| 2 | Recent glacial retreat in High Asia in China and its impact on water resource in Northwest China显示文摘Under the impact of climatic warming, the glaciers in the High Asia in China have been retreating continuously with negative glacial mass balance in recent several decades. The retreat became more intensive in the past 10 years. The spatial pattern of the glacial retreat in the High Asia in China is that the smallest magnitude of retreat is in the inland of the Tibetan Plateau, the magnitude increases from the inland to the margin of the Tibetan Plateau, and the largest magnitude at the margin of the Tibetan Plateau. The glacial retreat in the High Asia in China has an important impact on the water resource of the arid regions in Northwest China. This study shows that the glacial retreat in the 1990s has caused an increase of 5.5% in river runoff in Northwest China. | 3 | Geomorphologic evidence of phased uplift of the northeastern Qinghai-Tibet Plateau since 14 million years ago显示文摘A typical sequence of fluvial terraces and aeolian deposits overlying these terraces were multidisciplinary investigated. New evidences for uplift process of the northeastern Qinghai-Tibetan Plateau in the past 14 million years were obtained. At least 11 river terraces along Huangshui, the first-class tributary of Yellow River, at the Xining-Huzhu region are identified. While the first one (T1) is classified as an accumulation terrace, the others are all basement river terraces, which consist of the Tertiary sandstone and siltstone bedrock, fluvial gravel and pebbles and the overlying aeolian loess-Red Clay deposit. Samples from the aeolian deposits were examined for paleomagnetic stratigraphic reconstruction (1030 samples), luminescence dating (16 samples), magnetic susceptibility and grain-size distribution (more than 4000 samples). The luminescence dating and stratigraphic correlation suggest that terraces of T11, T10,T8, T7, T3, T2, T1 were formed at 14, 11.3, 1.55, 1.2, 0.15, 0.07 and 0.01 million years ago, respectively. Sedimentological analysis and geomorphological observation indicate that formation and evolution of these terraces were mainly driven by tectonic uplift. Therefore, the terrace sequence provides an ideal geological record of the uplift process of the northeastern Qinghai-Tibet during the past 14 million years, and the timings of the terraces formation are regarded as the timings of tectonic uplift. The significant uplifting events took place at 14, 11.3, 1.2 and 0.15 million years ago, respectively. The fluvial incision at the Xining-Huzhu region is less than 100 m during a period of ~12 million years in the Miocene era (between the T11 and T9), while the Huangshui River had incised 432 m during the past 1.2 million years (from T7 to the present floodplain). The river incision process clearly demonstrates that accelerated rising of the northeastern Qinghai-Tibet Plateau during the late Cenozoic, and provides new evidence of previous thoughts. There was a significant readjustment of the fluvial catchment during 1.55-1.2 million years ago: before this time, the paleoriver flowed to southwest. After this time the Huangshui River flows to southeast. | 4 | Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. | 7 | Changes of gut bacteria and immune parameters in liver transplant recipients显示文摘BACKGROUND: Liver transplantation is one of the most effective therapeutic options for patients with end-stage liver diseases, and gut microbiota is actively involved in potential infections in pretransplant and posttransplant patients. However, the diversity of gut microbiota and its relationship with the immune parameter of liver transplantation recipients are not well understood. METHODS: We collected fresh feces and blood samples from 190 participants in China from November 2004 to May 2008, including 28 healthy volunteers, 51 cirrhotic patients and 111 liver-transplanted patients. Six interesting gut bacteria, plasma endotoxin, serum cytokines (i.e., tumor necrosis factor alpha and interleukin-6) and fecal secretory IgA (SIgA) were investigated by real-time quantitative PCR, chromogenic limulus amoebocyte assay, sandwich-type enzyme-linked immunosorbent assay and radioimmunoassay, respectively. RESULTS: All Eubacteria, Bifidobacterium spp., Faecalibacterium prausnitzii and Lactobacillus spp. were significantly lower in the liver transplantation recipients while Enterobacteriaceae and Enterococcus spp. were significantly higher (P<0.05). Except for Enterococcus spp., other bacteria showed a tendency to restore to normal level along with the time after liver transplantation. Plasma endotoxin, interleukin-6 and fecal SIgA in cirrhotic patients increased