensemble dispersion modeling(EDM) and deterministic dispersion modeling(DDM),conducts the globally dispersion modeling cases for Fukushima nuclear accident,and analyzes and evaluates the simulation results using observation data.In this paper,EDM includes three different perturbation methods:meteorological perturbation method,turbulence perturbation method,and physical parameterization ensemble forecasting method.The simulation results show that the trajectories from EDM have a better performance,which is in better agreement with the atmospheric circulation and observation data; the spread from DDM is slower and not as far as EDM.Additionally,the results from EDM display a better performance in the modeling of transport from Japan to China East Sea on April 4.The reasons for these results are:the techniques of MET and TUR are performed by adding perturbations on mean wind and turbulent velocity,respectively; the various different flow fields will result in far spreading in horizontal and the simulation results closer to observation; PHY is performed by using different diffusion physical parameterizations and produces the perturbations on vertical wind,which results the spreading in smaller range and discontinuous in horizontal.Finally,the comparative analysis between modeling results and observation data shows that all cases results are in good agreement with trends of observed radionuclides surface concentration; however,the modeling surface concentration is smaller than observation,especially in DDM and PHY.Furthermore,the EDM results show that MET and TUR are of more evolutionary advantage than PHY in modeling of average and maximum concentration.Therefore,this study can serve as a reference to atmospheric dispersion and environmental emergency response(EER). | SHENG Li SONG ZhenXin HU JiangKai Lü Kai TONG Hua LI Bing QIAO QingDang | 2015 | Science China Earth Sciences2015,58,3: | 3 |
| 8 | Effects of the activities of key enzymes involved in starch biosynthesis on the fine structure of amylopectin in developing rice (Oryza sativa L.) endosperms显示文摘The dynamic changes of the activities of enzymes involving in starch biosynthesis, including ADP-glucose pyrophosphorylase (AGPase), soluble starch synthases (SSS), starch branching enzyme (SBE) and starch debranching enzymes (DBE) were studied, and changes of fine structure of amy- lopectin were characterized by isoamylase treatment during rice grain development, using trans anti-waxy gene rice plants. The relationships between the activities of those key enzymes were also analyzed. The amylose synthesis was significantly inhibited in transgenic Wanjing 9522, but the total starch content and final grain weight were less affected as compared with those of non-transgenic Wanjing 9522 rice cultivar. Analyses on the changes of activities of enzymes involving in starch bio- synthesis showed that different enzyme activities were expressed differently during rice endosperm development. Soluble starch synthase is relatively highly expressed in earlier stage of endosperm de- velopment, whilst maximal expression of granule-bound starch synthase (GBSS) occurred in mid-stage of endosperm development. No obvious differences in changes of the activities of AGPase and SBE between two rice cultivars investigated, except the DBEs. Distribution patterns of branches of amy- lopectin changed continually during the development of rice grains and varied between two rice culti- vars.