significantly, but not in liver transplantation recipients. Plasma endotoxin and interleukin-6 were negatively correlated with all Eubacteria and the Bacteroides-Prevotella group, while tumor necrosis factor alpha was not significantly correlated with these six gut bacteria in cirrhotic patients.CONCLUSIONS: Our study demonstrates that abundant gut bacteria were altered significantly in both cirrhotic and liver transplantation patients, while plasma endotoxin and interleukin-6 increased remarkably in cirrhotic patients, showing significant correlations with gut microbiota. As a typical gold-bearing quartz vein in this gold-rich area, the age data obtained from the Q875 also constrain on the metallogenic time of the lode gold deposits developed in the same geological settings. | 9 | Effect of carbon source on the denitrification in constructed wetlands显示文摘The ability of constructed wetlands with different plants in nitrate removal were investigated. The factors promoting the rates of denitrification including organic carbon, nitrate load, plants in wetlands, pH and water temperature in field were systematically investigated. The results showed that the additional carbon source (glucose) can remarkably improve the nitrate removal ability of the constructed wetland. It demonstrated that the nitrate removal rate can increase from 20% to more than 50% in summer and from 10% to 30% in winter, when the nitrate concentration was 30–40 mg/L, the retention time was 24 h and 25 mg/L dissolved organic carbon (DOC) was ploughed into the constructed wetland. However, the nitrite in the constructed wetland accumulated a little with the supply of the additional carbon source in summer and winter, and it increased from 0.15 to 2 mg/L in the effuent. It was also found that the abilities of plant in adjusting pH and temperature can result in an increase of denitrification in wetlands. It is a circulating hormone produced primarily by the adipose tissue. ob/ob mice with mutations of the gene encoding leptin become morbidly obese, infertile, hyperphagic, hypothermic,and diabetic. Since the cloning of leptin in 1994, our knowledge in body weight regulation and the role played by leptin has increased substantially. We now know that leptin signals through its receptor, OB-R, which is a member of the cytokine receptor superfamily. Leptin serves as an adiposity signal to inform the brain the adipose tissue mass in a negative feedback loop regulating food intake and energy expenditure. Leptin also plays important roles in angiogenesis, immune function, fertility and bone formation. | 12 | Metastatic human hepatocellular carcinoma models in nude mice and cell line with metastatic potential显示文摘Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature. | 13 | Discovery of seep carbonate nodules as new evidence for gas venting on the northern continental slope of South China Sea显示文摘Seep carbonate nodules were firstlycollected from the southwestern Dongsha area onnorthern continental slope of South China Sea formineralogical and geochemical studies. The resultsof X-ray diffraction show that carbonate nodules arecomposed of Fe-rich domolite,siderite and a few cal-cite and aragonite, as well as a small quantity ofnon-carbonate minerals such as goethite, quartz andclay minerals. Fe-rich dolomite and siderite charac-terized by euhedral microcrystalline structure weredirectly precipitated at cold seeps. The δ 13C valuesfor Fe-rich dolomite and siderite in the bulk nodulesvary from ?18.24‰ to ?36.07‰, and the δ 18O valuesrange from 0.42 to 2.76‰. Their moderate depleted13C reflects that carbon origin is possibly thermoge- netic gas or mixed gas, which is evidence of gasventings in the seafloor. Moreover, massive wormtube fossils related to cold seeps were found on thesurfaces of carbonate nodules, and the conduitsand/or channels in semi-solidified nodules whichwere not filled by carbonate or sediment were alsoobserved. | 15 | Analysis of DNA methylation in different maize tissues显示文摘DNA methylation plays an important role in gene expression regulation during biological development and tissue differentiation in plants. This study adopted methylation-sensitive Amplified fragment length polymorphism (AFLP) to compare the levels of DNA cyto- sine methylation at CCGG sites in tassel, bracteal leaf, and ear leaf from maize inbred lines, 18 White and 18 Red, respectively, and also examined specific methylation patterns of the three tissues. Significant differences in cytosine methylation level among the three tissues and the same changing tendency in two inbred lines were detected. Both MSAP (methylation sensitive amplification polymorphism) ratio and full methylation level were the highest in bracteal leaf, and the lowest in tassel. Meanwhile, different methylation levels were ob- served in the same tissue from the inbred lines, 18 White and 18 Red. Full methylation of internal cytosine was the dominant type in the maize genome. The differential methylation patterns in the three tissues were observed. In addition, sequencing of nine differentially me- thylated fragments and the subsequent blast search revealed that the cytosine methylated 5′-CCGG-3′sequences were distributed in re- peating sequences, in the coding and noncoding regions. Southern hybridization was used to verify the methylation polymorphism. These results clearly demonstrated the power of the MSAP technique for large-scale DNA methylation detection in the maize genome, and the complexity of DNA methylation change during plant growth and development. | 18 | Combined serum hepatoma-specific alpha-fetoprotein and circulating alpha-fetoprotein-mRNA in diagnosis of hepatocellular carcinoma显示文摘BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most common malignant tumors worldwide, its prognosis is poor, and early detection is of utmost importance. Although alpha-fetoprotein (AFP) is a useful marker for detecting and monitoring HCC development, the false-negative or false-positive rates with AFP alone may be as high as 30%-40% for patients with small HCCs. To enhance the specificity and accuracy of AFP measurements for HCC, we analyzed AFP expression states in livers, detected the hepatoma-specific AFP (HS-AFP) fraction and AFP-mRNA from peripheral blood mononuclear cells, and explored their clinical implications for HCC diagnosis. METHODS: AFP expression and hepatocyte distributions in liver specimens were investigated by an immunohistoche- mical assay. Total RNAs were extracted from circulating blood, synthesized to cDNA through random primers and reverse transcriptase, and fragments of the AFP gene were amplified by a nested-PCR assay. The HS-AFP fraction was separated by lectin-affinity chromatography and its level was detected by radioimmunoassay. RESULTS: The incidence of AFP was 73.3% in HCC tissues and its expression in HCCs with moderate or low differentiation was significantly stronger than that of HCCs with high differentiation (P<0.05). The incidence of AFP gene fragments was 100% in HCCs, and 60% in paracancerous tissues (P<0.01). In the HCC and liver cirrhosis groups, the incidence of HS-AFP was 91.7% and 18% (P<0.01), and of AFP-mRNA was 56.7% and 16% (P<0.01), respectively, and neither was found in controls.HS-AFP or AFP-mRNA was not significantly related to size or number of HCC, but to its differentiation, metastasis, and relapse (P<0.05). CONCLUSIONS: Different AFP expression is present in different parts of HCC tissues. | 20 | Total alkyl dibenzothiophenes content tracing the filling pathway of condensate reservoir in the Fushan Depression,South China Sea显示文摘The condensates are generally characterized by high maturity,low concentration of steranes and ter-panes biomarkers and low content of non-hydrocarbon fraction. In this case commonly used steranes,terpanes and carbazoles parameters cannot be effectively applied in the reservoir-filling tracing. The hydrogen bond formed by sulfur atom in the dibenzothiophenes (DBTs) results in molecule adsorption and fractionation during oil migration in reservoir. Like carbazoles,total DBTs content decreases with the increasing of oil migration distance. Therefore,a new parameter——total DBTs content is proposed to be used to trace the oil migration orientation and filling pathway. In present study,total DBTs con-tents of condensates and light oils are obtained by adding internal standard——eight deuterium atoms substituted DBT during Gas Chromatography-Mass Spectrometry analysis of aromatic fraction. Except for a few samples with much lower content of non-hydrocarbon fraction,the total DBTs content shows a fine positive correlation with that of carbazoles. Large errors can be caused in the process of pyrrolic nitrogen compounds separation. The application of this new parameter in the Fushan Depression of Beibu Gulf Basin,South China Sea indicates that this parameter is a reliable one to trace filling pathway in condensate reservoirs. Combined with other DBTs-related parameters,such as 4-/1-methydibenzo-thiophene and 2,4-/1,4-dimethyldibenzothiophene,oil migration orientation and filling pathway of the Fushan Depression was determined. The accumulations of Huachang oil field in the Fushan Depres-sion are mainly migrated and charged from northeast to southeast along the Huachang uplift. It can be predicated that the light oil and condensates in the Huachang oil field should be sourced from the source kitchen at the Bailian Sag.
